Diets based on red clover silage (RCS) typically increase the concentration of polyunsaturated fatty acids (PUFA) in ruminant meat and milk and lower the efficiency of N utilization compared with grass silages (GS). Four multiparous Finnish Ayrshire cows (108 d postpartum) fitted with rumen cannulas were used in a 4 × 4 Latin square design with 21-d periods to evaluate the effect of incremental replacement of GS with RCS on milk production, nutrient digestion, whole-body N metabolism, and milk fatty acid composition. Treatments comprised total mixed rations offered ad libitum, containing 600 g of forage/kg of diet dry matter (DM), with RCS replacing GS in ratios of 0:100, 33:67, 67:33, and 100:0 on a DM basis. Intake of DM and milk yield tended to be higher when RCS and GS were offered as a mixture than when fed alone. Forage species had no influence on the concentration or secretion of total milk fat, whereas replacing GS with RCS tended to decrease milk protein concentration and yield. Substitution of GS with RCS decreased linearly whole-tract apparent organic matter, fiber, and N digestion. Forage species had no effect on total nonammonia N at the omasum, whereas the flow of most AA at the omasum was higher for diets based on a mixture of forages. Replacing GS with RCS progressively lowered protein degradation in the rumen, increased linearly ruminal escape of dietary protein, and decreased linearly microbial protein synthesis. Incremental inclusion of RCS in the diet tended to lower whole-body N balance, increased linearly the proportion of dietary N excreted in feces and urine, and decreased linearly the utilization of dietary N for milk protein synthesis. Furthermore, replacing GS with RCS decreased linearly milk fat 4:0 to 8:0, 14:0, and 16:0 concentrations and increased linearly 18:2n-6 and 18:3n-3 concentrations, in the absence of changes in cis-9 18:1, cis-9, trans-11 18:2, or total trans fatty acid concentration. Inclusion of RCS in the diet progressively increased the apparent transfer of 18-carbon PUFA from the diet into milk, but had no effect on the amount of 18:2n-6 or 18:3n-3 at the omasum recovered in milk. In conclusion, forage species modified ruminal N metabolism, the flow of AA at the omasum, and whole-body N partitioning. A lower efficiency of N utilization for milk protein synthesis with RCS relative to GS was associated with decreased availability of AA for absorption, with some evidence of an imbalance in the supply of AA relative to requirements. Higher enrichment of PUFA in milk for diets based on RCS was related to an increased supply for absorption, with no indication that forage species substantially altered PUFA bioavailability.  相似文献

Our previous study showed that His was the first-limiting amino acid (AA) for milk protein production in cows fed grass silage and cereal-based supplement. The aim of this study was to identify the second-limiting AA and determine whether glucose was limiting responses to His. Abomasal infusion of His (6.5 g/d), glucose (250 g/d), His (6.5 g/d) + glucose (250 g/d), His 6.5 g/d) + Leu (12 g/d) and His (6.5 g/d) + Leu (12 g/d) + glucose (250 g/d) on milk production and utilization of amino acids by mammary gland was in an incomplete 5 x 6 Latin square design with 14-d periods. The diet was based on restrictively fermented grass silage fed ad libitum and 8 kg/d of concentrate comprised of barley, oats, unmolassed sugar beet pulp, urea, and minerals. The infusions did not affect feed intake, diet digestibility, or rumen fermentation pattern. The molar proportion of propionate in rumen VFA was low (15.5%), suggesting that glucose supply from the basal diet could be limiting. Milk and milk protein yields were increased by His infusion. Infusion of His increased plasma His concentration from 19 to 52 microM but decreased extraction efficiency of His. Infusion of glucose increased plasma glucose concentration, milk lactose concentration, and yield and tended to increase milk protein yield. Responses in milk protein yield to combined infusions of His and glucose were additive, suggesting that the utilization of the first-limiting AA His was limited by glucose supply. Infusion of Leu increased plasma Leu concentration but did not produce any further milk protein yield response compared with the infusions without Leu. It was concluded that the efficiency of utilization of the first-limiting AA His could be improved by increasing the supply of glucose, when the basal diet produces a rumen fermentation pattern low in propionate. Leu was not the second-limiting AA in cows fed grass silage-based diets.  相似文献

