This work demonstrates the efficiency of almond gum polysaccharides (AGPs) as bioactive compounds. AGPs were first extracted using H2O2, in the presence of NaOH, at different times and temperatures. The optimal extraction conditions were 4% H2O2 and 2 N NaOH, for 7 h at 50 °C, leading to an extraction yield of 58.2% (w/w). After a purification step, the retained AGPs were characterised using high‐performance liquid chromatography showing a molecular weight of 99.3 kDa. The monosaccharide composition of AGPs were assessed using gas chromatography–mass spectrometry. AGPs were found to be a complex heteropolysaccharide with a repeating unit mainly composed of galactose, arabinose, xylose, mannose, rhamnose, and glucuronic acid with the respective ratios: 45:26:7:10:1:11. The acidic nature of the polysaccharide is due to the presence of glucuronic acid. Total antioxidant activity, free radical‐scavenging activity and reducing power assay of AGPs were investigated. The obtained results showed high antioxidant activities of AGPs. Furthermore, beyond 60 mg mL?1, AGPs exhibited bacterial growth inhibition for five pathogenic strains: Escherichia coli, Staphylococcus aureus, Enterococcus feacalis, Pseudomonas aeruginosa and Salmonella typhimurium.  相似文献

Oligodendrocytes and their myelin membranes are the apparent target of the autoimmune response that characterizes the human adult central nervous system-demyelinating disease multiple sclerosis. Human oligodendrocytes do not express MHC class II molecules, a requirement for MHC-restricted injury mediated by myelin-reactive CD4+ T cells, the cell type implicated in initiating the disease process. In this study we observed that human adult central nervous system-derived oligodendrocytes can be susceptible to non-MHC-restricted lysis mediated by myelin basic protein-reactive CD4+ T cell lines. Cytotoxicity was significantly greater (37 +/- 4 vs 7 +/- 3%) with cell lines in which a high proportion of cells (mean, 28 +/- 6%) expressed CD56 compared with cytotoxicity mediated by low CD56 cell lines (8 +/- 2%). High CD56 cell lines, when rested in IL-2, lost cytotoxic activity and had reduced expression of CD56 (mean, 5 +/- 2%). CD4+ T cells isolated from short term (3-day) anti-CD3/IL-2-activated mononuclear cell cultures did not express CD56 and were not cytotoxic to oligodendrocytes unless lectin was added. In contrast, an enriched population of non-T cells extracted from the same activated MNC cultures expressed CD56 as well as other NK cell-associated surface molecules and was cytotoxic. These results indicate the potential susceptibility of human oligodendrocytes to non-MHC-restricted injury mediated by both Ag-reactive and nonspecific cellular immune effector cells, with CD56 expression being a common feature of the effector cells.  相似文献

The room temperature slow crack growth resistance in air ( 50% relative humidity) and in water for large cracks in MgO-partially stabilized zircoba (PSZ) improves with increase in critical fracture toughness,K lc. Ageing the as-fired PSZ at 1400° C for 8 h results in decreasing Klc from 8.5 MPa m1/2 to 6 M Pa M1/2. The ageing treatment also promotes the growth of eutectoid decomposition products on grain boundaries that is accompanied by a decrease in the dependence (A the change in Region I crack velocity with a change in the applied stress intensity. Calculated times to failures are markedly decreased in the aged as compared to the as-fired PSZ ceramic.  相似文献

Thick film adherence fracture energy: influence of alumina substrates   总被引:1,自引:0,他引:1  
The adherence of a glass bonded Pt-Au thick film conductor to various alumina substrates is degraded by changes in the surface composition of and by the presence of (0001) crystallographic texture in the substrate. Using the critical fracture energy (γIC) required to separate the thick film from the substrate, it was found that γIC was reduced from a maximum of 3.7 J m−2 using an as-received 96+ wt% alumina substrate to ∼2J m−2 using an as-received 99+ wt% alumina. In addition, the thick film adherence γIC using (0001) sapphire substrates was less than that using (11ˉ23) sapphire. The 96 wt% substate exhibited essentially a random crystallographic surface texture and a considerable amount of surface silicates. The as-received 99+ wt% Al2O3 substrate was charactarized by a high (0001) surface texture and, while exhibiting a similar composition silicate layer as that of the as-received 96 wt% alumina, the total amount of the glass layer is greatly diminished. Fractographic analysis of the separated thick films and substrates showed that changes in the substrate crystallographic texture and the glass layer diminish the interpenetrating nature of the glass—metal interface and weaken the glass—substrate interface. Such changes in thick film microstructure lead to poorer thick film adherence.  相似文献

