A simulation model that assesses the risk of acquiring salmonellosis from consumption and handling of chicken was created in an Excel ™ spreadsheet and was simulated using Risk ™. The model simulated the distribution, preparation, and consumption of 1,000 chickens and was designed to determine the relationship between the level of Salmonella contamination on chickens at the processing plant exit and the risk of salmonellosis for consumers of the chickens. A scatter plot of the probability of acquiring salmonellosis from consumption of the chickens simulated versus the Salmonella load on the chickens at the processing plant exit clearly showed that highly (i.e., > 100 Salmonella/ bird) contaminated chickens at the plant exit did not necessarily pose greater risk of salmonellosis than lightly (i.e., < 10 Salmonella/ bird) contaminated chickens at the plant exit. Rather, greater risk of salmonellosis was realized from lightly contaminated chickens when they were temperature-abused, undercooked, and consumed by someone from the high risk population.  相似文献

Lithium‐ion battery performance is intrinsically linked to electrode microstructure. Quantitative measurement of key structural parameters of lithium‐ion battery electrode microstructures will enable optimization as well as motivate systematic numerical studies for the improvement of battery performance. With the rapid development of 3‐D imaging techniques, quantitative assessment of 3‐D microstructures from 2‐D image sections by stereological methods appears outmoded; however, in spite of the proliferation of tomographic imaging techniques, it remains significantly easier to obtain two‐dimensional (2‐D) data sets. In this study, stereological prediction and three‐dimensional (3‐D) analysis techniques for quantitative assessment of key geometric parameters for characterizing battery electrode microstructures are examined and compared. Lithium‐ion battery electrodes were imaged using synchrotron‐based X‐ray tomographic microscopy. For each electrode sample investigated, stereological analysis was performed on reconstructed 2‐D image sections generated from tomographic imaging, whereas direct 3‐D analysis was performed on reconstructed image volumes. The analysis showed that geometric parameter estimation using 2‐D image sections is bound to be associated with ambiguity and that volume‐based 3‐D characterization of nonconvex, irregular and interconnected particles can be used to more accurately quantify spatially‐dependent parameters, such as tortuosity and pore‐phase connectivity.  相似文献

One strain of 11 serotypes or 11 strains of Salmonella, which were isolated from the ceca of broilers, were surveyed for their growth kinetics on sterile ground chicken breast burgers incubated at 25C to determine the variation of lag time and specific growth rate. Growth curves, four per strain, were fit to a two‐phase linear model to determine lag time (h) and specific growth rate (log10/h). Repeatability of growth kinetics measurements for individual strains had a mean coefficient of variation of 11.7% for lag time (range: 5.8 to 17.3%) and a mean coefficient of variation of 6.7% for specific growth rate (range: 2.7 to 13.3%). Lag time among strains ranged from 2.2 to 3.1 h with a mean of 2.8 h for all strains, whereas specific growth rate among strains ranged from 0.3 to 0.38 log10 per h with a mean of 0.35 log10per h for all strains. One‐way analysis of variance indicated that lag time (P =0.029) and specific growth rate (P =0.025) differed slightly among strains. S. Haardt had a shorter (P < 0.05) lag time than S. Agona and S. Brandenburg, whereas the specific growth rate of S. Enteritidis was less than (P < 0.05) the specific growth rates of S. Typhimurium and S. Brandenburg. All other strains had similar lag times and specific growth rates. The coefficient of variation among strains was 9.4% for lag time and 5.7% for specific growth rate. These results indicate that there were only minor differences in the lag times and specific growth rates among the strains of Salmonella surveyed. Thus, the growth kinetic values obtained with one strain of Salmonella may be useful for predicting the growth of other strains of Salmonella for which data do not currently exist.  相似文献

The aim of this study was to investigate fecal shedding and transmission of E. coli O157 in cohorts of cattle within a feedlot, to assess subsequent contamination of carcasses with this pathogen and to identify risk factors associated with fecal shedding of E. coli O157. A cohort of 133 heifers housed infour adjacent pens was examined over a five month period, from entering the feedlot to slaughter. Individual rectal fecal samples and pen environmental samples were taken at monthly intervals. The entire outer and inner surfaces of a carcass side of each animal were swabbed immediately following slaughter.
E. coli O157 was isolated from 136 (23%) of the 600 rectal fecal samples; 96% of which contained virulent markers. One hundred and sixty environmental samples were examined and E. coli O157 was isolated from 46 (29%), all of which contained virulent markers. E. coli O157 was not isolated from any of the dressed carcasses. The prevalence of E. coli O157 fecal shedding may be related to the pen and E. coli O157 contamination of the pen floor feces, water trough and feed.
E. coli O157 should be considered as a pathogen shed in the feces of a substantial proportion of feedlot cattle. However, with good hygienic practice at harvest, a very low level of this pathogen can be achieved on dressed carcasses.  相似文献

Previous morphometric methods for estimation of the volume of components, surface area and thickness of the diffusion barrier in fish gills have taken advantage of the highly ordered structure of these organs for sampling and surface area estimations, whereas the thickness of the diffusion barrier has been measured orthogonally on perpendicularly sectioned material at subjectively selected sites. Although intuitively logical, these procedures do not have a demonstrated mathematical basis, do not involve random sampling and measurement techniques, and are not applicable to the gills of all fish. The present stereological methods apply the principles of surface area estimation in vertical uniform random sections to the gills of the Brazilian teleost Arapaima gigas. The tissue was taken from the entire gill apparatus of the right‐hand or left‐hand side (selected at random) of the fish by systematic random sampling and embedded in glycol methacrylate for light microscopy. Arches from the other side were embedded in Epoxy resin. Reference volume was estimated by the Cavalieri method in the same vertical sections that were used for surface density and volume density measurements. The harmonic mean barrier thickness of the water‐blood diffusion barrier was calculated from measurements taken along randomly selected orientation lines that were sine‐weighted relative to the vertical axis. The values thus obtained for the anatomical diffusion factor (surface area divided by barrier thickness) compare favourably with those obtained for other sluggish fish using existing methods.  相似文献

Dairy laboratories are responsible for measuring the quality of ingredients, intermediate products and finished products. A particular challenge in the food processing sector is that consistently high quality products must be made from raw material which varies widely in composition and to some extent in quality. Consequently, routine milk sampling and analysis form part of the quality control system in any dairy plant. Sampling and data collection begin at the milk collection point on the farm, and so the collection truck must be considered part of the informationlquality control management system. Computerized information management systems relieve the workload on the laboratory manager by assisting with tasks such as organizing the workload; tracking samples; monitoring instruments; acquiring, storing, reporting and analysing test data; and with additional functions such as Good Laboratory Practice and IS0 9000. Of particular significance to laboratories is that computer interfaces and computer technology inside analytical instruments have opened new ways of integrating on-line measurements in the process, and on-line laboratory data. With the increasing rationalization of the dairy industry there are fewer laboratories, and a laboratory often processes samples for several factories, which requires good integration.
