The effect of illumination on intracellular free calcium concentration, [Ca2+]i, was studied in a cell line (WiDr cells) derived from a primary adenocarcinoma of the rectosigmoid colon. In these cells the biosynthesis of protoporphyrin IX was stimulated by 5-aminolevulinic acid to reach levels of 600-700 pmol of protoporphyrin IX per mg cell protein. A brief (1-min) exposure of the cells to light (70% of light energy at 340-380 nm) resulted in an increase in [Ca2+]i. This increase was not reversible over a period of at least 20 min following illumination. Elevation of [Ca2+]i most probably represented an influx of calcium ions from the medium to the cell, since it was completely abolished in the presence of extracellular EGTA. The increased [Ca2+]i did not reflect general membrane damage, as determined by trypan blue staining as well as measurement of the intercalation of ethidium bromide into cellular DNA, and neither did the sustained elevation of [Ca2+]i lead to any substantial loss of clonogenicity following illumination of protoporphyrin-containing cells. Together these results indicate that an increased [Ca2+]i level is not per se a cause of cell death during photodynamic therapy.  相似文献

The direct discharge of wastewater from ships is prohibited by the International Maritime Organization, especially discharge of oily bilge water. This study aims to develop a compact onboard integrated wastewater treatment system for all the wastewater streams on ships, including grey water, black water and bilge water, using biofilm-MBR technology. Both dead-end side-stream and recycle side-stream configurations of a biofilm-MBR concept have been investigated. Flat sheet ceramic membranes from KeraNor AS were applied in the membrane filtration unit. A good membrane permeate quality was achieved in each process configuration, with oil concentrations < 5 mg/L. However, the membrane was seriously fouled when the dead-end side-stream configuration was operated with a high filtration unit recovery (93%). A significantly improved membrane performance, and better permeate quality, was achieved by recycling the concentrate solution back to the biofilm reactor due to better bio-flocculation and biodegradation of oil compounds in the process. The effect of process configuration was investigated by comparing oil concentration, SMP, TSS, FCOD, TCOD, PSD of the concentrate solutions around the membrane in filtration unit. The membrane fouling rate was found to be directly related to oil concentration and oily biomass characteristics in the membrane filtration unit.  相似文献

Biaxial strength, fracture toughness and subcritical crack growth are reported for coarse grained porous alumina ceramics. The materials were prepared with a varying amount of a silica sintering aid, which resulted in the formation of a glassy secondary phase at the grain boundaries. Crystalline mullite was additionally found in the material with the highest silica content. The biaxial strength, measured by Ball-on-Ring and Ball-on-3-Balls, was highest for the material without mullite at the grain boundaries, and the biaxial strength decreased with increasing porosity. The fracture toughness of the materials was in the range of 1.7–1.9 MPa m0.5. Measurements of subcritical crack growth by a modified lifetime method in air and aqueous environments demonstrated a higher crack growth rate in water and acid relative to in air. The effect of porosity and grain boundary phase were discussed in relation to subcritical crack growth and fracture mode in the coarse grained alumina ceramics.  相似文献

The invasive rainbow smelt (Osmerus mordax) was an abundant food source for Lake Winnipeg walleye (Sander vitreus), especially in the north basin of the lake, until the smelt’s collapse in approximately 2013. We quantified changing length-at-age (≈ growth rates) and relative mass (≈ body condition) in Lake Winnipeg walleye caught for a gillnet index data set. Here, walleye showed smaller length-at-age, particularly young fish in the north basin, over time. This approach to assessing growth suggests a constraint in the north basin fish, possibly a nutritional limitation between 2017 and 2018, that was not present in the south. We then analyzed a separate group of walleye (≥452 mm in fork length) sampled in 2017 as part of a large-scale tracking study, which had a similar slope in length-mass relationship to large walleye caught in that year for the gillnet index data. A panel of metabolites in whole blood samples associated with amino acid metabolism and protein turnover was compared. These metabolites revealed elevated essential amino acids in fish caught in the Dauphin River, and suggest that protein degradation may be elevated in north basin walleye. Therefore, based on both growth estimates and metabolites associated with protein balance, we suggest there were spatially distinct separations affecting Lake Winnipeg walleye with decreased nutritional status of walleye in the north basin of Lake Winnipeg being of particular concern.  相似文献

