Although nanotechnology has led to important advances in in vitro diagnostics, the development of nanosensors for in vivo detection remains very challenging. Here, we demonstrated the proof-of-principle of in vivo detection of nucleic acid targets using a promising type of surface-enhanced Raman scattering (SERS) nanosensor implanted in the skin of a large animal model (pig). The in vivo nanosensor used in this study involves the “inverse molecular sentinel” detection scheme using plasmonics-active nanostars, which have tunable absorption bands in the near infrared region of the “tissue optical window”, rendering them efficient as an optical sensing platform for in vivo optical detection. Ex vivo measurements were also performed using human skin grafts to demonstrate the detection of SERS nanosensors through tissue. In this study, a new core–shell nanorattle probe with Raman reporters trapped between the core and shell was utilized as an internal standard system for self-calibration. These results illustrate the usefulness and translational potential of the SERS nanosensor for in vivo biosensing.

Potassium carbonate-based sorbents are prospective materials for direct air capture (DAC). In the present study, we examined and revealed the influence of the temperature swing adsorption (TSA) cycle conditions on the CO2 sorption properties of a novel aerogel-based K2CO3/ZrO2 sorbent in a DAC process. It was shown that the humidity and temperature drastically affect the sorption dynamic and sorption capacity of the sorbent. When a temperature at the sorption stage was 29 ℃ and a water vapor pressure in the feed air was 5.2 mbar (1 bar = 105 Pa), the composite material demonstrated a stable CO2 sorption capacity of 3.4% (mass). An increase in sorption temperature leads to a continuous decrease in the CO2 absorption capacity reaching a value of 0.7% (mass) at T = 80 ℃. The material showed the retention of a stable CO2 sorption capacity for many cycles at each temperature in the range. Increasing PH2O in the inlet air from 5.2 to 6.8 mbar leads to instability of CO2 sorption capacity which decreases in the course of 3 consecutive TSA cycles from 1.7% to 0.8% (mass) at T = 29 ℃. A further increase in air humidity only facilitates the deterioration of the CO2 sorption capacity of the material. A possible explanation for this phenomenon could be the filling of the porous system of the sorbent with solid reaction products and an aqueous solution of potassium salts, which leads to a significant slowdown in the CO2 diffusion in the composite sorbent grain. To investigate the regeneration step of the TSA cycle in situ, the macro ATR-FTIR (attenuated total reflection Fourier-transform infrared) spectroscopic imaging was applied for the first time. It was shown that the migration of carbonate-containing species over the surface of sorbent occurs during the thermal regeneration stage of the TSA cycle. The movement of the active component in the porous matrix of the sorbent can affect the sorption characteristics of the composite material. The revealed features make it possible to formulate the requirements and limitations that need to be taken into account for the practical implementation of the DAC process using the K2CO3/ZrO2 composite sorbent.  相似文献

