BALB/c mice were fed milk or Lactobacillus casei BL23 in milk for 14 d and fecal samples were collected at d 0, 4, and 7 as well as 1 and 8 d after the last administration. According to high-throughput DNA sequencing of the 16S rRNA genes extracted from the fecal microbiota, the bacterial diversity in the fecal samples of all mice increased over time. After 14 d of administration, the consumption of milk and milk containing L. casei BL23 resulted in distinct effects on the microbial composition in the intestine. Specifically, the proportions of bacteria in the Lactobacillaceae, Porphyromonadaceae, and Comamonadaceae were significantly higher in mice fed the L. casei BL23-milk culture compared with one or more of the other groups of mice. The relative amounts of Lachnospiraceae were higher and Streptococcaceae were lower in mice fed milk alone. The changes were not found at d 4 and 7 during milk and L. casei feeding and were no longer detected 8 d after administration was stopped. This study shows that consumption of milk or probiotic L. casei-containing milk results in non-overlapping, taxa-specific effects on the bacteria in the distal murine intestine.  相似文献

Thyromimetics, whose physicochemical characteristics are analog to thyroid hormones (THs) and their derivatives, are promising candidates as novel therapeutics for neurodegenerative and metabolic pathologies. In particular, sobetirome (GC-1), one of the initial halogen-free thyromimetics, and newly synthesized IS25 and TG68, with optimized ADME-Tox profile, have recently attracted attention owing to their superior therapeutic benefits, selectivity, and enhanced permeability. Here, we further explored the functional capabilities of these thyromimetics to inhibit transthyretin (TTR) amyloidosis. TTR is a homotetrameric transporter protein for THs, yet it is also responsible for severe amyloid fibril formation, which is facilitated by tetramer dissociation into non-native monomers. By combining nuclear magnetic resonance (NMR) spectroscopy, computational simulation, and biochemical assays, we found that GC-1 and newly designed diphenyl-methane-based thyromimetics, namely IS25 and TG68, are TTR stabilizers and efficient suppressors of TTR aggregation. Based on these observations, we propose the novel potential of thyromimetics as a multi-functional therapeutic molecule for TTR-related pathologies, including neurodegenerative diseases.  相似文献

This study seeks to investigate the impact of an exposure to a university's social media on publics’ perceptions during a crisis. Data from this study showed that participants (= 269) expressed more favourable evaluations towards the university and their perceived severity was mitigated after viewing its official statement on university's Facebook posts. The results of this empirical research offer a meaningful suggestion to crisis communication scholars and practitioners interested in assessing the impact of universities’ statements in social media by providing ways to understand the importance of the organizational usage of social media during a crisis, why that is important, and why practitioners should make effective social media communication tactics a priority during a crisis.  相似文献

Novel sphingolipid metabolite conjugated poly(amino acid)-derivative, poly-α,β-(2-hydroxylethyl l-aspartamide)-g-phytosphingosine copolymer, was designed as an anticancer prodrug-type carrier for enhanced intracellular uptake and physicochemical properties of polymeric micelle-like aggregates were evaluated. The resultant micelle-like aggregates showed a spherical shape and uniform size with a diameter less than 20 nm in an aqueous solution upon the increased number of phytosphingosine grafts. By the steady-state fluorescence of pyrene in aqueous polymer solutions, critical aggregation concentration (CAC) was obtained in the range of 0.166–0.0036 mg/ml and Kv, equilibrium partition constants of the pyrene in the self-aggregate solutions were estimated to be from 2.39 × 103 to 1.96 × 106. Phytosphingosine-grafted polymeric micelle-like aggregates as small as 20 nm were efficiently delivered into various cancer cell lines including oral, breast and colon carcinoma with the extent which is comparable to the level of targeting carrier system. The enhanced cellular uptake and anticancer therapeutic effect was evaluated by flow cytometry, confocal laser scanning microscopy (CLSM), and MTT assay.  相似文献

Major efforts are underway to harness motor proteins for technical applications. Yet how to best attach cargo to microtubules that serve as kinesin-driven "molecular shuttles" without compromising transport performance remains challenging. Furthermore, microtubule-associated proteins (MAPs) can block motor protein-powered transport in neurons, which can lead to neurodegenerative diseases. Again it is unclear how different physical roadblock parameters interfere with the stepping motion of kinesins. Here, we employ a series of MAPs, tailored (strept)avidins, and DNA as model roadblocks and determine how their geometrical, nanomechanical, and electrochemical properties can reduce kinesin-mediated transport. Our results provide insights into kinesin transport regulation and might facilitate the choice of appropriate cargo linkers for motor protein-driven transport devices.  相似文献

A new sensitivity controllable pixel structure is proposed for CMOS active-pixel image sensor. The proposed pixel structure has a sensitivity control gate overlaid on the photodiode. The sensitivity of the pixel is controlled by the bias voltage of the control gate that forms a variable accumulation-mode MOS capacitor. The prototype sensor is fabricated with a 0.35-mum CMOS process and consists of 60 times 240 pixels with 5.6-mum pixel pitch. Measurement results show that the sensitivity of the photodiode can be controlled by a factor of 4.  相似文献

We demonstrate three different transfer patterns that can be achieved by use of a surface reconstructed block copolymer film where metal is evaporated onto the surface of the film, providing the contrast. Thin films of diblock copolymers having cylindrical microdomains oriented normal to the surface with long-range lateral order were used. Solvent reconstruction of the film, followed by a glancing angle metal evaporation and thermal annealing, led to three different decorations of the films with gold. These films were used as masks for pattern transfer of pores, columns, and rings to underlying substrate with high fidelity.  相似文献
