Esophageal cancer (EC) is a life-threatening disease, demanding the discovery of new biomarkers and molecular targets for precision oncology. Aberrantly glycosylated proteins hold tremendous potential towards this objective. In the current study, a series of esophageal squamous cell carcinomas (ESCC) and EC-derived circulating tumor cells (CTCs) were screened by immunoassays for the sialyl-Tn (STn) antigen, a glycan rarely expressed in healthy tissues and widely observed in aggressive gastrointestinal cancers. An ESCC cell model was glycoengineered to express STn and characterized in relation to cell proliferation and invasion in vitro. STn was found to be widely present in ESCC (70% of tumors) and in CTCs in 20% of patients, being associated with general recurrence and reduced survival. Furthermore, STn expression in ESCC cells increased invasion in vitro, while reducing cancer cells proliferation. In parallel, an ESCC mass spectrometry-based proteomics dataset, obtained from the PRIDE database, was comprehensively interrogated for abnormally glycosylated proteins. Data integration with the Target Score, an algorithm developed in-house, pinpointed the glucose transporter type 1 (GLUT1) as a biomarker of poor prognosis. GLUT1-STn glycoproteoforms were latter identified in tumor tissues in patients facing worst prognosis. Furthermore, healthy human tissues analysis suggested that STn glycosylation provided cancer specificity to GLUT1. In conclusion, STn is a biomarker of worst prognosis in EC and GLUT1-STn glycoforms may be used to increase its specificity on the stratification and targeting of aggressive ESCC forms.  相似文献

The aim of the present work was to develop a transient mathematical model focused on microalgae biomass drying, considering two phases: solid (wet biomass) and gas (drying air). Mass and thermal energy balances were written for each phase producing a system of ordinary differential equations (ODE). The solution of the ODE set delivers the temperature and air humidity ratio and biomass profiles with respect to time. The numerical results were directly compared with temperature experimental measurements—for both phases—and with the biomass humidity content. Data from experiment 1 were used to carry out the mathematical model adjustment, whereas data from experiment 2 were used for the experimental validation of the model. The model was adjusted by proposing a new correlation for the mass transfer coefficient and by calibrating the heat transfer coefficient. The transient numerical results were in good quantitative and qualitative agreement with the experimental results, ie, within the experimental error bars. Then the experimentally validated mathematical model was utilized to optimize the following parameters: (i) the electric heater power ( ) and the dry air mass flow rate ( ) and (ii) the convection oven length to width ratio (L/W). The goal was to minimize system energy consumption (objective function). The optimization procedure was subject to the following physical constraints: (i) fixed convection oven total volume and (ii) fixed biomass and drying air contact surface area. For the oven original geometry,  = 3.0 kW and  = 9 g s?1 were numerically found for minimum energy consumption, so that 36.9% and 43.5% energy consumption decreases were obtained, respectively, in comparison with the measurements of experiment 1. Next, the numerical geometric optimization found (L/W)opt = 9, with and , which was capable to reach a 51.6% energy consumption reduction in comparison with the original system tested in experiment 1. The novelty of this work consists of the development and experimental validation of a physically based microalgae biomass drying mathematical model, ie, instead of using empirical correlations to predict the drying time and temperature profiles and then minimize system energy consumption. Therefore, the results show that it is reasonable to state that the model could be used to design, control, and optimize drying systems with configurations similar to the one analyzed in this study.  相似文献

People without disabilities seamlessly control devices with their hands. Interestingly, their hands can perform coarse and fine control. Implementing smooth control for computerized systems is not straightforward and most of the time it is not intuitive either. Here we offer a solution to that problem: smooth control through humming. Voice commands have become ubiquitous in modern technology. Speech-to-text applications abound. Smooth control, on the other hand, has not been tackled yet. Here we design and implement a humming control technique, and demonstrate a hardware implementation with a powered wheelchair. Once actuated, the speed with which the chair moves will depend on the subtle variation on the fundamental frequency of the user's humming, acquired through an accelerometer measuring vocal cord vibration. We also discuss two signal processing techniques that handle commonly encountered issues when trying to resolve frequencies in real time data. The hardware implementation shows performance of 80% and higher in speech recognition for signal-to-noise ratio (SNR) higher than 8 dB and 100% in smooth control and frequency detection for all tested SNRs. We also discuss potential applications of smooth humming control to other assistive technology.  相似文献

The antioxidant properties of different ecotypes of chestnut nut (cv. Judia) were studied. Total phenolics and flavonoids were also determinated. Total phenolics amount ranged from 9.6 mg/g of GAE (hottest ecotype, Murça) to 19.4 mg/g of GAE (coldest ecotype, Valpaços). Gallic and ellagic acid were the predominant compounds and Valpaços had the highest values while, Murça had the lowest ones. The antioxidant capacity of ethanolic extracts were evaluated through several biochemical essays: ABTS (2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid)) and DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-scavenging activity, FRAP (ferric reducing/antioxidant power) and inhibition of oxidative haemolysis in erythrocytes. In order to evaluate the antioxidant efficiency of each ecotype, the EC50 values were calculated. Once again Valpaços revealed the best antioxidant properties, presenting much lower EC50 values. Climatic conditions influence seems to be a limiting factor for production of phenolic compounds and consequently for the antioxidant properties of chestnut nuts.  相似文献

