ABSTRACT

In this article, we explore how visitors engage with a science museum exhibit that displays controversial topics. Through a case study methodology, we focus on the Brazilian exhibit Preventing Youth Pregnancy that delves into teen pregnancy, sexual practices, and sexually transmitted diseases. Using the lens of science communication (an emergent field of research) we explore (a) communication patterns established between the exhibit and the public and (b) the dimensions of engagement that visitors experienced. Data were collected through observations and interviews with visitors, field notes, exit comment cards, and interviews with staffers. Our findings revealed the potential of articulating different models of science communication and exposed a visitor experience in which informed decision making, sharing of personal stories, co-production of knowledge, and learning how to care (and act) were at play while negotiating the complexities of the issues raised by the displays.  相似文献

The main problem for experiments using the bolometric technique to search for rare events is the contribution of surface contamination to the background. In this paper a new technique for the identification of the origin of events will be described. The idea is to shield the main bolometer with bolometric shields. In an effort to understand molecular mechanisms of intracellular lipid transport, we have focused upon specific events required for de novo aminophospholipid synthesis in the yeast Saccharomyces cerevisiae. A genetic system for examining the steps between phosphatidylserine (PtdSer) synthesis in the endoplasmic reticulum and its transport to and decarboxylation by PtdSer decarboxylase 2 in the Golgi/vacuole has been developed. We have isolated a mutant, denoted pstB1, that accumulates PtdSer and has diminished phosphatidylethanolamine formation despite normal PtdSer decarboxylase 2 activity. The lesion in PtdSer metabolism is consistent with a defect in interorganelle lipid transport. A genomic DNA clone that complements the mutation was isolated, and sequencing revealed that the clone contains the STT4 gene, encoding a phosphatidylinositol 4-kinase. The pstB1 mutant exhibits a defect in Stt4p-type phosphatidylinositol 4-kinase activity, and direct gene replacement indicates that STT4 is the defective gene in the mutant. Creation of an STT4 null allele (stt4Delta::HIS3) demonstrates the gene is essential. These results provide evidence that implicates phosphoinositides in the regulation of intracellular aminophospholipid transport.  相似文献

An underground station for the measurement of low-level radioactivity is in operation in Northern Italy in the town of Como under the Baradello hill. The rock cover is -300 m water equivalent. This paper reports about the preliminary measurements carried out to characterise the site of the Baradello hill and the installation of a high-purity Ge detector with a radio-pure copper shielding. Features and levels of the achieved background are reported and discussed, in comparison with other above ground and underground low-radioactivity stations. Sensitivities to radioactivity in typical environmental matrices are evaluated, confirming the very low-gamma background attained in the laboratory.  相似文献

BACKGROUND: Color kinesis (CK) is a new echocardiographic technique for the assessment of left ventricular (LV) wall motion based on acoustic quantification. Using integrated backscatter data, this technique identifies the pixel value transitions from blood to myocardial tissue throughout systole and tracks endocardial motion in real time. The color-encoded images, built on a frame-by-frame basis by adding one color at a time, provide an integrated display of the timing and amplitude of endocardial motion in a single end-systolic frame. Recent studies have shown that CK is a promising clinical tool for quantitative assessment of regional LV function. OBJECTIVES: The aim of this study was to evaluate the feasibility and accuracy of CK in identifying the regional wall-motion abnormalities diagnosed by conventional two-dimensional (2-D) echocardiography in patients after acute myocardial infarction (AMI). METHODS: The end-systolic color overlays were analyzed using a method to quantify the regional timing and amplitude of endocardial systolic excursion (ESE) based on the count of the numbers of colors. At this point, the total duration (ESE timing) and distance (ESE amplitude) of endocardial excursion from end-diastolic to end-systolic color-frame was calculated in each segment. In 54 patients after AMI, we compared the feasibility and ability of CK superimposed on 2-D superimposed on 2-D superimposed on 2-D echocardiographic images and visual 2-D echo analysis to evaluate the endocardial border excursion in parasternal short-axis (SAX) and apical four-(AP4CH) and two-(AP2CH) chamber views. In 20 normal subjects, the end-systolic color overlays were used to evaluate the variability of the measurements of ESE timing (msec) and amplitude (cm) and to define the reference values. Image quality was considered adequate if at least 12 of 16 segments could be evaluated for systolic function by conventional visual 2-D echo. Among 54 patients, 35 with adequate studies were selected to determine the accuracy of quantitative analysis of CK images in identifying regional wall-motion abnormalities. RESULTS: The SAX view was obtained in 36 of 54 patients; of the possible 216 segments, 210 (97%) were adequately visualized by 2-D echocardiography and 207 (96%) by CK. Apical views were obtained in 50 patients (93%); of the possible 300 segments, 93% were visualized by 2-D echocardiography and 90% by CK in the AP4CH view and 94% and 92%, respectively, were visualized by the two methods in the AP2CH view. In normal subjects, measurements of ESE timing and amplitude were found to be consistent and the mean values were 346 msec (range 280-360) and 0.99 cm (range 0.72-1.26) respectively. In the 35 selected patients, 2-D echocardiography identified 355 normokinetic segments in which ESE timing and amplitude were similar to the reference values. In 83 hypokinetic segments and 108 akinetic segments, ESE timing and amplitude were significantly inferior to values of normokinetic segments (p < 0.001). An ESE timing below the reference values of 280 msec identified all of the 191 asynergic segments (sensitivity and specificity = 100%) and an ESE amplitude of less than 0.70 cm identified 188 asynergic segments (sensitivity = 98% and specificity = 99%). CONCLUSIONS: CK showed good feasibility and diagnostic accuracy in identifying regional wall motion abnormalities in patients with acute myocardial infarction. The model used in our study for the quantitative analysis of color kinesis images, which provided easy and feasible indices of timing and amplitude of endocardial excursion, enabled fast and objective evaluation of LV regional wall motion.  相似文献

