The comparison pairs of cores (each 100 mm apart) from 68 reinstatements sites from various parts of the UK revealed that, the compounding consequences of generic non-homogeneous characteristics of hand laid recipe mixed materials (specified in Specification for the Reinstatement of Openings in Highways (SROH)) and high likeliness of being biased during air void (AV) testing makes the coring method extremely unreliable with very low repeatability and reproducibility. The wide-ranging maximum density reported in every instance in the comparison pair coring experiments meaningfully rationalizes the distorted homogeneity of materials. Although not only maximum density but also bulk density of adjacent cores located only 100 mm apart were found to be varied in the case of every pair in this study. Furthermore, the in situ performance shown by from 50 reinstatements after experiencing 1.5–10 years real-life ageing from various parts of the UK predictably indicates that either the linkage between the reinstatement with non-compliant AV and its impact on footways durability is non-proven or the reported AV content is extremely over estimated. At 95% level of significance, there exists enough evidence to conclude that, due to high uncertainty, very low repeatability and reproducibility and poor reliability with high chances of bias, the assessment of hand laid reinstatement work by AV testing will expose both the contractor and the client to unacceptable risk.  相似文献

In classical public‐key infrastructure (PKI), the certificate authorities (CAs) are fully trusted, and the security of the PKI relies on the trustworthiness of the CAs. However, recent failures and compromises of CAs showed that if a CA is corrupted, fake certificates may be issued, and the security of clients will be at risk. As emerging solutions, blockchain‐ and log‐based PKI proposals potentially solved the shortcomings of the PKI, in particular, eliminating the weakest link security and providing a rapid remedy to CAs' problems. Nevertheless, log‐based PKIs are still exposed to split‐world attacks if the attacker is capable of presenting two distinct signed versions of the log to the targeted victim(s), while the blockchain‐based PKIs have scaling and high‐cost issues to be overcome. To address these problems, this paper presents a secure and accountable transport layer security (TLS) certificate management (SCM), which is a next‐generation PKI framework. It combines the two emerging architectures, introducing novel mechanisms, and makes CAs and log servers accountable to domain owners. In SCM, CA‐signed domain certificates are stored in log servers, while the management of CAs and log servers is handed over to a group of domain owners, which is conducted on the blockchain platform. Different from existing blockchain‐based PKI proposals, SCM decreases the storage cost of blockchain from several hundreds of GB to only hundreds of megabytes. Finally, we analyze the security and performance of SCM and compare SCM with previous blockchain‐ and log‐based PKI schemes.  相似文献

We propose a microsphere array device with microspheres having controllable positions for error-free target identification. We conduct a statistical design analysis to select the optimal distance between the microspheres as well as the optimal temperature. Our design simplifies the imaging and ensures a desired statistical performance for a given sensor cost. Specifically, we compute the posterior Cramér-Rao bound on the errors in estimating the unknown target concentrations. We use this performance bound to compute the optimal design variables. We discuss both uniform and sparse concentration levels of targets, and replace the unknown imaging parameters with their maximum likelihood estimates. We illustrate our design concept using numerical examples. The proposed microarray has high sensitivity, efficient packing, and guaranteed imaging performance. It simplifies the imaging analysis significantly by identifying targets based on the known positions of the microspheres. Potential applications include molecular recognition, specificity of targeting molecules, protein-protein dimerization, high throughput screening assays for enzyme inhibitors, drug discovery, and gene sequencing.  相似文献

This study evaluates the effects of surface roughness, saliva and bracket materials on in vitro frictional resistance of contemporary and aesthetic orthodontic brackets during the sliding of archwires against elastomeric ligature. Eight various brackets were investigated together with stainless steel (SS) archwires of several shapes. The sliding force involved in a ligated bracket-archwire combination was measured in an Instron by a self-designed jig. The interfaces were also examined using scanning electron microscope (SEM) to simulate sliding mechanics and abrasive/adhesive wear of the bottom surfaces of the bracket slots, which were observed under dry condition. Round SS archwires (SRO-OR) demonstrated the lower frictional force compared to coated round SS archwire (SRO-AW) and the flat SS archwire (rectangular and square) surfaces for most of the brackets. Braided composite bracket without a metal slot had the least resistance when sliding with SRO-OR archwire whilst the composite bracket (Spirit MB) with a metal slot demonstrated the lower resistance. Ceramic bracket (Inspire) without a metal slot, which also reflected severe wear patterns produced a significantly larger frictional force for SRO-AW. The presence of saliva had a consistent effect in increasing the friction, which counters the inconsistency reported in the literature. The soft-coated surface of SRO-AW archwire confirms sliding difficulty in sliding on the hard surface of bracket slot. A smoother surface correlates with a lower frictional force according to the surface roughness values.  相似文献

