A rapid and sensitive method for detection of staphylococcal enterotoxin B (SEB) in raw and dry milk samples with the use of antibody-based immunomagnetic separation (IMS) in conjunction with flow cytometry (FCM) was developed. Sheep anti-SEB immunoglobulin G (IgG) was immobilized on Dynabeads M-280. The SEB initially binds to the capturing antibody, which is bound on the magnetic beads. The rabbit anti-SEB IgG binds to the captured toxin and is further labeled with a Cy5-labeled goat anti-rabbit IgG antibody. The percentage of the beads that were fluorescent was measured by FCM. FCM was carried out for 1 min, and the data obtained were expressed as histograms for particle size (forward light scatter) and histograms for fluorescence intensity. A peak corresponding to the magnetic beads was clearly distinguished from a peak derived from contaminating particles in the sample solution. In the absence of SEB, about 10% of the beads emitted fluorescence. The percentage of fluorescent beads and the fluorescence intensity increased with increasing SEB concentrations. For this IMS-FCM assay, the lower limits of detection for SEB were estimated to be 0.01 and 0.25 ng/ml for buffer and milk samples, respectively.  相似文献

The kinetics of conformational change of troponin C (TN-C) induced by binding and removal of calcium ions were studied by measuring the fluorescence of tyrosine by stopped-flow spectrofluorometry. When the concentration of free calcium ions in the solution [Ca2+] was increased rapidly from 4X10(-9)M to 1X10(-3)M at neutral pH, a first-order reaction with a rate constant of 13.7 sec-1, which was preceded by a much faster reaction, was observed. In contrast, when [Ca2+] was reduced from 2X10(-3)M to 4.4X10(-8)M, two first-order reactions with rate constants of 7.4 and 0.78 sec-1, preceded by a much faster reaction, were observed.  相似文献

Two-longitudinal-mode distributed-Bragg-reflector laser diodes have been fabricated for wavelength-division-multiplexing communication systems, two-wavelength optical measurement systems, and optical data processing systems. A Bragg reflector with a periodic-phase-shift grating is adopted for two-longitudinal-mode operation. IgE metabolic turnover studies with purified radioiodinated IgE were performed in normal individuals and in a patient with IgE myeloma. The validity of the turnover studies was established in several ways, including comparisons of radioiodinated IgE turnover with the turnover of endogenously labeled 14C-IgE and with the turnover of infused unlabeled IgE in a patient with hypogammaglobulinemia. The geometric mean serum IgE concentration in 73 normal adults was 96 ng/ml with a 68% confidence interval of 24 to 386 ng/ml. Metabolic turnover studies in 10 control individuals disclosed a geometric mean total circulating IgE of 4.1 mug/kg, a mean percentage of the total exchangeable IgE in the intravascular space of 41%, a mean half-time of survival of IgE of 2.7 days, a mean fractional catabolic rate of 94% of intravascular pool per day, and a geometric mean synthetic rate of 3.8 mug/kg/day. IgE has the lowest synthetic rate and highest fractional catabolic rate of the five major classes of immunoglobulin molecules. In contrast to these normal values, a patient with IgE myeloma had a serum IgE comcentration of 42 mg/ml, a total circulating IgE of 1.7 g/kg, and a synthetic rate of 270 mg/kg/day. Furthermore, although the synthetic rate was vastly increased, the survival time was prolonged and the fractional catabolic rate was decreased to 5.1 days and 16 to 22% of the intravascular pool per day, respectively. These data are compatible with the concept that IgE is catabolized in part by a mechanism common to all immunoglobulin classes and in part by a unique mechanism not available to other immunoglobulins. At very high IgE serum concentrations, such as those encountered in patients with IgE myeloma, the unique mechanism would be saturated and only the catabolic pathways available to all immunoglobulins would be available to IgE.  相似文献