In this prospective controlled study, the authors examined (a) parenting experiences among couples with successful assisted reproduction treatment (ART; n = 367) and fertile spontaneously conceiving controls (n = 371) and (b) the impact of ante- and perinatal factors and child characteristics on parenting experiences. The results show that positive mothering experiences increased more during the 1st year of parenting and were generally higher among ART mothers than control mothers. No differences were found between ART fathers and controls in their fathering experience. Unpleasant birth experiences, low birth weight, and difficulty soothing the child were associated with high levels of parental stress in the control group, but this was not so among the ART parents. Psychosocial interventions in maternal care should take into account the various meanings that couples give to the history of infertility and conception and ante- and perinatal experiences. Four rumen cannulated Finnish Ayrshire cows in midlactation were used in an experiment designed as a 4 x 5 incomplete Latin square with 2-wk periods to compare effects of glucogenic substrates on grass silage-based diets. The five treatments were continuous infusions of 1) water (control), 2) casein 300 g/d, 3) glucose 300 g/d, 4) propionic acid 247 g/d, and 5) barley starch 270 g/d. Substrates were infused either into the rumen (propionic acid) or into the abomasum (other substrates). As a basal diet, cows were fed a formic acid treated grass silage ad libitum (digestible organic matter 690 g/kg dry matter [DM], crude protein [CP] 131 g/kg DM) and a barley-rapeseed concentrate (CP 141g/kg DM) at a rate of 7 kg/d. Production responses to glucogenic substrates other than casein were negligible, suggesting that glucose supply of the cows did not primarily limit milk production. However, with casein cows produced significantly more milk, milk protein, and lactose than with other glucogenic substrates. Casein increased urea and essential amino acid (EAA), and decreased nonessential AA (NEAA) in arterial plasma compared with other substrates, suggesting that casein provided precursors both in terms of NEAA for gluconeogenesis and EAA for milk protein synthesis. This puts forward that providing the AA needs of the mammary gland for milk protein synthesis are met, glucose supply may become the next limiting factor for milk protein synthesis in cows fed diets based on restrictively fermented grass silage. The limited supply of AA from the basal diet, and possibly the low production levels of cows partly invalidated the hypothesis of monitoring differing glucogenic substrates for grass silage-based diets.  相似文献

Five Finnish ruminally cannulated Ayrshire cows were used in a 5 x 5 Latin square trial with 14-d periods to determine whether branched-chain amino acids (AA) are the second- or colimiting AA for milk protein synthesis on grass silage-cereal based diet. Mammary metabolism of AA as well as AA supply from the basal diet were also studied. Grass silage (17.5% crude protein) was given ad libitum with 9 kg/d as a cereal-based concentrate (13.8% crude protein). Treatments were basal diet without AA infusion (Control), abomasal infusion of AA mixture of His, Ile, Leu, and Val at 8.5, 14.9,27.9, and 18.3 g/d, respectively, AA mixture minus Ile, AA mixture minus Leu, and AA mixture minus Val. Glucose was infused on all treatments at 250 g/d. Amino acid infusions had no effect on dry matter intake (mean 19.2 kg/d), yields of milk (mean 25.3 kg/d), energy-corrected milk (mean 25.9 kg/d), milk protein (mean 807 g/d), lactose (mean 1261 g/d), or fat (mean 1056 g/d). Milk composition was not affected by the treatments. Plasma concentrations of His and Val responded to AA infusions but concentration of Ile increased only on treatment AA mixture minus Leu, and concentration of Leu only on treatment AA mixture minus Ile. Infusion of AA mixture of His, Ile, Leu, and Val decreased plasma concentrations of Arg, Lys, Met, Phe, and Tyr. Amino acid infusions did not affect concentrations of plasma urea and energy metabolites or AA utilization by the mammary gland. Based on unchanged production parameters, the supply of His or branched-chain AA seemed not to be limiting under the current dietary conditions. Changes in plasma AA concentrations suggest either antagonism between individual AA in absorption or increased partitioning of AA into the muscle tissues. About 75% of omasal canal nonammonia nitrogen flow (427 g/d) was of microbial origin, and AA profiles of microbial protein and omasal canal digesta were fairly similar. The current modulation characteristics of a short external cavity tunable laser are presented. The 1540 nm Fabry-Perot diode laser is wavelength-tuned by the electrostatically controlled silicon micromachined Fabry-Perot interferometer device. OBJECTIVES: To investigate the molecular epidemiology and genetic structure of the virus strain(s) causing an outbreak of HIV-1 infection in the Kaliningrad province of the Russian Federation and to investigate the relationship of this outbreak to some other emerging HIV-1 epidemics in the countries of the former Soviet Union. DESIGN: A molecular epidemiological investigation was conducted in the city of Kaliningrad amongst individuals recently diagnosed as HIV-1-positive. Samples were also collected from neighbouring Lithuania and from the Ukraine. METHODS: Incident and population data was collected from official health statistics in Kaliningrad. A standardized questionnaire was administered to newly diagnosed individuals to assess risk factors for HIV-1 infection. For genotyping, two regions of the virus (env C2-V3 and gag NCp7) were directly sequenced. RESULTS: The number of newly diagnosed individuals testing seropositive for HIV-1 infection in Kaliningrad rose from less than one per month to more than 100 per month during the period of July-October 1996. A total of 1335 new infections were identified between 1 July 1996 and 30 June 1997. The main reported risk factor for HIV-1 infection (80%) was injecting drug use, in particular with a locally produced opiate. Sequence analysis of patient viruses in Kaliningrad (n = 50) showed that the epidemic was caused by a highly homogenous HIV-1 strain, recombinant between the genetic subtypes A and B. Comparison with subtype A strains prevalent amongst injecting drug users (IDU) in the Ukraine showed that one of these strains was the direct subtype A parent of the epidemic A/B recombinant strain in Kaliningrad. CONCLUSIONS: The HIV-1 epidemic in Kaliningrad probably started from a single source, with rapid spread of the virus through the IDU population. The origin of the epidemic strain is a recombination event occurring between the subtype A strain virus prevalent among IDU in some southern CIS countries, and a subtype B strain of unknown origin.  相似文献

