By protein engineering we have investigated changes to two aminoacid residues (Trp93 and Ser48) in the substrate pocket of yeastalcohol dehydrogenase 1. Upon changing Thr48 to serine we producedan enzyme which has markedly greater activity towards aliphaticalcohols with chain length up to 8, together with a generalincrease in catalytic activity (V/K). Changes at position 93were less pronounced, with the Phe enzyme being more activethan the parent towards the range of alcohols but with the alanineenzyme showing very little difference from the wild-type. Enzymeswith the double changes at 48 and 93 showed increased activitytowards alcohols with 3–8 carbons but the increases werenot additive over the single changes. The enzymes with changesat the two positions would metabolize both stereoisomers of2-octanol whereas the parent ADH would attack only one of them.None of the engineered enzymes would attack cyclohexanol oraromatic alcohols. The results are in general agreement withthe prediction that reducing the size of amino acids in thesubstrate pocket would enhance the ability to oxidize alcoholslarger than ethanol.  相似文献

Biaxial creep behaviors of stress-relieved and recrystallized thin-walled tubing of Ti-3Al-2.5V and Zircaloy-4 are considered under equal hoop and axial stresses by internal pressurization superimposed with axial load. Both hoop and axial strains were monitored and the ratio of the strain rate along the hoop to that along the axial directions is considered to represent the degree of anisotropy. In both of these alloys, relatively weak hoop direction in cold-worked stress-relieved (CWSR) materials became slightly stronger following recrystallization. Crystallographic texture was considered in terms of x-ray pole figures from which the crystallite orientation distribution functions (CODFs) were derived and crystal plasticity model with slip dominant on prism planes was combined with the CODFs to predict the creep anisotropy. While good correlation was noted for recrystallized materials, distinct deviations are observed for CWSR that are believed to arise from grain shape anisotropy. The relatively small (equiaxed) grain size along the hoop and radial directions results in grain boundary sliding leading to stress enhancements along these directions. This anisotropy in grain boundary sliding is shown to explain the observed deviations.  相似文献

The aim of the study was to investigate the effect of a sustained release of bone morphogenetic protein2 (BMP-2) incorporated in a polymeric implant coating on bone healing. In vitro analysis revealed a sustained, but incomplete BMP-2 release until Day 42. For the in vivo study, the rat tibia osteotomy was stabilized either with control or BMP-2 coated wires, and the healing progress was followed by micro computed tomography (μCT), biomechanical testing and histology at Days 10, 28, 42 and 84. MicroCT showed an accelerated formation of mineralized callus, as well as remodeling and an increase of mineralized/total callus volume (p = 0.021) at Day 42 in the BMP-2 group compared to the control. Histology revealed an increased callus mineralization at Days 42 and 84 (p = 0.006) with reduced cartilage at Day 84 (p = 0.004) in the BMP-2 group. Biomechanical stiffness was significantly higher in the BMP-2 group (p = 0.045) at Day 42. In summary, bone healing was enhanced after sustained BMP-2 application compared to the control. Using the same drug delivery system, but a burst release of BMP-2, a previous published study showed a similar positive effect on bone healing. Distinct differences in the healing outcome might be explained due to the different BMP release kinetics and dosages. However, further studies are necessary to adapt the optimal release profiles to physiological mechanisms.  相似文献

Efforts to generate detectable concentrations of free radicals in explosives, binders, and their mixtures are described. Radicals were readily produced in polycaprolactone and polyethylene glycol binders at liquid nitrogen temperature using stresses as low as 0.4 kbar. These radicals were all of the peroxy type, and presumably formed by reaction of mechano-radicals with oxygen present in the polymer. No mechano-radicals were observed from HMX or RDX using samples cooled to liquid nitrogen temperature and applied stresses up to 4 kbar. In neither impacted samples that failed to explode nor the residues remaining after a partial explosion were radicals detected by ESR. Low temperature γ-irradiation of these materials was also carried out. Free radical signals originating in both the polymer and the explosive could be identified. The reactivity of NO2 radicals from γ-irradiated HMX is enhanced in the presence of binder. In γ-irradiated HMX/polycaprolactone mixtures, the NO2 radical anneals rapidly at 150 K, versus 240 K for HMX alone. Evidence is presented to suggest that the relatively stable NO2 radical (such as produced by γ-irradiation) in HMX does not play a dominant role in mechanical initiation.  相似文献

