Acute nicotine administration stimulated [3H]norepinephrine ([3H]NE) release from cultured fetal locus coeruleus (LC) cells. The effect was concentration dependent, with an EC50 of 0.9 microM, and was abolished by removal of calcium from, or addition of tetrodotoxin (500 nM) to, the assay buffer. Other nicotinic receptor agonists stimulated [3H]NE release, with the rank order of potency being (+)-epibatidine > (-)-nicotine > 1,1-dimethyl-4-phenylpiperazinium (DMPP). Whereas (-)-nicotine and (+/-)-epibatidine exhibited equal maximal responses, DMPP was a partial agonist and (-)-cytisine had no agonist activity. Nicotine-stimulated release of [3H]NE was blocked by nicotinic receptor antagonists, with an order of potency of mecamylamine > lobeline > cytisine > methyllycaconitine > dihydro-beta-erythroidine. The pharmacological profile of this nicotinic receptor is largely consistent with that described previously for an alpha4beta2 subunit combination, although discrepancies in the efficacies of agonists were observed. No additivity in NMDA- and nicotine-stimulated [3H]NE release was observed, suggesting a common signal transduction mechanism. However, the pharmacological characteristics of MK-801 blockade of nicotine-induced responses were not consistent with those of an NMDA receptor. We therefore conclude that nicotine directly releases [3H]NE from LC cells and does not act indirectly via activation of glutamate release.  相似文献

Summary The effects of cooking and sterilization at several temperatures on the free amino acids (FAA) content in albacore (Thunnus alalunga) muscle were studied during the processing of canned tuna. FAAs were derivatized witho-phtalaldehyde, separated on a C18 column by HPLC and detected by both fluorescence and ultra-violet detectors. After cooking the loss of FAAs was not significant. However, in the final product sterilized at 115 °C and 110 °C (throughout the whole process) there were significant losses with regard to the start material, but not at 118 °C (all temperatures leading to the same lethal F-value). The influence of the thermal process time at 115 °C was evaluated for 60 and 100 min. Significant losses were found between both canned products (25%) and between the raw fish and the final product (12% and 34%, process time 60 and 100 min, respectively). The determination of the content of FAA present in canned albacore may be a useful indication of the severity of the thermal processing.

Zusammenfassung Die Wirkung des Kochens und Sterilisierens bei verschiedenen Temperaturen auf den Gehalt an freien Aminosäuren (FAA) im Thunfischmuskel (Thunnus alalunga) wurde während der Herstellung von Thunfischkonserven untersucht. Die freien Aminosäuren wurden mito-Phtalaldehyd derivatisiert, auf einer C-18-Kolonne mit HPLC abgetrennt und durch Fluorescenz und UV-Detektoren nachgewiesen. Nach dem Kochen war der Verlust an FAA nicht signifikant, jedoch in dem bei 110° und 115 °C sterilisierten Endprodukt ergaben sich signifikante Verluste bezogen auf das Ausgangsmaterial, aber nicht auf das bei 118 °C erhitzte; alle Erhitzungstemperaturen führen zu demselben letalen F-Wert. Der Zeiteinfluß der Erhitzung bei 115 °C wurde bei 60 und 100 min bewertet. Signifikante Verluste sind bei beiden Dosenkonserven aufgetreten (25%) und zwischen diesen und dem Rohfisch ( 12% und 34%) bei einer Erhitzungszeit von 60 und 100 min). Die Bestimmung des FAA-Gehalts in Thunfischkonserven kann für den Nachweis der Einwandfreiheit des thermischen Prozesses sehr nützlich sein.

Proximal tubular (PT) acidosis, which alkalinizes the urinary filtrate, together with Ca2+ supersaturation in PT can induce luminal calcium phosphate (CaP) crystal formation. While such CaP crystals are known to act as a nidus for CaP/calcium oxalate (CaOx) mixed stone formation, the regulation of PT luminal Ca2+ concentration ([Ca2+]) under elevated pH and/or high [Ca2+] conditions are unknown. Since we found that transient receptor potential canonical 3 (TRPC3) knockout (KO; -/-) mice could produce mild hypercalciuria with CaP urine crystals, we alkalinized the tubular pH in TRPC3-/- mice by oral acetazolamide (0.08%) to develop mixed urinary crystals akin to clinical signs of calcium nephrolithiasis (CaNL). Our ratiometric (λ340/380) intracellular [Ca2+] measurements reveal that such alkalization not only upsurges Ca2+ influx into PT cells, but the mode of Ca2+ entry switches from receptor-operated to store-operated pathway. Electrophysiological experiments show enhanced bicarbonate related current activity in treated PT cells which may determine the stone-forming phenotypes (CaP or CaP/CaOx). Moreover, such alkalization promotes reactive oxygen species generation, and upregulation of calcification, inflammation, fibrosis, and apoptosis in PT cells, which were exacerbated in absence of TRPC3. Altogether, the pH-induced alteration of the Ca2+ signaling signature in PT cells from TRPC3 ablated mice exacerbated the pathophysiology of mixed urinary stone formation, which may aid in uncovering the downstream mechanism of CaNL.  相似文献

