Sirtuins (SIRTs), enzymes from the family of NAD+-dependent histone deacetylases, play an important role in the functioning of the body at the cellular level and participate in many biochemical processes. The multi-directionality of SIRTs encourages scientists to undertake research aimed at understanding the mechanisms of their action and the influence that SIRTs have on the organism. At the same time, new substances are constantly being sought that can modulate the action of SIRTs. Extensive research on the expression of SIRTs in various pathological conditions suggests that regulation of their activity may have positive results in supporting the treatment of certain metabolic, neurodegenerative or cancer diseases or this connected with oxidative stress. Due to such a wide spectrum of activity, SIRTs may also be a prognostic markers of selected pathological conditions and prove helpful in assessing their progression, especially by modulating their activity. The article presents and discusses the activating or inhibiting impact of individual SIRTs modulators. The review also gathered selected currently available information on the expression of SIRTs in individual disease cases as well as the biological role that SIRTs play in the human organism, also in connection with oxidative stress condition, taking into account the progress of knowledge about SIRTs over the years, with particular reference to the latest research results. 相似文献

Transactions have been around since the Seventies to provide reliable information processing in automated information systems.
Originally developed for simple ‘debit-credit’ style database operations in centralized systems, they have moved into much
more complex application domains including aspects like distribution, process-orientation and loose coupling. The amount of
published research work on transactions is huge and a number of overview papers and books already exist. A concise historic
analysis providing an overview of the various phases of development of transaction models and mechanisms in the context of
growing complexity of application domains is still missing, however. To fill this gap, this paper presents a historic overview
of transaction models organized in several ‘transaction management eras’, thereby investigating numerous transaction models
ranging from the classical flat transactions, via advanced and workflow transactions to the Web Services and Grid transaction
models. The key concepts and techniques with respect to transaction management are investigated. Placing well-known research
efforts in historical perspective reveals specific trends and developments in the area of transaction management. As such,

Two ruthenium(III) complexes, namely trans-indazolium[tetrachlorobis(indazole)- ruthenate(III)], HInd[RuInd(2)Cl(4)] and trans-imidazolium[tetrachlorobis(imidazole)- ruthenate(III)], HIm[RuIm(2)Cl(4)] exhibit high anticancer activity in an autochthonous colorectal carcinoma model in rats. Recently, it has been shown that both complexes bind specifically to human serum apotransferrin and the resulting adducts have been studied through spectroscopic and chromatographic techniques with the ultimate goal of preparing adducts with good selectivity for cancer cells due to the fact that tumour cells express high amounts of transferrin receptors on their cell surface.In order to investigate whether the cellular uptake of the complexes was mediated by apotransferrin or transferrin, we compared the antiproliferative efficacy of HInd[RuInd(2)Cl(4)] and HIm[RuIm(2)Cl(4)] with its apotransferrin- and transferrin-bound form in the human colon cancer cell line SW707 using the microculture tetrazolium test (MTT).Our results show that especially the transferrin-bound forms exhibit high antiproliferative activity, which exceeds that of the free complex, indicating that this protein can act as a carrier of the ruthenium complexes into the tumor cell. 相似文献

In precision machining, due to the recent developments in cutting tools, machine tool structural rigidity and improved CNC controllers, hard turning is an emerging process as an alternative to some of the grinding processes by providing reductions in costs and cycle-times. In industrial environments, hard turning is established for geometry features of parts with low to medium requirements on part quality. Better understanding of cutting forces, stresses and temperature fields, temperature gradients created during the machining are very critical for achieving highest quality products and high productivity in feasible cycle times. To enlarge the capability profile of the hard turning process, this paper introduces prediction models of mechanical and thermal loads during turning of 51CrV4 with hardness of 68 HRC by a CBN tool. The shear flow stress, shear and friction angles are determined from the orthogonal cutting tests. Cutting force coefficients are determined from orthogonal to oblique transformations. Cutting forces, temperature field for the chip and tool are predicted and compared with experimental measurements. Capillary electrophoresis (CE) was coupled to ICPMS in order to combine the good performance of this separation technique with the high sensitivity of the ICPMS for the analysis of plutonium and neptunium oxidation states. The combination of a fused-silica capillary with a MicroMist AR 30-I-FM02 nebulizer and a Cinnabar small-volume cyclonic spray chamber yielded the best separation results. With this setup, it was possible to separate a model element mixture containing neptunium (NpO2(+)), uranium (UO2(2+)), lanthanum (La3+), and thorium (Th4+) in 1 M acetic acid. The same conditions were also suitable for the separation of various oxidation states of plutonium and neptunium in different aqueous samples. All separations were obtained within less than 15 min. A detection limit of 50 ppb identical with 2 x 10(-7) M (3-fold standard deviation of a blank) was achieved. To prove the negligible disturbance of the plutonium and neptunium redox equilibria during the CE separations, plutonium and neptunium speciation by CE-ICPMS in acidic solutions was compared with the results of UV/visible absorption spectroscopy and was found to be in good agreement. The impact of seminal plasma components on the fertilization outcomes in humans is still under question. The increasing number of couples facing problems with conception raises the need for predictive biomarkers. Detailed understanding of the molecular mechanisms accompanying fertilization remains another challenge. Carbohydrate–protein recognition may be of key importance in this complex field. In this study, we analyzed the unique glycosylation pattern of seminal plasma proteins, the display of high-mannose and hybrid-type oligosaccharides, by means of their reactivity with mannose-specific Galanthus nivalis lectin. Normozoospermic infertile subjects presented decreased amounts of lectin-reactive glycoepitopes compared to fertile donors and infertile patients with abnormal semen parameters. Glycoproteins containing unveiled mannose were isolated in affinity chromatography, and 17 glycoproteins were identified in liquid chromatography-tandem mass spectrometry with electrospray ionization. The N-glycome of the isolated glycoproteins was examined in matrix-assisted laser desorption ionization mass spectrometry. Eleven out of 27 identified oligosaccharides expressed terminal mannose residues, responsible for lectin binding. We suggest that lowered content of high-mannose and hybrid type glycans in normozoospermic infertile patients may be associated with impaired sperm protection from preterm capacitation and should be considered in the search for new infertility markers. 相似文献