Osmotic changes are common challenges for marine microorganisms. Bacteria have developed numerous ways of dealing with this stress, including reprogramming of global cellular processes. However, specific molecular adaptation mechanisms to osmotic stress have mainly been investigated in terrestrial model bacteria. In this work, we aimed to elucidate the basis of adjustment to prolonged salinity challenges at the proteome level in marine bacteria. The objects of our studies were three representatives of bacteria inhabiting various marine environments, Shewanella baltica, Vibrio harveyi and Aliivibrio fischeri. The proteomic studies were performed with bacteria cultivated in increased and decreased salinity, followed by proteolytic digestion of samples which were then subjected to liquid chromatography with tandem mass spectrometry analysis. We show that bacteria adjust at all levels of their biological processes, from DNA topology through gene expression regulation and proteasome assembly, to transport and cellular metabolism. The finding that many similar adaptation strategies were observed for both low- and high-salinity conditions is particularly striking. The results show that adaptation to salinity challenge involves the accumulation of DNA-binding proteins and increased polyamine uptake. We hypothesize that their function is to coat and protect the nucleoid to counteract adverse changes in DNA topology due to ionic shifts.  相似文献

Very high concentrations of polybrominated diphenyl ethers (PBDE) have been reported in fish from Lake Mj?sa in Norway. This study was performed to examine the serum concentrations of PBDE and hexabromocyclododecane (HBCD) in consumers of fish from this lake and to investigate possible relationships between serum concentrations, self-reported fish intake and calculated total dietary PBDE exposure. Serum concentrations of the sum of the seven PBDE (BDE-28, 47, 99, 100, 153, 154 and 183) were significantly higher than those of a reference group of Norwegians eating only food with background levels of contamination (medians: 18 ng/g lipids men, 8.4 ng/g lipids women). The median dietary intake of Sum 7 PBDE was 2549 ng/day (30 ng/kg body weight/day), the highest dietary intake of PBDE reported. The contribution from fish caught from the contaminated lake comprised 98.7% of the total dietary exposure. For men, serum levels of PBDE were strongly correlated with the calculated dietary exposure, except for BDE-209. This suggests that sources other than the diet are important for human BDE-209 exposure. The median serum HBCD concentration was 4.1 and 2.6 ng/g lipids for men and women, respectively, and was also found to be associated with consumption of fish from Lake Mj?sa.  相似文献

The creep resistance at temperatures between 1200° and 1300°C in air of alumina—SiC-whisker composites was investigated via four-point flexure to examine (1) the effect of whisker content and (2) the influence of densification additives (i.e., Y2O3 (plus MgO)). The creep resistance of polycrystalline alumina is greatly improved with the addition of ≤ 20 vol% SiC whiskers. The interlocking/pinning of grains by whiskers which limits grain-boundary sliding contributes to the improvement in creep resistance. However, the creep rates of alumina composites in air increase at whisker contents ≥ 30 vol%. Electron microscopy observations suggested that the degradation in creep resistance for whisker content ≥ 30 vol% originated from (1) the promotion of creep cavitation and subsequent microcrack generation from the higher number density of nucleation sites and (2) more extensive formation of grain-boundary amorphous phase(s) associated with an observed increased oxidation rate. Along this one, the excellent creep resistance of alumina composites containing 20 vol% SiC whiskers was significantly degraded by the presence of the intergranular amorphous phases introduced by the addition of the Y2O3 densification additive.  相似文献