A thermal energy storage system, consisting of a packed bed of rocks as storing material and air as high-temperature heat transfer fluid, is analyzed for concentrated solar power (CSP) applications. A 6.5 MWhth pilot-scale thermal storage unit immersed in the ground and of truncated conical shape is fabricated and experimentally demonstrated to generate thermoclines. A dynamic numerical heat transfer model is formulated for separate fluid and solid phases and variable thermo-physical properties in the range of 20–650 °C, and validated with experimental results. The validated model is further applied to design and simulate an array of two industrial-scale thermal storage units, each of 7.2 GWhth capacity, for a 26 MWel round-the-clock concentrated solar power plant during multiple 8 h-charging/16 h-discharging cycles, yielding 95% overall thermal efficiency.  相似文献

Biofuels are of great topical interest, especially in the light of recent European directives. Biodiesel is both already available and thoroughly regulated. A critical aspect of the transesterification process, used in biodiesel production, is maximization of the contact surface between reagents. This is currently achieved using discontinuous processes with mechanical agitation, which however afford poor mixing efficiency. Ultrasound appears to be an effective method to enhance mixing intensity. The aim of this work was to assess the advantages offered by this technique, especially in terms of process velocity. For this purpose a small scale experimental apparatus for biodiesel production integrated with a modular ultrasound system was set up. Plant dimension and energy inputs were chosen to enable eventual industrial implementation. Plant performances were assessed as the velocity of oil conversion to fatty acids methyl esters. The product was analyzed according to EN 14105. In general, the efficiency of ultrasound in increasing reaction velocity was markedly affected by residual triglyceride content. Conversion rates in line with EN 14214 and ASTM D6751 standard specifications, which allow a maximum residual triglycerides content of 0.20%, were obtained with reductions in reaction time of about 20%. In less stringent conversion conditions, e.g. 3% triglycerides, the reaction time was roughly halved. Below 1% of triglycerides residues sonication should be interrupted and the process continued with conventional agitation.  相似文献

Fluoroelastomers (FEs) usually have working temperatures above 150°C and a great resistance to aggressive agents such as oils, fuels, aliphatic and aromatic solvents, steam, moderate acid, and basic environments. Liquid crystal polymers (LCPs) can be effective processing aids and reinforcing agents for elastomers. These characteristics are very attractive to lower melt viscosity and to stiffen and strengthen the final product through a simple blending. Among the LCPs, the semirigid LCPs seem the most appealing for blending with flexible thermoplastics (FTs) because their processing temperatures can be arranged to be in the same processing temperature range of FTs and because the presence of flexible segments can improve the compatibility with the flexible matrix. This is very important especially for FEs that are subjected not only to mechanical degradation, but also to thermal degradation. Blends of FEs with two types of semirigid LCPs show easy processability and enhanced mechanical and thermomechanical properties. These improvements were observed both for pure FEs and for filled vulcanized systems. The improvement of the mechanical properties is below that expected on the basis of the additive rule due to the incompatibility between the components; nevertheless, an impressive increase of the elastic modulus, up to 40 times, was noticed by adding 20% of LCP. In vulcanized systems, the tensile strength is also increased, whereas the elongation at break is slightly reduced. The working temperature is also drastically increased both in vulcanized and in unvulcanized systems. © 1996 John Wiley & Sons, Inc.  相似文献

The homology modeling of GPCRs has benefitted vastly from the availability of some resolved structures, which allow the generation of many reliable GPCR models. However, the dynamic behavior of such receptors has been only minimally examined in silico, although several pieces of evidence have highlighted some conformational switches that can orchestrate the activation mechanism. Among such switches, Pro-containing helices play a key role in determining bending in TM helices and thereby the width of the TM bundle. The approach proposed herein involves the generation of a set of possible models (conformational chimeras) by exhaustively combining the two main conformations (straight and bent) that a Pro-containing helix can assume. This approach was validated by generating conformational chimeras for the Cys-LTR1 receptor, which is involved in contractile and inflammatory processes. The generated chimeras were then used for docking a small set of representative ligands. The results revealed the flexibility mechanisms of Cys-LTR1, showing how the docked agonists vary their stabilizing interactions, shifting from the open to closed state, and how the examined antagonists are able to block the receptor in an open and inactive conformation, thus behaving as inverse agonists. This study emphasizes the promising potential of chimera modeling, confirms the key role of proline residues in receptor activation, and suggests that docking results can be improved by considering the often-overlooked flexibility of receptors.  相似文献