Old age, adiposity, and metabolic disorders are known as risk factors for chronic tendinopathy, which is a common problem in both athletes and the general population. However, the importance of these influencing factors has not yet been well understood. This study investigated alterations in gene expression and histology of Achilles tendons of young (10 weeks) and old (100 weeks) rats bred for low (low capacity runners, LCR) and high (high capacity runners, HCR) intrinsic aerobic exercise capacity. In this rat model, LCR displayed a phenotype of reduced exercise capacity, higher body weight, and metabolic dysfunctions compared to HCR. We hypothesized that the risk factors for tendinopathy in old LCR could lead to more pronounced impairments in Achilles tendon tissue. In quantitative real-time PCR (qPCR), age-related downregulation of tenocyte markers e.g., tenomodulin, genes related to matrix modeling and remodeling (e.g., collagens, elastin, biglycan, fibronectin, tenascin C) as well as transforming growth factor beta 3 (Tgfb3) have been detected. Inflammation marker cyclooxygenase 2 (Cox2) was downregulated in old rats, while microsomal prostaglandin E synthase 2 (Ptges2) was upregulated in old HCR and old LCR. In all groups, interleukin 6 (Il6), interleukin 1 beta (Il1b), and tumor necrosis factor alpha (Tnfa) showed no significant alteration. In histological evaluation, tendons of old rats had fewer and more elongated tenocyte nuclei than young rats. Even though a higher content of glycosaminoglycans, a sign of degeneration, was found in old HCR and LCR, no further signs of tendinopathy were detectable in tendons of old rats by histological evaluation. Low intrinsic aerobic exercise capacity and the associated phenotype did not show significant effects on gene expression and tendon histology. These findings indicate that aging seems to play a prominent role in molecular and structural alterations of Achilles tendon tissue and suggests that other risk factors associated with intrinsic aerobic exercise capacity are less influential in this rat model.  相似文献

The relationships between transient vehicle operation and ultrafine particle emissions are not well-known, especially for low-emission alternative bus technologies such as compressed natural gas (CNG) and diesel buses equipped with particulate filters/traps (TRAP). In this study, real-time particle number concentrations measured on a nominal 5 s average basis using an electrical low pressure impactor (ELPI) for these two bus technologies are compared to that of a baseline catalyst-equipped diesel bus operated on ultralow sulfur fuel (BASE) using dynamometer testing. Particle emissions were consistently 2 orders of magnitude lower for the CNG and TRAP compared to BASE on all driving cycles. Time-resolved total particle numbers were examined in terms of sampling factors identified as affecting the ability of ELPI to quantify the particulate matter number emissions for low-emitting vehicles such as CNG and TRAP as a function of vehicle driving mode. Key factors were instrument sensitivity and dilution ratio, alignment of particle and vehicle operating data, sampling train background particles, and cycle-to-cycle variability due to vehicle, engine, after-treatment, or driver behavior. In-cycle variability on the central business district (CBD) cycle was highest for the TRAP configuration, but this could not be attributed to the ELPI sensitivity issues observed for TRAP-IDLE measurements. Elevated TRAP emissions coincided with low exhaust temperature, suggesting on-road real-world particulate filter performance can be evaluated by monitoring exhaust temperature. Nonunique particle emission maps indicate that measures other than vehicle speed and acceleration are necessary to model disaggregated real-time particle emissions. Further testing on a wide variety of test cycles is needed to evaluate the relative importance of the time history of vehicle operation and the hysteresis of the sampling train/dilution tunnel on ultrafine particle emissions. Future studies should monitor particle emissions with high-resolution real-time instruments and account for the operating regime of the vehicle using time-series analysis to develop predictive number emissions models.  相似文献

We evaluated short-term systemic antimicrobial prophylaxis for catheter-associated bacteriuria in women undergoing elective gynecological operations in a prospective, controlled, double-masked study. Nine of 100 placebo-treated patients acquired bacteriuria during catheterization compared with 3 of 96 of the drug-treated group. However, at the time of hospital discharge, clean-voided urine specimens were positive as frequently in the drug-treated group (8 of 82 patients cultured) as in the placebo group (8 of 75 patients cultured). No difference in febrile morbidity due to bacteriuria was noted between the prophylaxis and placebo groups. The incidence of catheter-associated bacteriuria may be reduced by antimicrobial prophylaxis. However, because the protective effect is transient and is associated with the selection of resistant organisms, prophylaxis is not indicated for patients at low risk for acquired bacteriuria and in whom the sequelae of catheter-associated infections are infrequent.  相似文献
