The measurement of minute samples is desirable in many areas of research and analysis, including biological, environmental, and forensic sciences. The use of manual solution direct deposition, combined with surface evaporation, is a very useful and convenient method for the transfer of many analytes to a spectroscopic sampling window. For Fourier transform infrared (FT-IR) spectrometric analysis, high quality attenuated total reflection (ATR) and transmission analyses are possible with suitable solution deposits. If the sample area is very small, placement of the analyte deposit on the active area of the sampling accessory must be done carefully. To achieve this, a novel direct deposition system has been developed. This system, a series of valves attached to a glass nebulizer, generates deposits that are reproducible, and placement of the deposits is precise. The valves allow the sample to be loaded, helium airflow to expel the sample, and the nebulizer to be cleaned after deposition. To help contain the sample to a small area once deposition has taken place, a vacuum line is attached to the nebulizer. This simple semiautomated deposition system allows for higher sensitivity and run-to-run reproducibility for minute sample analysis.  相似文献

Over one in ten students surveyed have admitted to copying programs in courses with computer assignments. The ease with which digital coursework can be copied and the impracticality of manually checking for plagiarized projects in large courses has only compounded the problem. As current research has focused predominantly on detecting plagiarism for textual applications such as source code and documents, there exists a gap in detecting plagiarism in graphically-driven applications. This paper focuses on the effectiveness of a technological tool in detecting plagiarized projects in a course using Microsoft Access. Seven semesters of data were collected from a large technology-oriented course in which the tool had been in use. Comparing semesters before and after the technological tool was introduced demonstrates a significant decrease in the number of projects being duplicated. The results indicate combining technology and policy can be effective in curtailing blatant plagiarism within large technology courses.  相似文献

The mechanical properties for zirconium diboride (ZrB2) were measured at two laboratories and compared. Two billets of ZrB2 were prepared by hot-pressing commercial powder. The relative densities of the billets were >99% and with an average grain size of 5.9 ± 4.5 µm. Both laboratories prepared American Society for Testing and Materials (ASTM) C1161 B-bars for strength and ASTM C1421 bars with notch configuration A for fracture toughness. Specimens were machined by diamond grinding at the Army Research Laboratory (ARL) and electrical discharge machining (EDM) at Missouri S&T. Strength bars tested at Missouri S&T were polished to a .25 μm finish while the bars were tested as-ground at ARL. Strengths were 473 ± 79 MPa for the Missouri S&T bars and 438 ± 68 for the ARL bars while the fracture toughness values were 3.9 ± .7 MPa•m1/2 for the Missouri S&T bars and 4.4 ± .6 MPa•m1/2 for the ARL bars. Vickers hardness was measured by both laboratories over a range of indentation loads. The resulting hardness values were on the low end of previously reported values and were quite different from each other especially at indentation loads ≤20N. The study demonstrated that the properties of materials tested to ASTM standards at different laboratories can be compared directly. In addition, strength and fracture toughness were nearly identical for bars prepared by conventional diamond grinding or EDM.  相似文献

BACKGROUND: Allergoids are widely used in specific immunotherapy for the treatment of IgE-mediated allergic diseases. OBJECTIVE: The aim of this study was to analyse whether a modification of birch pollen allergens with formaldehyde affects the availability of T-cell epitopes. METHODS: Efficient modification of the allergens was verified by determining IgE and IgG binding activity using ELISA inhibition tests. T-cell responses to birch pollen allergoids were analysed in polyclonal systems, using peripheral blood mononuclear cells (PBMC) of five birch pollen-allergic individuals, as well as birch pollen extract-reactive T-cell lines (TCL), established from the peripheral blood of 14 birch pollen-allergic donors. To determine whether the modification of natural (n)Bet v 1 with formaldehyde or maleic anhydride results in epitope-specific changes in T-cell reactivities, 22 Bet v 1-specific T-cell clones (TCC), established from nine additional birch pollen-allergic individuals, were tested for their reactivity with these products. RESULTS: The majority of PBMC and TCL showed a reduced response to the birch pollen extract allergoid. Bet v 1-specific TCC could be divided into allergoid-reactive and -non-reactive TCC. No simple correlation between possible modification sites of formaldehyde in the respective T-cell epitopes and the stimulatory potential of the allergoid was observed. Mechanisms of suppression or of anergy induction were excluded as an explanation for the non-reactivity of representative TCC. All TCC could be stimulated by maleylated and unmodified nBet v 1 to a similar extent. CONCLUSION: These results demonstrate differences in the availability of T-cell epitopes between allergoids and unmodified allergens, which are most likely due to structural changes within the allergen molecule.  相似文献

