Growth hormone (GH) has recently been shown to affect polymorphonuclear neutrophil granulocyte (PMN) function and to be secreted by mononuclear cells, indicating that the hormone may be active in an immunophysiologic network, acting as an endo- or paracrine priming agent. The purpose of the present study was to evaluate the chemotactic responsiveness of canine peripheral PMN in a dog with acromegaly, caused by spontaneous, progesterone-induced hypersecretion of GH and, secondary to this, a seven-fold increase in insulin-like growth factor I (IGF-I). The chemotactic responsiveness towards zymosan-activated serum (ZAS) and leukotriene B4 (LTB4) was evaluated at a time when the dog suffered from acromegaly and again 57 days after corrective surgery (ovariohysterectomy). The experiments showed that PMN from the patient exhibited enhanced chemotactic migration that appeared to be associated with the hypersomatotropic condition as judged from the reversibility of the phenomenon. The glucose intolerance and elevated serum alkaline phosphatase that were observed in the acromegalic dog were also shown to be reversible following surgery.  相似文献

Intensive fertilizer use has led to nitrate contamination of groundwater in many areas. This article examines the possibility of removing nitrates through the construction of artificial wetlands. A pilot study conducted in Denmark has found that the method is effective and economical, at least for small waterworks, though further treatment is necessary before an acceptable microbiological water quality is attained.  相似文献

The G-protein coupled receptor GPR39 is abundantly expressed in various tissues and can be activated by changes in extracellular Zn2+ in physiological concentrations. Previously, genetically modified rodent models have been able to shed some light on the physiological functions of GPR39, and more recently the utilization of novel synthetic agonists has led to the unraveling of several new functions in the variety of tissues GPR39 is expressed. Indeed, GPR39 seems to be involved in many important metabolic and endocrine functions, but also to play a part in inflammation, cardiovascular diseases, saliva secretion, bone formation, male fertility, addictive and depression disorders and cancer. These new discoveries offer opportunities for the development of novel therapeutic approaches against many diseases where efficient therapeutics are still lacking. This review focuses on Zn2+ as an endogenous ligand as well as on the novel synthetic agonists of GPR39, placing special emphasis on the recently discovered physiological functions and discusses their pharmacological potential.  相似文献

Interactive topology optimization on hand-held devices   总被引:1,自引:1,他引:0  
This paper presents an interactive topology optimization application designed for hand-held devices running iOS or Android. The TopOpt app solves the 2D minimum compliance problem with interactive control of load and support positions as well as volume fraction. Thus, it is possible to change the problem settings on the fly and watch the design evolve to a new optimum in real time. The use of an interactive app makes it extremely simple to learn and understand the influence of load-directions, support conditions and volume fraction. The topology optimization kernel is written in C# and the graphical user interface is developed using the game engine Unity3D. The underlying code is inspired by the publicly available 88 and 99 line Matlab codes for topology optimization but does not utilize any low-level linear algebra routines such as BLAS or LAPACK. The TopOpt App can be downloaded on iOS devices from the Apple App Store, at Google Play for the Android platform, and a web-version can be run from www.topopt.dtu.dk.  相似文献

Tests comparing image sets can play a critical role in PET research, providing a yes-no answer to the question "Are two image sets different?" The statistical goal is to determine how often observed differences would occur by chance alone. We examined randomization methods to provide several omnibus test for PET images and compared these tests with two currently used methods. In the first series of analyses, normally distributed image data were simulated fulfilling the requirements of standard statistical tests. These analyses generated power estimates and compared the various test statistics under optimal conditions. Varying whether the standard deviations were local or pooled estimates provided an assessment of a distinguishing feature between the SPM and Montreal methods. In a second series of analyses, we more closely simulated current PET acquisition and analysis techniques. Finally, PET images from normal subjects were used as an example of randomization. Randomization proved to be a highly flexible and powerful statistical procedure. Furthermore, the randomization test does not require extensive and unrealistic statistical assumptions made by standard procedures currently in use.  相似文献