Elevated levels of fibrinogen/fibrin degradation products (FDP) occur in uremia, and have been thought to be in part related to intravascular coagulation in the kidney. More recent data indicated that delayed catabolism of fibrinogen fragment D occurred in anephric animals. To further evaluate FDP catabolism in the kidney, turnover studies of purified dog 131I-Fg-D and 125I-Fg-E were performed on dogs before and after acute subtotal nephrectomies, and later during chronic uremia. 131I-fibrinogen clearances were also perfomed. Slowed catabolism of Fg-D and Fg-E was observed in both the acute and chronic uremic stages. Altered urinary excretion was not a factor as only minimal amounts of Fg-D and Fg-E were excreted in the urine of the control animals. In the 131I-fibrinogen studies, there were significant changes in plasma volume, fibrinogen t 1/2, and intravascular/extravascular distribution, but not in fractional catabolic rate. To differentiate fully, the effects of uremia from those of loss of catabolic renal tissue, the Fg-D and Fg-E turnover studies were repeated on other animals with intact kidneys whose ureters were diverted into the peritoneum and compared to subsequent studies after total nephrectomy. The control and ureter-severed studies had the same clearance pattern, whereas decreased catabolism occurred in the nephrectomized dogs. The results demonstrate uremia per se does not have a major effect upon the catabolism of fibrinogen, Fg-D, and Fg-E. Loss of renal tissue does impair the clearance of Fg-D and Fg-E, indicating these proteins are normally catabolized in part by the kidneys. Thus elevated plasma FRA in uremic patients may reflect decreased Fg-D and Fg-E catabolism rather than increased FDP production from primary or secondary fibrinolysis.  相似文献

Fucoxanthin-containing oil extracted from microalga, Chaetoseros sp., was subjected to genotoxicity studies, the bacterial reverse mutation test and the micronucleus test in mice. The number of revertant colonies in fucoxanthin oil-treated plates of all strains tested was less than twice the number of colonies in the negative control, regardless of the presence of the metabolic activator in the bacterial reverse mutation test. In the micronucleus test, 500, 1,000, 2,000 mg/kg body weight of fucoxanthin oil was administered orally to mice. There was no significant increase in micronucleus frequency in bone marrow cells. These results suggest that fucoxanthin oil does not exhibit genotoxicity.  相似文献

On 16 April 2016, a Mw 7.0 earthquake occurred in Kumamoto city, Japan. The main shock induced two large landslides, namely the Aso Bridge landslide and the Aso Volcanological Laboratory landslide. Their topographical and geological conditions and motion features were investigated by using an unmanned aerial vehicle (UAV) and portable dynamic cone penetration tests (PPTs). The Aso Bridge landslide lies between elevations of 385 m and 725 m, with a total estimated volume of about 1,980,000 m3. The main body is composed of cohesive soil with lapilli and block. The Aso Volcanological Laboratory landslide lies on a slope between 483 m to 582 m, and the total volume is about 81,000 m3, with an average thickness of 4.5 m. The main body is composed of Kusasenrigahama volcanic pumice tephra beds. The material compositions and deposits of both landslides have low cohesion and easily induced shear failure for the two landslides. The sliding distance of the Aso Bridge landslide was long, the sliding direction almost unchanged from the scarp to the toe, and the sliding speed was rapid. The sliding distance of the Aso Volcanological Laboratory landslide, however, was short, the sliding direction changed from the N-direction at the scarp to the NW-direction at the toe, and the sliding speed was slow.

Viscoelastic properties of non-entangled concentrated aqueous solutions of three sodium polystyrenesulfonates with low molecular weights were measured in oscillation flows. Master curves of dynamic moduli were obtained by shifting the moduli data measured at different concentrations along the frequency (ω) axis for each molecular weight. The concentration shift factor used in making the master curves composes a single line irrespective of molecular weight. Storage and loss moduli are proportional to ω2 and ω, respectively, in the terminal region, while they are proportional to ω1/2 in the transition region. All the master curves can be well represented by the modified Rouse theory using the experimental values of concentration, molecular weight and viscosity in a reference state.  相似文献
