Collagen I Modifies Connexin-43 Hemichannel Activity via Integrin α2β1 Binding in TGFβ1-Evoked Renal Tubular Epithelial Cells

Chronic Kidney Disease (CKD) is associated with sustained inflammation and progressive fibrosis, changes that have been linked to altered connexin hemichannel-mediated release of adenosine triphosphate (ATP). Kidney fibrosis develops in response to increased deposition of extracellular matrix (ECM), and up-regulation of collagen I is an early marker of renal disease. With ECM remodeling known to promote a loss of epithelial stability, in the current study we used a clonal human kidney (HK2) model of proximal tubular epithelial cells to determine if collagen I modulates changes in cell function, via connexin-43 (Cx43) hemichannel ATP release. HK2 cells were cultured on collagen I and treated with the beta 1 isoform of the pro-fibrotic cytokine transforming growth factor (TGFβ1) ± the Cx43 mimetic Peptide 5 and/or an anti-integrin α2β1 neutralizing antibody. Phase microscopy and immunocytochemistry observed changes in cell morphology and cytoskeletal reorganization, whilst immunoblotting and ELISA identified changes in protein expression and secretion. Carboxyfluorescein dye uptake and biosensing measured hemichannel activity and ATP release. A Cytoselect extracellular matrix adhesion assay assessed changes in cell-substrate interactions. Collagen I and TGFβ1 synergistically evoked increased hemichannel activity and ATP release. This was paralleled by changes to markers of tubular injury, partly mediated by integrin α2β1/integrin-like kinase signaling. The co-incubation of the hemichannel blocker Peptide 5, reduced collagen I/TGFβ1 induced alterations and inhibited a positive feedforward loop between Cx43/ATP release/collagen I. This study highlights a role for collagen I in regulating connexin-mediated hemichannel activity through integrin α2β1 signaling, ahead of establishing Peptide 5 as a potential intervention.

A scalable approach to mapping annual land cover at 250 m using MODIS time series data: A case study in the Dry Chaco ecoregion of South America

Land use and land cover (LULC) maps from remote sensing are vital for monitoring, understanding and predicting the effects of complex human-nature interactions that span local, regional and global scales. We present a method to map annual LULC at a regional spatial scale with source data and processing techniques that permit scaling to broader spatial and temporal scales, while maintaining a consistent classification scheme and accuracy. Using the Dry Chaco ecoregion in Argentina, Bolivia and Paraguay as a test site, we derived a suite of predictor variables from 2001 to 2007 from the MODIS 250 m vegetation index product (MOD13Q1). These variables included: annual statistics of red, near infrared, and enhanced vegetation index (EVI), phenological metrics derived from EVI time series data, and slope and elevation. For reference data, we visually interpreted percent cover of eight classes at locations with high-resolution QuickBird imagery in Google Earth. An adjustable majority cover threshold was used to assign samples to a dominant class. When compared to field data, we found this imagery to have georeferencing error < 5% the length of a MODIS pixel, while most class interpretation error was related to confusion between agriculture and herbaceous vegetation. We used the Random Forests classifier to identify the best sets of predictor variables and percent cover thresholds for discriminating our LULC classes. The best variable set included all predictor variables and a cover threshold of 80%. This optimal Random Forests was used to map LULC for each year between 2001 and 2007, followed by a per-pixel, 3-year temporal filter to remove disallowed LULC transitions. Our sequence of maps had an overall accuracy of 79.3%, producer accuracy from 51.4% (plantation) to 95.8% (woody vegetation), and user accuracy from 58.9% (herbaceous vegetation) to 100.0% (water). We attributed map class confusion to limited spectral information, sub-pixel spectral mixing, georeferencing error and human error in interpreting reference samples. We used our maps to assess woody vegetation change in the Dry Chaco from 2002 to 2006, which was characterized by rapid deforestation related to soybean and planted pasture expansion. This method can be easily applied to other regions or continents to produce spatially and temporally consistent information on annual LULC.

The Shopping Jacket: Wearable computing for the consumer

As part of the Bristol Wearable Computing Initiative, we are exploring location-sensing systems suitable for use with wearable computing. In this paper we present our findings, and in particular a wearable application — the ‘Shopping Jacket’ — which relies on a minimal infrastructure to be effective. We use two positioning devices, ‘pingers’ and GPS. The pinger is used to signal the presence of a shop, and to indicate the type of shop and its website. The GPS is used to disambiguate which branch of a high street chain is being passed. The wearable uses this information to determine whether the wearer needs to be alerted that they are passing an interesting shop, or to direct the wearer around a shopping mall. The Shopping Jacket integrates a wearable CardPC, GPS and pinger receivers, a near-field radio link, hand-held display, GSM data telephone and a speech interface into a conventional sports blazer.     

Euclidean controllable realizations of linear hereditary systems

A realization problem for a class of linear hereditary systems is formulated and state space representations are constructed from inputoutput relations. The resulting hereditary models are shown to be Euclidean controllable, and comparisons are made between these hereditary realizations and other recently developed theories.This research was supported in part by the Air Force Flight Dynamics Laboratory under grant AFOSR-77-3221.     

Planning in the Ethno-culturally Diverse City: A Comment

This article examines how major retailers, in their attempts to secure sites for very large stores (over 7000 m2 sales area), have had to adopt new strategies in order to overcome resistance from local authority and central government retail planning policy. These strategies include recycling of old retail developments with ‘open A1 consent’, participation in urban regeneration projects and development or redevelopment of ‘district shopping centres’ in urban areas. Discussion of these strategies reveals weaknesses and inconsistencies in retail planning policy in the UK.