In exploring the structural features which determine the antitumor activity of 2,4,6-tris-[(hydroxymethyl)methylamino]-1,3,5-triazine (trimelamol, 1), we have synthesized analogues in which the methyl groups have been replaced by the electron-withdrawing substituents 2,2,2-trifluoroethyl (5), propargyl (13), and cyanomethyl (15) via the respective tris(alkylamino)triazines 3, 12, and 14. Three mono[(hydroxymethyl)amino]triazines (4, 7, and 10) were also prepared. All the new tris(hydroxymethyl) derivatives showed cytotoxicities toward a variety of experimental rodent and human ovarian tumor cell lines similar to those shown by 1, the cyanomethyl analogue (15) having the most favorable profile. Mono(hydroxymethyl) derivatives (4 and 7) were ca. one-third as toxic. The new tris(hydroxymethyl) analogues were more stable to aqueous hydrolysis than was 1. Half-life (pH 7.5) values were, for 1, 120 min, for 5, 690 min, for 13, 450 min, and for 15, 275 min, but at pH 2.0, 15 (t1/2 350 min) was the most stable. This cyanomethyl analogue was also the most water-soluble, being comparable to 1 whereas 5 and 13 were poorly soluble.  相似文献

In spontaneously beating sinus venosus of the frog Caudiverbera caudiverbera the effects of bekanamycin and dibekacin, two aminoglycoside antibiotics, on action potentials of cardiac primary pacemaker cells were studied by intracellular recording. Bekanamycin and dibekacin induced a concentration-dependent decrease of the amplitude, overshoot and the rate of rise of the action potential. Both also flattened the slow diastolic depolarization leading to a marked decrease in beat rate. At the highest concentration used (1 x 10(-3) M), the aminoglycosides produced a complete inhibition of primary cells action potentials. It was preceded by the appearance of subthreshold oscillations of the membrane potential which were observed for a few minutes until the electrical activity of pacemaker cell ceased. During absence of impulse initiation a stable membrane potential about -40 mV was observed. Aminoglycoside effects, excepting those on SCL, were completely suppressed when external calcium was increased to 3.6 mM. The results support the conclusion that bekanamycin and dibekacin depress the electrical activity of pacemaker cells. It is suggested that this effect is induced by aminoglycosides blockade of the slow calcium current involved in both upstroke and slow diastolic depolarization and through modification of potassium outward current. Bekanamycin at a lower concentration than that needed to induce electrophysiological effects potentiated verapamil 1 x 10(-8) effects on cardiac pacemaker cells.  相似文献

Chronic in utero methamphetamine treatment, throughout gestation in rats, resulted in alterations in both behavior and brain monoamine function in the adult offspring. The higher dose of methamphetamine (10 mg/kg/b.i.d.) caused a significant decrease in square crossing and rearing in an open field, as well as a regional increase of serotonin and dopamine uptake sites. In contrast, the lower dose of in utero methamphetamine (2 mg/kg/b.i.d.) resulted in a significant decrease in regional densities of serotonin and dopamine uptake sites, and only decreased rearing behavior. Across treatment groups, there were significant correlations between open-field square crossing activity and the number of uptake sites in specific brain areas. Other measured behaviors, such as the neonate righting reflex and the adult Morris water maze performance, were unaffected by either in utero drug regimen. These results are discussed in terms of the known neurotoxicity of amphetamines and the ability of the immature nervous system to compensate for fetal exposure to methamphetamine.  相似文献

The onset of sonically induced cavitation in liquid helium at frequencies between 30 and 40 kHz has been studied. In helium II, two types of cavitation activity were identified: acoustic cavitation whose characteristic noise can be detected, and visible cavitation in which vaporous cavities grow to visible size. The onset of acoustic cavitation is statistical in nature with increasing event rates as the sound pressure amplitude is increased and whose threshold depends on the waiting time at that particular amplitude. The acoustic threshold sound pressure amplitude in helium II between 1.8° K andT was found to lie within 0.15 mb of 0.3 mb, the variation of ±0.15 mb occurring from one determination to another, whereas the sound pressure amplitude corresponding to the visible threshold was about a hundred times larger. These two distinct types of sonically induced cavitation appear to be unique to liquid helium. However, aboveT the two thresholds were found to coincide at a sound pressure amplitude within 0.4 mb of 0.8 mb. The characteristics of the onset of acoustic cavitation were found to be independent of applied static pressure of up to 1.5 atm above and belowT and in helium II they were unaffected by filtering, heat flushing, or rotating the liquid. The results suggest that liquid helium is nucleated by random events initiated by the ambient cosmic radiation or by vortices generated in the liquid, and they imply that at ultrasonic frequencies this liquid cannot withstand a tensile stress and behaves in this respect like water saturated with gas and containing dust motes. Attempts to determine the onset of acoustic cavitation by scattering light off the bubbles or by detecting sonoluminescence were not successful: The upper limit to the size of these bubbles was shown to be about 30 µm and the intensity of any sonoluminescence must have been less than 10–4 of that from cavitating water. The possibilities of exploiting the two types of cavitation activity in liquid helium in the construction of a posttriggerable ultrasonic bubble chamber for visualizing the tracks of ionizing particles are discussed, as are the theoretical background and future development of the work presented in this paper.  相似文献