The EFVM (Vitoria Minas Railroad) is one of the main railways in Brazil. It transports freight trains of ore, 220 wagons each. These wagons have 2 boogies of 2 axles each and 32 metric tons on metre gauge. Elastic strains were measured on a special part of this railway due to these trains. The main load to evaluate stresses and strains was a G 16 Locomotive, a C-C kind from Vale, a Brazilian Company. The measurements were obtained by dynamic deflectometer installed on a main line of this railway, near Ipatinga, a city from Minas Gerais, one of Brazil states. This track was equipped to obtain stresses under an equal repeated static load A simulation of the stresses was made under critical strain by Ferrovia 1.0 software. It was also made an evaluation of unequal results from neighbor sleepers taking in comparison two equipped parts of this railway, one with compacted ballast and no compaction to the other. The results were strain limited, avoiding breakage or damage to the studied rails. This work analyses these measurements focusing on the improvement of track quality.  相似文献

The thermomechanical degradation and thermostabilization of poly(lactic acid) (PLA) have been studied using subsequent extrusion cycles under different temperature profiles. Primary and secondary antioxidants were used to avoid degradation process during extrusion. Melt flow index (MFI), size exclusion chromatography (SEC), and infrared spectroscopy (FTIR) analysis were used to evaluate the degradation and stabilization of PLA. The MFI and SEC analysis show that the main thermodegradation mechanism of PLA is governed by scission reactions. FTIR analysis confirmed the SEC results and showed that the synergetic effect between primary and secondary antioxidant is a suitable way to thermostabilize the PLA. © 2013 Wiley Periodicals, Inc. J. Photobacteriosis is a septicaemic bacterial disease affecting several marine species around the globe, resulting in significant economic losses. Although many studies have been performed related to the pathogen virulence and resistance factors, information regarding the host defence mechanisms activated once an infection takes place is still scarce. The present study was designed to understand innate immune responses of farmed juvenile gilthead seabream (Sparus aurata) after Photobacterium damselae subsp. piscicida (Phdp) infection. Therefore, two groups of seabream juveniles were intraperitoneally injected with 100 µL of PBS (placebo) or 100 µL of exponentially growing Phdp (1 × 106 CFU/mL; infected). The blood, plasma, liver, and head kidney of six fish from each treatment were sampled immediately before infection and 3, 6, 9, 24 and 48 h after infection for the broad screening of fish immune and oxidative stress responses. Infected animals presented marked anaemia, neutrophilia and monocytosis, conditions that are correlated with an increased expression of genes related to inflammation and phagocytic activity. Similar studies with different fish species and bacteria can be useful for the definition of health biomarkers that might help fish farmers to prevent the occurrence of such diseases.  相似文献

The aim of this study was to evaluate the effect of solidification cooling rates on the as-cast microstructural morphologies of a Pb-1 wt%Sn alloy, and to correlate the resulting microstructure with the corresponding electrochemical corrosion resistance in a 0.5 M H2SO4 solution at 25 °C. Cylindrical low-carbon steel and insulating molds were employed permitting the two extremes of a significant range of solidification cooling rates to be experimentally examined. Electrochemical impedance spectroscopy (EIS) diagrams, potentiodynamic polarization curves and an equivalent circuit analysis were used to evaluate the electrochemical corrosion response of Pb-1 wt%Sn alloy samples. It was found that lower cooling rates are associated with coarse cellular arrays which result in better corrosion resistance than fine cells which are related to high cooling rates. The experimental results have shown that that the pre-programming of microstructure cell size of Pb-Sn alloys can be used as an alternative way to produce as-cast components of lead-acid batteries with higher corrosion resistance.  相似文献

The latex obtained from Hancornia speciosa is used in folk medicine for treatment of several diseases, such as acne, warts, diabetes, gastritis and inflammation. In this work, we describe the biocompatibility assessment and angiogenic properties of H. speciosa latex and its potential application in medicine. The physical–chemical characterization was carried out following different methodologies (CHN elemental analyses; thermogravimetric analyses and Fourier transform infrared spectroscopy). The biocompatibility was evaluated through cytotoxicity and genotoxicity tests in fibroblast mouse cells and the angiogenic properties were evaluated using the chick chorioallantoic membrane (CAM) assay model. The physical–chemical results showed that the structure of Hancornia speciosa latex biomembrane is very similar to that of Hevea brasiliensis (commercially available product). Moreover, the cytotoxicity and genotoxicity assays showed that H. speciosa latex is biocompatible with life systems and can be a good biomaterial for medical applications. The CAM test showed the efficient ability of H. speciosa latex in neovascularization of tissues. The histological analysis was in accordance with the results obtained in the CAM assay. Our data indicate that the latex obtained from H. speciosa and eluted in water showed significant angiogenic activity without any cytotoxic or genotoxic effects on life systems. The same did not occur with H. speciosa latex stabilized with ammonia. Addition of ammonia does not have significant effects on the structure of biomembranes, but showed a smaller cell survival and a significant genotoxicity effect. This study contributes to the understanding of the potentialities of H. speciosa latex as a source of new phytomedicines.  相似文献