Because of their large specific surface area, aqueous‐based pseudolatex systems of ethylcellulose can absorb large amounts of drugs. In addition, the stability of polymeric particles in biological fluids delays the release of the drug as in controlled drug delivery systems. The aim of the present study was to characterize the rheological properties of latex particles as a measure of their colloidal stability. Here, we report the effect of three variables: pH, electrolyte concentration, and temperature. The rheograms clearly show that the polymer suspensions displayed Bingham plastic behavior. Internal structuring of the latex was greatest at acid and natural pH values, particularly at the highest ionic strength. In acid solutions, only temperature appeared to play a fundamental role; both the shear stress corresponding to the onset of nonlinear viscoelasticity and the elastic modulus at all frequencies were higher at 37°C than those at room temperature. This is assumed to be a consequence of deformation of the polymer particles upon heating. The effect of ionic strength was noticeable only at the natural pH (pH ? 6.5). At high concentrations of sodium chloride, the particles aggregated because of the decrease in double layer repulsion, and as a result, the latex became structured and its elastic modulus subsequently increased. Interestingly, when the temperature was increased further, this structure presumably broke, down, at least partially, and the storage modulus was reduced. © 2006 Wiley Periodicals, Inc. In this paper we propose a new class of incomplete Riemann solvers, based on approximations in the \(L^\infty \) -norm to the absolute value function in \([-1,1]\) by means of rational functions, for the numerical approximation of the solution of hyperbolic systems of conservation laws. The main idea relies on the construction of a numerical approximation to the viscosity matrix by using an appropriate rational real function \(R(x)\) , that approximates the function \(|x|\) uniformly in \([-1,1]\) , evaluated at the Jacobian of the fluxes of the hyperbolic system computed at some average value (for example, Roe averages). In addition to the Jacobians of the fluxes we shall use either the maximum in absolute value of the characteristic speeds in each cell or an upper bound of them. Thus, the resulting approximate Riemann solver is incomplete in the sense that we do not use the complete spectral decomposition of the Jacobian. Moreover, the new class of Riemann solvers consists of a hierarchy of schemes running from the more dissipative to the less dissipative ones, and having as limiting case a Roe-like scheme. According to the order of the approximation of the generating rational function used, the degree of dissipation can be dosed for particular applications. We study different rational approximations: Newman-type functions, iterative generated Halley functions, and also Chebyshev polynomial approximants. We test our basic algorithms for different initial value Riemann problems for ideal gas dynamics (HD) and magnetohydrodynamics (MHD) to observe their behavior with respect to challenging scenarios in numerical simulations, including some standard numerical pathologies (e. g., heat conduction, postshock oscillations and overheating) and the formation of compound waves in ideal MHD. We also examine our proposed schemes, by computing the numerical approximation of different initial value problems for nonconservative multilayer shallow water equations, where it has been observed that intermediate waves can be properly captured for an appropriate degree of approximation of the generating rational function used. Our numerical tests indicate that the proposed schemes are robust, running stable and accurate with a satisfactory time step restriction (CFL constant), and the computational cost is more advantageous with respect to schemes that use a complete spectral decomposition of the Jacobians.  相似文献

Summary The paper deals with the synthesis of the phenylazo-formamidoethyl 4-t-butylazo-4-cyanovalerate, a product which — by its two azo groups having different thermal stabilities — may be used in initiating processes of stepwise radicalic polymerization. The preparation is based on the condensation of the 4-t-butylazo-4-cyanovaleric acid chloride with N-hydroxy ethyl-phenylazoformamide (HEPF) in anhydrous chloroform, in the presence of pyridine. The initiator purification involves its passing over an alumina column, on using methylene chloride as eluent. HEPF is a new intermediate obtained by the reaction of the ethyl phenylazocarboxylate with ethanolamine. Both the HEPF structure and that of the bis-azo initiator is confirmed by elemental analysis and spectroscopic measurements (IR and H-NMR spectra), as well.  相似文献

A thin film of yttrium Y (150 nm) protected by a 6 nm coating of nickel Ni on a glass substrate was completely hydrogenated in a 1 M NaOH electrolyte at a constant negative current until the transparent Y tri-hydride phase was achieved and hydrogen gas evolution from the electrode began. A series of impedance measurements were performed in situ during the electrochemical experiment to study the properties of the system as dependent on hydrogenation degree and time of relaxation. The equivalent electrical circuit (EEC) simulations were performed with a Randles-like scheme R0[R1CPE1], where R0 is the thin film electrode resistance, R1 the charge transfer resistance and CPE1 is the capacitive constant phase element. The behavior of all the components of the EEC undergoes a clear transition when the hydrogenation degree of the electrode is approximated to its maximum value (H to Y ratio 2.7) and electrochemical process changes from hydrogen uptake to hydrogen evolution.  相似文献

A capillary tube model was solved to investigate the influence of mass transfer coefficient, temperature, and front depth on the evaporation rate during the penetrating-front period of water-filled porous media dried in hot air. The results show that increasing the flow rate of the drying air is not so efficient as increasing the sample temperature. Due to attenuating diffusion rate, the rate of liquid front migration decreases with time. The calculations explain the falling rate period behavior of sandstone heated at 121°C. Trends depicted by the model may be useful for the design of heating conditions for drying processes.  相似文献