Phenolic compounds derived from the olive plant (Olea europaea L.), particularly hydroxytyrosol and oleuropein, have many beneficial effects in vitro. Olive leaves are the richest source of olive phenolic compounds, and olive leaf extract (OLE) is now a popular nutraceutical taken either as liquid or capsules. To quantify the bioavailability and metabolism of oleuropein and hydroxytyrosol when taken as OLE, nine volunteers (five males) aged 42.8 ± 7.4 years were randomized to receive either capsulated or liquid OLE as a single lower (51.1 mg oleuropein, 9.7 mg hydroxytyrosol) or higher (76.6 mg oleuropein, 14.5 mg hydroxytyrosol) dose, and then the opposite strength (but same formulation) a week later. Plasma and urine samples were collected at fixed intervals for 24 h post‐ingestion. Phenolic content was analyzed by LC‐ESI‐MS/MS. Conjugated metabolites of hydroxytyrosol were the primary metabolites recovered in plasma and urine after OLE ingestion. Peak oleuropein concentrations in plasma were greater following ingestion of liquid than capsule preparations (0.47 versus 2.74 ng/mL; p = 0.004), but no such effect was observed for peak concentrations of conjugated (sulfated and glucuronidated) hydroxytyrosol (p = 0.94). However, the latter peak was reached earlier with liquid preparation (93 versus 64 min; p = 0.031). There was a gender effect on the bioavailability of phenolic compounds, with males displaying greater plasma area under the curve for conjugated hydroxytyrosol (11 600 versus 2550 ng/mL; p = 0.048). All conjugated hydroxytyrosol metabolites were recovered in the urine within 8 h. There was wide inter‐individual variation. OLE effectively delivers oleuropein and hydroxytrosol metabolites to plasma in humans.  相似文献

For crystalline silicon solar cells, the efficient collection of light at wavelengths in the infrared is a challenge because of long absorption lengths. Especially for thinner wafers, an efficient light‐trapping scheme, such as the patch texture, is required for high short‐circuit current densities. We have measured the light‐trapping properties of patch textures produced by laser assisted texturing (LAST) on polished ⟨100⟩silicon wafers, and compared them with ray‐tracing simulations. Single‐sided random pyramid textures are created for comparison. Excellent agreement between simulations and measurements is achieved by employing diffuse scattering with a narrow angular distribution in the simulations, confirming the successful implementation of the process. We use our optical measurements of the textures for simulations of textures with rear reflectors, where we also investigate the influence on light‐trapping properties when varying geometry and reflectance properties. The results from the optical simulations are imported into the solar cell simulation program PC1D. For a 50 μm‐thick solar cell, we simulate an improvement in Jsc of up to 0.4 mA/cm2 when going from single‐sided random pyramid textures to patch textures, even when the performance of the texture is limited by process inaccuracies. Removing the physical inaccuracies of the laser system, the potential gain in Jsc on a 50 μm‐thick cell with a patch texture covering the complete wafer surface is 0.8 mA/cm2. We therefore conclude that the LAST method for creating patch textures is suitable to achieve an improved Jsc in thin monocrystalline silicon solar cells. A wide range of problems can be modelled as constraint satisfaction problems (CSPs), that is, a set of constraints that must be satisfied simultaneously. Constraints can either be represented extensionally, by explicitly listing allowed combinations of values, or implicitly, by special-purpose algorithms provided by a solver. Such implicitly represented constraints, known as global constraints, are widely used; indeed, they are one of the key reasons for the success of constraint programming in solving real-world problems. In recent years, a variety of restrictions on the structure of CSP instances have been shown to yield tractable classes of CSPs. However, most such restrictions fail to guarantee tractability for CSPs with global constraints. We therefore study the applicability of structural restrictions to instances with such constraints. We show that when the number of solutions to a CSP instance is bounded in key parts of the problem, structural restrictions can be used to derive new tractable classes. Furthermore, we show that this result extends to combinations of instances drawn from known tractable classes, as well as to CSP instances where constraints assign costs to satisfying assignments.  相似文献