Despite the existence of patterns able to discriminate between consensual and non-consensual intercourse, the relevance of genital lesions in the corroboration of a legal rape complaint is currently under debate in many countries. The testimony of the physicians when assessing these lesions has been questioned in court due to several factors (e.g., a lack of comprehensive knowledge of lesions, wide spectrum of background area, among others). Therefore, it is relevant to provide automated tools to support the decision process in an objective manner. In this work, we evaluate the performance of state-of-the-art deep learning architectures for the forensic assessment of sexual assault. We propose a deep architecture and learning strategy to tackle the class imbalance on deep learning using ranking. The proposed methodologies achieved the best results when compared with handcrafted feature engineering and with other deep architectures.  相似文献

A new potassium tetraamidoboranealuminate, K[Al(NH2BH3)4], has been synthesized by a mechanochemical reaction between KAlH4 and NH3BH3. The compound, K[Al(NH2BH3)4], crystallizes in a triclinic unit cell with space group symmetry P?1. The crystal structure consists of [K(NH2BH3)6]5? octahedra which facilitate the bridging between K+ in 1D chains, while also bridging K+ to Al3+ to connect the 1D chains in a 3D network. Thermal analysis reveals that K[Al(NH2BH3)4] decomposes in two exothermic steps at T ~ 94 and 138 °C and releases primarily hydrogen. The total gas release amounts to ~6.0 wt% H2. The decomposition products are investigated ex situ by powder X-ray diffraction, infrared spectroscopy, and 11B and 27Al NMR and identified as KBH4 and amorphous phases, possibly BN3, N2BH, and/or NBH2 whereas aluminum is found in four-, five-, and six-fold coordination. Unfortunately, the decomposed sample shows no hydrogen absorption at T = 260 °C and p(H2) = 110 bar.  相似文献

By mapping the small-angle X-ray scattering (SAXS) from a looped poly-(para-phenylene terephtalamide) (aramid, PPTA) filament using a synchrotron X-ray microbeam, we investigate the effects of axially compressive and tensile strain on internal fibrillar structures. Unique observations of oscillations in the highly anisotropic SAXS patterns indicate a regular internal structure. Upon increase of the applied compressive strain, a significant decrease in oscillation frequency is observed in the scattering pattern. With an increase in imposed tensile strain an increase in oscillation frequency in the scattering pattern is observed. One model capable of describing the intensity is a model of stacked cylinders. These cylinders could be part of the fibrillar structure present in the PPTA fibres, which consist of cylinder- to tape-like objects, the presence of which is supported by SEM images. One hypothetical physical interpretation presented here for the appearance of a regular internal structure is the occurrence of fibrillar separation in regions undergoing axially compressive strain, and the appearance of strain relief/slip planes between packs of fibrils in regions undergoing tensile strain. Compaction of these packs upon increase of tensile strain could increase the repetition distance in the structure as multiple packs combine to form a single pack.  相似文献

Insulin-like growth factor-1 (IGF-1) is a serum protein whichunexpectedly folds to yield two stable tertiary structures withdifferent disulphide connectivities; native IGF-1 [18–61,6–48,47–52]and IGF-1 swap [18–61,6–47, 48–52]. Here we demonstratein detail the biological properties of recombinant human nativeIGF-1 and IGF-1 swap secreted from Saccharomyces cerevisiae.IGF-1 swap had a ~30 fold loss in affinity for the IGF-1 receptoroverexpressed on BHK cells compared with native IGF-1.The parallelincrease in dose required to induce negative cooperativity togetherwith the parallel loss in mitogenicity in NIH 3T3 cells impliesthat disruption of the IGF-1 receptor binding interaction ratherthan restriction of a post-binding conformational change isresponsible for the reduction in biological activity of IGF-1swap. Interestingly, the affinity of IGF-1 swap for the insulinreceptor was ~200 fold lower than that of native IGF-1 indicatingthat the binding surface complementary to the insulin receptor(or the ability to attain it) is disturbed to a greater extentthan that to the IGF-1 receptor. A 1.0 ns high-temperature moleculardynamics study of the local energy landscape of IGF-1 swap resultedin uncoiling of the first A-region -helix and a rearrangementin the relative orientation of the A- and B-regions. The modelof IGF-1 swap is structurally homologous to the NMR structureof insulin swap and CD spectra consistent with the model arepresented. However, in the model of IGF-1 swap the C-regionhas filled the space where the first A-region -helix has uncoiledand this may be hindering interaction of Val44 with the secondinsulin receptor binding pocket.  相似文献