The present study was conducted to investigate ruminal N metabolism in dairy cows using 15N-labeled N sources and dynamic models. The data summarized in this study were obtained from 2 of 4 treatments whose effects were determined in a 4 × 4 Latin square design. Soluble N (SN) isolated from timothy grass silage labeled with 15N and ammonia N (AN) labeled with 15N were administered into the rumen contents of 4 ruminally cannulated dairy cows. Ruminal N pool sizes were determined by manual evacuation of rumen contents. The excess 15N-atom% was determined in N-fractions of rumen digesta grab samples that were collected frequently between 0 to 72 h and used to determine 15N metabolism in the rumen. Calculations of area under the curve ratios of 15N were used to estimate proportions of N fractions originating from precursor N pools. A model including soluble nonammonia N (SNAN), AN, bacterial N, and protozoal N pools was developed to predict observed values of 15N atomic excess pool sizes. The model described the pool sizes accurately based on small residuals between observed and predicted values. An immediate increase in 15N enrichment of protozoal N suggests physical attachment of bacteria pool to protozoa pool. The mean proportions of bacterial N, protozoal N, and feed N in rumen solid phase were 0.59, 0.20, and 0.21, respectively. These observations suggest that protozoal N accounted for 0.25 of rumen microbial N. About 0.90 of the initial dose of AN was absorbed or taken up by microbes within 2 h. Faster 15N enrichment of bacterial N with SN than with AN treatment indicates a rapid adsorption of SNAN to microbial cells. Additionally, the recovery of 15N as microbial and feed N flow from the rumen was approximately 0.36 greater for SN than for the AN treatment, indicating that SNAN was more efficiently used for microbial growth than AN. The present study indicated that about 0.15 of microbial N flowing to the duodenum was of protozoal origin and that 0.95 of the protozoal N originated from engulfed bacterial N. The kinetic variables indicated that 0.125 of SNAN escaped ruminal degradation, which calls into question the use of in situ estimations of protein degradation to predict the flow of rumen undegradable protein.  相似文献

该试验研究磨浆时间对玉米波杂颗粒微观结构均匀性的影响,并以分层率和离心沉淀率为评价指标,探讨不同增稠剂的稳定效果,解决玉米波杂储藏过程中分层、口感粗糙等问题。通过单因素试验及正交试验,优化复合增稠剂的配比。结果表明,最适磨浆时间为80 s,最佳增稠剂配比为黄原胶0.02%,海藻酸钠0.02%,卡拉胶0.03%。在(4±1) ℃冷藏25 d无分层,第30天的分层率为0.6%,离心沉淀率为50%,玉米波杂口感细腻,稳定性良好。  相似文献
