Mincle agonists have been shown to induce inflammatory cytokine production, such as tumor necrosis factor-alpha (TNF) and promote the development of a Th1/Th17 immune response that might be crucial to development of effective vaccination against pathogens such as Mycobacterium tuberculosis. As an expansion of our previous work, a library of 6,6′-amide and sulfonamide α,α-d -trehalose compounds with various substituents on the aromatic ring was synthesized efficiently in good to excellent yields. These compounds were evaluated for their ability to activate the human C-type lectin receptor Mincle by the induction of cytokines from human peripheral blood mononuclear cells. A preliminary structure–activity relationship (SAR) of these novel trehalose diamides and sulfonamides revealed that aryl amide-linked trehalose compounds demonstrated improved activity and relatively high potency cytokine production compared to the Mincle ligand trehalose dibehenate adjuvant (TDB) and the natural ligand trehalose dimycolate (TDM) inducing dose-dependent and human-Mincle-specific stimulation in a HEK reporter cell line.  相似文献

As part of a characterization research on hydroxyapatite (HAp) plasma deposits for hip prosthesis, crystallinity and residual stresses weredetemiined at the interface Hap'TA6V. A traditional method employed by several laboratories is based on X-ray diffraction by a powder mixture of the investigated HAp sample with a standard of crystallinity Al2O3 Four new methods were investigated for determining the crystallinity of HAp sample, two of which are based on x-ray diffraction, one on neutron diffraction and one on the infrared absorption spectrometry. Whereas the three techniques require the use of powders, the neutron technique can be applied also to the intact HAp deposits. All four methods provide reasonable results, not very different one from each other, with a greater accuracy than for the x-ray and IR methods.  相似文献

The sodium chloride surface is one of the most common platforms for the study of catalysts, thin film growth, and atmospheric aerosols. Here we report a nanoscale periodic modulation pattern on the surface of a cleaved NaCl single crystal, revealed by non-contact atomic force microscopy with a tuning fork sensor. The surface pattern shows two orthogonal domains, extending over the entire cleavage surface. The spatial modulations exhibit a characteristic period of 5.4 nm, along <110> crystallographic directions of the NaCl. The modulations are robust in vacuum, not affected by the tip-induced electric field or gentle annealing (<300?°C); however, they are eliminated after exposure to water and an atomically flat surface can be recovered by subsequent thermal annealing after water exposure. A strong electrostatic charging is revealed on the cleavage surface which may facilitate the formation of the observed metastable surface reconstruction.  相似文献

An important development in recent synthesis strategies is the formation of electronically coupled one and two-dimensional organic systems for potential applications in nanoscale molecule-based devices. Here, we assemble one-dimensional spin chains by covalently linking basic molecular building blocks on a Au(111) surface. Their structural properties are studied by scanning tunneling microscopy and the Kondo effect of the basic molecular blocks inside the chains is probed by scanning tunneling spectroscopy. Tunneling spectroscopic images reveal the existence of separate Kondo regions within the chains while density functional theory calculations unveil antiferromagnetic coupling between the spin centers.  相似文献