Objective: Understanding the psychological processes that contribute to smoking reduction will yield population health benefits. Negative mood may moderate smoking lapse during cessation, but this relationship has been difficult to measure in ongoing daily experience. We used a novel form of ecological momentary assessment to test a self-control model of negative mood and craving leading to smoking lapse. Design: We validated short message service (SMS) text as a user-friendly and low-cost option for ecologically measuring real-time health behaviors. We sent text messages to cigarette smokers attempting to quit eight times daily for the first 21 days of cessation (N-obs = 3,811). Main outcome measures: Approximately every two hours, we assessed cigarette count, mood, and cravings, and examined between- and within-day patterns and time-lagged relationships among these variables. Exhaled carbon monoxide was assessed pre- and posttreatment. Results: Negative mood and craving predicted smoking two hours later, but craving mediated the mood–smoking relationship. Also, this mediation relationship predicted smoking over the next two, but not four, hours. Conclusion: Results clarify conflicting previous findings on the relation between affect and smoking, validate a new low-cost and user-friendly method for collecting fine-grained health behavior assessments, and emphasize the importance of rapid, real-time measurement of smoking moderators. The present paper investigates the performance of a shifted factorized sparse approximate inverse as a parallel preconditioner for the iterative solution to the linear systems arising in the finite element discretization of non‐linear groundwater flow models. The shift strategy is based on an inexpensive preconditioner update exploiting the structure of the coefficient matrix. The proposed algorithm is experimented with in the parallel simulation of a large‐scale real multi‐aquifer system characterized by a stochastic distribution of the hydraulic conductivity. The numerical results show that the shifted factorized sparse approximate inverse algorithm may yield an overall computational gain up to 300% with respect to the non‐shifted scheme with an excellent parallel efficiency. Although the role of social support in promoting employees' health and well-being has been studied extensively, the evidence is inconsistent, sometimes even suggesting that social support might have negative effects. The authors examined some psychological processes that might explain such effects. On the basis of the threat-to-self-esteem model, the authors tested the hypothesis that receiving imposed support elicits negative reactions, which are moderated by someone's need for support. The authors distinguished 3 different reactions: (a) self-related, (b) interaction-related, and (c) physiological. The results of an experiment with 48 temporary administrative workers generally confirmed the hypothesis. Imposed support elicited negative reactions, except when there was an unsolvable problem, but even then the effect of imposed support was not positive but neutral. Summary A series expansion of theLaurent type which is the three-dimensional counterpart of a result obtained byReichardt (1960) for two dimensions is derived. With the aid of this expansion we find the radiation conditions valid for arbitrary complex frequencies. For these conditions it is then possible to give a proof of the fact that the eigenfrequencies for the first and second boundary value problems are always damped. The solution to the first boundary value problem and its uniqueness proof in caseGreen's function exists are given. The proof of the existence of theGreen's function to the first and second boundary value problems is completely analogous to the one given byReichardt and is therefore omitted here. An application of theLaurent expansion to the solution of the first boundary value problem for a spherical layer concludes the report.
Zusammenfassung Es wird eineLaurent-Entwicklung hergeleitet, die das dreidimensionale Gegenstück zu einem Ergebnis vonReichardt (1960) für zwei Dimensionen darstellt. Mit Hilfe dieser Entwicklung werden die Ausstrahlungsbedingungen für beliebige komplexe Frequenzen ermittelt. Für diese Bedingungen kann dann gezeigt werden, daß die Eigenfrequenzen für das erste und zweite Randwertproblem stets gedämpft sind. Für den Fall der Existenz einerGreenschen Funktion wird die Lösung des ersten Randwertproblems angegeben und ihre Eindeutigkeit gezeigt. Der beweis der Existenz einerGreenschen Funktion für das erste und zweite Randwertproblem ist vollkommen analog zu dem vonReichardt angegebenen und wird deshalb hier weggelassen. Eine Anwendung derLaurent-Entwicklung auf die Lösung des ersten Randwertproblems für die Kugelschichte beschließt die Arbeit.

In continuation of our studies on dynamic tolerance after condylar nailing in pertrochanteric fractures, we have examined the femora of patients who died within the first 2 weeks after surgery and studied their stress tolerance. We appreciated the lack of the weight bearing medial cortical bone and the need for abduction at the fracture site by the behaviour under stress. The results under stress could be compared with an experimental fracture model. From these experiments conclusions have been made for the correct placement of condylar nails for their best static advantage.  相似文献

The incorporation of β-amino acids into a peptide sequence has gained particular attention as β- and α/β-peptides have shown remarkable proteolytic stability, even after a single homologation at the scissile bond. Several peptidases have been shown to cleave such bonds with high specificity but at a much slower rate compared to α-peptide bonds. In this study, a series of analogs of dipeptidyl peptidase-4 (DPP-4) substrate inhibitors were synthesized in order to investigate whether β-amino acid homologation at the scissile bond could be a valid approach to improving peptide stability towards DPP-4 degradation. DPP-4 cleaved the α/β-peptide bond after the N-terminal penultimate Pro with a broad specificity and retained full activity regardless of the β3-amino acid side chain and peptide length. Significantly improved half-lives were observed for β3Ile-containing peptides. Replacing the penultimate Pro with a conformationally constrained Pro mimetic led to proteolytic resistance. DPP-4 cleavage of α/β-peptide bonds with a broad promiscuity represents a new insight into the stability of peptide analogs containing β-amino acids as such analogs were thought to be stable towards enzymatic degradation.  相似文献
