Usher syndrome is an autosomal recessive disorder characterized by congenital hearing loss combined with retinitis pigmentosa, and in some cases, vestibular areflexia. Three clinical subtypes are distinguished, and MYO7A and USH2A represent the two major causal genes involved in Usher type I, the most severe form, and type II, the most frequent form, respectively. Massively parallel sequencing was performed on a cohort of patients in the context of a molecular diagnosis to confirm clinical suspicion of Usher syndrome. We report here 231 pathogenic MYO7A and USH2A genotypes identified in 73 Usher type I and 158 Usher type II patients. Furthermore, we present the ACMG classification of the variants, which comprise all types. Among them, 68 have not been previously reported in the literature, including 12 missense and 16 splice variants. We also report a new deep intronic variant in USH2A. Despite the important number of molecular studies published on these two genes, we show that during the course of routine genetic diagnosis, undescribed variants continue to be identified at a high rate. This is particularly pertinent in the current era, where therapeutic strategies based on DNA or RNA technologies are being developed. 相似文献

La Reunion island in the South West Indian Ocean is now endemic for dengue following the introduction of dengue virus serotype 2 (DENV-2) cosmopolitan-I genotype in 2017. DENV-2 infection causes a wide spectrum of clinical manifestations ranging from flu-like disease to severe dengue. The nonstructural glycoprotein 1 (NS1) has been identified as playing a key role in dengue disease severity. The intracellular NS1 exists as a homodimer, whereas a fraction is driven towards the plasma membrane or released as a soluble hexameric protein. Here, we characterized the NS1 glycoproteins from clinical isolates DES-14 and RUN-18 that were collected during the DENV-2 epidemics in Tanzania in 2014 and La Reunion island in 2018, respectively. In relation to hepatotropism of the DENV, expression of recombinant DES-14 NS1 and RUN-18 NS1 glycoproteins was compared in human hepatoma Huh7 cells. We observed that RUN-18 NS1 was poorly stable in Huh7 cells compared to DES-14 NS1. The instability of RUN-18 NS1 leading to a low level of NS1 secretion mostly relates to lysine residues on positions 272 and 324. Our data raise the issue of the consequences of a defect in NS1 stability in human hepatocytes in relation to the major role of NS1 in the pathogenesis of the DENV-2 infection. 相似文献

the structural, functional, and immunological properties of this lipoprotein. Here we present data showing that, under appropriate
conditions of cryopreservation, Lp(a) retains the properties of the freshly isolated product, and we provide examples supporting

Ceramic parts manufactured by lithography‐based ceramic manufacturing (LCM) excel in resolution and surface quality. The material for LCM is a photosensitive ceramic particle‐filled slurry which needs to have homogeneous properties over time and during each processing step. The goal of this study was to use “mechanical” stabilization for a tricalcium phosphate‐filled slurry done by increasing slurry viscosity, solids loading, or inducing thixotropic behavior. The modified slurries were compared with a nonstable reference slurry. While all methods lead to increased storage stability, only the stabilized slurry with 0.5 wt% fumed silica is stable during the printing process. 相似文献

Discretised structural models such as by finite elements imply discretised support conditions. In some cases such as plates on elastic foundation or slabs on large interacting columns an improved formulation of the continuous support conditions is desirable. This can be achieved by means of linear constraint equations. The numerical treatment of linear constraints is discussed for the method of elimination of variables as well as for the method of Lagrange multipliers. Then specific constraint equations for different accuracy requirements are derived, which can be used to constrain rectangular flat shell elements of arbitrary shape functions. These constraints introduce six generalized displacements according to the rigid body motions of the element and transmit the corresponding generalized reactions on the nodal degrees of freedom in a way consistent with distributed reactions. The effect on the strain energy of a square shell element is shown for the different constraint equations. As an application, the linear constraints are used to represent the continuous interaction of columns with the plate in a flat slab structure. Comparison of the finite element solutions with analytical results shows that the derived constraint equations allow a considerably improved formulation of continuous support conditions. 相似文献

Flour beetles are cosmopolitan and common pests in grain stores and flour mills. Their ability to exploit a wide variety of stored products has contributed to their status as major pests of stored food. Although it was previously reported that the same aggregation pheromone, 4,8-dimethyldecanal (DMD), is shared by three flour beetles species (Tribolium castaneum, T. confusum, and T. freemani), the volatiles released by the other Tribolium species associated with stored products have not yet been examined. In the present study, the volatiles produced by males and females of eight Tribolium species were examined by solid phase microextraction (SPME). SPME samples were analyzed by gas chromatography coupled to mass spectrometry (GC-MS). Experiments were conducted to identify volatiles emitted by the adults of different Tribolium species and to determine whether DMD is a common aggregation pheromone. We observed that DMD is not a common pheromone of the eight species tested, but is common to T. castaneum, T. confusum, T. freemani, and T. madens. Two other volatiles were detected, 1-pentadecene, which is shown here to be a common semiochemical of flour beetles, and 1,6-pentadecadiene, which was detected in five species (T. audax, T. brevicornis, T. destructor, T. freemani, and T. madens). 相似文献

ComfSim – Interactive indoor thermal comfort simulation on high performance computers. The paper outlines the current state of the development of a ‘computational steering’ environment for interactive indoor thermal comfort simulation and assessment. The system consists of a parallel CFD kernel, a fast 3D mesh generator and an integrated virtual reality‐based visualization component. The numerical method is based on a hybrid thermal lattice Boltzmann method with extensions for simulations of turbulent convective flows. Utilizing high‐performance supercomputing facilities allows for modifying both the geometric model and the boundary conditions during runtime with immediate visualization of changes in the results. We currently enhance our model by a radiation model with fast visibility check and integrate a local thermal comfort model developed in cooperation with our partners. The application is demonstrated by turbulent convection in a train's passenger carriage. 相似文献