Composites are usually brittle materials and have low impact properties. Structural dimensions, stacking sequence, ply materials, ply thicknesses and ply angles are standard variables that influence composite‘s performance against impact loads. Stacking sequence in hybrid laminates affects the failure and impact resistance. Failure mechanisms at the low-velocity impact of a rigid object in hybrid laminates are complex, and the subsurface damage in a composite laminate cannot be detected directly. However, various simulation platforms make it easy to see the impact damage between the plies of laminate. This paper numerically investigated the effect of stack sequence and hybridization of two fiber types against low-velocity impact. The current study adopted four-layer composite laminates of carbon and glass fiber layers with a stacking plan [C/C/C/C], [C/G/C/G] and [G/C/G/C], having lay-up angles as [0°/45°/−45°/90°]. Keeping the impactor mass and the incident velocity constant, the laminates were subjected to low-velocity impact. The damage contours for a failure mode were recorded and compared at the ply level. The numerical study resulted in impact imitations showing comparisons of the damage contours using Hashin failure criteria. Hybrid laminates display better performance in absorbing impact energies; however, hybrid laminates experienced more subsurface damage due to more impact energy absorption.  相似文献

Aflatoxin M1 (AFM1) is a metabolite of aflatoxin B1 (AFB1) and secreted in milk of animals that are kept on a moldy diet. AFB1 is produced by several toxigenic species of fungi. In this study, the comparative efficacy of two mycotoxin binders was evaluated in terms of reducing excretion of AFM1, total viable bacterial count and somatic cell count in milk obtained from goats fed with AFB1 contaminated diet. A total of 32 lactating Beetal goats were reared and divided into 4 equal groups: Animals of group A were kept as control, while those in groups B, C, and D were individually fed with AFB1 at 40 µg alone, 40 µg AFB1 along with mycotoxin binder Toxfin® at 3 g kg?1 of cotton seed cake, and 40 µg AFB1 along with mycotoxin binder Elitox® at a dose rate of 1 g kg?1 of cotton seed cake, respectively. Toxfin® significantly decreased the excretion of AFM1 in goats’ milk by 49.57 % while Elitox® non-significantly reduced the excretion of AFM1 in goats’ milk by 19.49 %. Total viable bacterial count in milk samples of group C and D, and somatic cell count in milk samples of group C did not reduce significantly. However, somatic cell count in milk samples of group D reduced significantly. We conclude that the addition of Toxfin® in the moldy diet significantly reduce the excretion of AFM1 in animals’ milk and minimize the risk of mycotoxin toxicity to public health.  相似文献

In oligonucleotide microarray experiments, noise is a challenging problem, as biologists now are studying their organisms not in isolation but in the context of a natural environment. In low photomultiplier tube (PMT) voltage images, weak gene signals and their interactions with the background fluorescence noise are most problematic. In addition, nonspecific sequences bind to array spots intermittently causing inaccurate measurements. Conventional techniques cannot precisely separate the foreground and the background signals. In this paper, we propose analytically based estimation technique. We assume a priori spot-shape information using a circular outer periphery with an elliptical center hole. We assume Gaussian statistics for modeling both the foreground and background signals. The mean of the foreground signal quantifies the weak gene signal corresponding to the spot, and the variance gives the measure of the undesired binding that causes fluctuation in the measurement. We propose a foreground-signal and shape-estimation algorithm using the Gibbs sampling method. We compare our developed algorithm with the existing Mann-Whitney (MW)- and expectation maximization (EM)/iterated conditional modes (ICM)-based methods. Our method outperforms the existing methods with considerably smaller mean-square error (MSE) for all signal-to-noise ratios (SNRs) in computer-generated images and gives better qualitative results in low-SNR real-data images. Our method is computationally relatively slow because of its inherent sampling operation and hence only applicable to very noisy-spot images. In a realistic example using our method, we show that the gene-signal fluctuations on the estimated foreground are better observed for the input noisy images with relatively higher undesired bindings.  相似文献

Although single-photon fluorescence lifetime imaging microscopy (FLIM) is widely used to image molecular processes using a wide range of excitation wavelengths, the captured emission of this technique is confined to the visible spectrum. Here, we explore the feasibility of utilizing near-infrared (NIR) fluorescent molecular probes with emission >700 nm for FLIM of live cells. The confocal microscope is equipped with a 785 nm laser diode, a red-enhanced photomultiplier tube, and a time-correlated single photon counting card. We demonstrate that our system reports the lifetime distributions of NIR fluorescent dyes, cypate and DTTCI, in cells. In cells labelled separately or jointly with these dyes, NIR FLIM successfully distinguishes their lifetimes, providing a method to sort different cell populations. In addition, lifetime distributions of cells co-incubated with these dyes allow estimate of the dyes' relative concentrations in complex cellular microenvironments. With the heightened interest in fluorescence lifetime-based small animal imaging using NIR fluorophores, this technique further serves as a bridge between in vitro spectroscopic characterization of new fluorophore lifetimes and in vivo tissue imaging.  相似文献

In rural Bangladesh various components of four ponds, including aquatic plants, snails, water, oysters, and sediment were studied for faecal contamination for three years, from July 1994, to June 1997. Faecal coliform counts ranged from 2.80–5.45, 2.95–5.64, 1.49–2.05, 2.51–4.60 and 1.30–3.15 log10cfu/gm or ml in plant, snail, water, oyster and sediment samples respectively during the study period. Snail and sediment samples showed highest and lowest counts, respectively. All components of the studied ponds included in this study yielded coliform counts higher than the accepted limit for potability and domestic use, as recommended by the WHO. Therefore these ponds have a high potential for transmission of diarrhoeal and other water‐borne diseases.  相似文献
