一种便携快速微流芯片PCR仪性能评价

目的 评价一种便携快速微流芯片PCR仪的主要性能指标。方法 按照RB/T 160-2017《分析化学仪器设备验证与综合评价指南》、SN/T 2102.2-2008《食源性病原体PCR检测技术规范第2部分: PCR仪性能试验要求》等相关标准制定的评价指标及要求, 选取标准菌株鼠伤寒沙门氏菌ATCC14028、肠炎沙门氏菌ATCC13076和大肠埃希氏菌O157:H7 ATCC43895食源性致病菌为检测目标物, 对便携快速微流芯片PCR仪的基线噪声、检测低限、线性、精密度、样品检测及重复性进行验证实验。结果 检测基线稳定、线性拟合相关系数r>0.98, 精密度均小于5%, 满足YY/T 1173-2010《聚合酶链反应分析仪》的要求。结论 基线噪声、检测低限、线性、精密度、样品检测及重复性6个指标可以有效的评价便携快速微流芯片PCR仪。     

海藻岩藻多糖抗肿瘤活性研究新进展

岩藻多糖是一种主要来自海洋褐藻的多糖,由硫酸化的聚糖残基所组成,具有广泛的生物学活性,如抗肿瘤、抗氧化、抗病毒、抗炎等。大量研究发现,岩藻多糖不仅通过诱导细胞凋亡、阻滞细胞周期等途径直接发挥抗癌作用,还可以通过激活自然杀伤细胞、巨噬细胞等间接杀伤癌细胞。岩藻多糖因其生物活性高、来源广泛、副作用低、耐药性好等优点,可开发成为一种新型的抗肿瘤药物,或者与化疗药物联合用于临床上肿瘤的辅助治疗。本文综述了岩藻多糖抗肿瘤的作用机理,及其在抗肝癌、肺癌、乳腺癌、白血病等方面的研究进展,及岩藻多糖的代谢和毒性,以期为临床上癌症的预防和治疗提供更多的理论依据。     

Investigation into the triggering mechanism of loess landslides in the south Jingyang platform,Shaanxi province

Loess landslide incidents have resulted in significant economic losses and human casualties in Northwest China. To investigate the triggering mechanism of such loess landslides in the south Jingyang platform, Shaanxi province, an area which is subject to loess landslides, we performed a series of field measurements for loess landslide LD37 and also undertook laboratory. Comparisons between the field measurements of LD37 and the results of the constant shear drained (CSD) triaxial test indicate that the seepage from irrigation-water infiltration triggered the loess landslides in the study area where the initiation of the local sliding surfaces was linked to gentle stratum erosion and their subsequent development was because of excessive stratum erosion. The development of slow–rapid strain cycles in the ε–p′ plot from the CSD triaxial test results indicates that loess landslides are a feature of progressive failure of the loess slope. Preventive measures to reduce irrigation-water infiltration into loess slopes should be taken to prevent similar incidents from recurring in the future.

Effects of CYP3A43 Expression on Cell Proliferation and Migration of Lung Adenocarcinoma and Its Clinical Significance

The cytochrome P450s (CYP450s) include key oxidative enzymes involved in the metabolism of various carcinogens and anticancer drugs. Bioinformatic studies have demonstrated the association of CYP3A43 with liver cancer and ovarian cancer. However, the biological function of CYP3A43 in tumor progression remains unclear. To further reveal the role of CYP3A43 in tumor progression, we first analyzed the data from the UALCAN database and found that CYP3A43 was negatively correlated to the cancer staging and lymph node metastasis of lung adenocarcinoma (LUAD). We established stable CYP3A43-knockdown LUAD H1299 cell line and found that its knockdown enhanced cell proliferation, colony formation, and migration in vitro, and promoted the growth of tumor xenograft in vivo. Interestingly, when CYP3A43 was ectopically-expressed in the LUAD cell lines, decreased cell proliferation and ERK1/2 phosphorylation level were observed. Lastly, we also identified CYP3A43 co-expressed genes in LUAD from LinkedOmics database followed by GO and KEGG analyses. In conclusion, our results indicate the unprecedented role of CYP3A43 in the suppression of LUAD and provide new possibilities for targeted therapy of this life-threatening disease.
