Closures for the drag and virtual mass forces acting on a single air bubble rising in initially quiescent pure water have been numerically investigated using direct numerical simulation techniques. A 3D Front Tracking model was used and the results were compared with simulation results obtained with a 2D Volume of Fluid model to assess the influence of the third dimension. In the simulations realistic values were taken for the physical properties, i.e., a density ratio of 800. The computed time-averaged terminal rise velocity and mean aspect ratio for individual air bubbles ranging in equivalent diameter from 1 to 10 mm rising in pure water compare well with available experimental data.  相似文献

One of the impacts of disease is its effect on milk production. In the present study the effect of an outbreak of bovine herpesvirus type 1 on milk production at the herd level of certified bovine herpesvirus type 1-free dairy farms was modeled. The objective was to study several linear models to quantify the effects of a bovine herpesvirus type 1 outbreak on milk production accounting for the repeated measurements and incorporating our assumptions about the most likely duration of effects of this virus. Because milk production is measured at regular intervals, the data consisted of repeated measurements at the herd and cow levels. A marginal model, a subject-specific random-effect model, and a transition model were developed. The effect of a bovine herpesvirus type 1 outbreak was statistically significant in the random-effect model, and this model fitted the investigated farms best. However, a transition model might be a better model for generalizing the results to the whole population of Dutch dairy farms. The effect of a bovine herpesvirus type 1 outbreak on milk production derived from the random-effect model amounted to, on average, a loss of 0.92 kg of milk per cow per day during a period of 9 wk. The milk production loss varied from almost none to 2 kg of milk per cow per day. This reduction resulted in an average loss of Dfl 372 (Dfl1 = $US $0.50) with lower and upper confidence limits of, respectively, Dfl 12 and Dfl 730 per bovine herpesvirus type 1 outbreak.  相似文献

Expert opinions were elicited about the characteristics at the commercial-farm level of on-line information technology (IT) applications that are able to detect oestrus and mastitis in dairy cows. Since actual data of these characteristics are not available, judgmental data provided an alternative means to interpret the implications of research results for commercial farms. Applications included were activity measurement, milk-production measurement, electrical conductivity of quarter milk, automated concentrate feeders and milk-temperature measurement. Sensitivity and specificity of detection of oestrus (OD), clinical-mastitis (CMD) and subclinical-mastitis (SCMD) were ascertained. Conjoint-analysis was used to assess the effect of each application indirectly by decomposing the evaluated overall detection characteristics of a predefined number of IT combinations. The individual experts were consistent in evaluating the alternatives, but there was variation in estimates among experts. Estimations of the main effects of the applications and important first-order interactions were incorporated into the detection models. Implementation of all applications under study resulted in overall sensitivities and specificities of 82% and 90%, 73% and 87%, 58% and 82% for OD, CMD and SCMD, respectively. Further research is necessary that should take into account costs and benefits of the different detection systems based on the current status of farm performance (e.g. OD and mastitis incidence) and farm structure (e.g. farm size, years in operation of the milking parlour and parlour layout). A study was carried out to determine the possibility of a more-closed farming system for (Dutch) dairy farms. The objective of the study was to provide effective and economically profitable management advice for improving the animal-health status of farms. Management measures will only be successfully applied if supported by farmers and their advisors (such as veterinarians). Therefore, the perception of farmers and advisors of the importance of various risk factors for the introduction of diseases to a farm was determined by using bovine herpes virus type 1 (BHV1) as an example. As part of the study, an evening-long workshop was organized and run thrice. In total, 49 farmers, veterinarians and AI technicians participated in these workshops. The computerized questionnaire technique was based on adaptive conjoint analysis (ACA). ACA has the advantage that participants can work with a large number of risk factors in a relatively short period of time. Another advantage of ACA (compared with standard questionnaires) is that the answers from each participant can be checked with regard to consistency with respect to the importance assigned to them. Data from participants with inconsistent responses can be excluded from further analyses. The results of the ACA interview were compared with the risk factors reported in the literature as being associated with BHV1 status (e.g. purchase of cattle, participation in cattle shows) and with farmers' actual management to prevent the introduction of diseases. The workshop participants were all operating in the dairy sector and they seemed well aware of the risk of direct animal contacts for the introduction of BHV1. Farmers thought visitors to be more risky than did AI technicians and (especially) veterinarians. Farmers who purchased cattle or participated in cattle shows were of the opinion that the risks of direct animal contacts were more important than did farmers who were not involved in those practices. Farmers whose farms were BHV1-positive (and participated in cattle shows more often) thought the risk of participation smaller than did farmers with BHV1-negative farms.  相似文献

This research was carried out to analyse the visits specialists of the Dutch Animal Health Service made to growing and fattening pig farms. The type and frequency of the visits and identified herd-health management factors that did not meet accepted standards were investigated. In total 373 visit reports were studied. The majority of the visits (n = 306 of 373) were made to investigate the cause of health, welfare, and performance problems ('problem-solving visits'). Respiratory disorders were the main reason for requesting a specialist to assess farm conditions and management (n = 156). In the other 67 of 373 visit reports the specialists screened for herd-health management factors that did not meet standards for the prevention of disease ('screening visits'). For both types of visits, the main factors detected were abrupt changes in feeding regimens (e.g. changes in feed type, feed composition or feed supplier) (37%), inadequate measures to prevent introduction of pathogens by people and trucks (83%), and incorrect adjustment of the ventilation system (58-60%). The specialists focusing on housing-climate management, identified the majority of factors in an equal number irrespective of whether the visit was a problem-solving visit or a screening visit. This implies that even on farms that appear not to have health or performance problems, factors that relate to disease are present and may cause problems sooner or later. Although veterinary practitioners and other farm advisors assist farmers in their management to optimize herd health, the findings of the research suggest that advisors could provide additional support in situations where environmental and managerial factors play a role in pig health and performance. The knowledge of advisors about integrated herd-health management can be broadened by means of textbooks, courses, or computer programs.  相似文献

Prenatal smoke exposure (PreSE) is a risk factor for nicotine dependence, which is further enhanced by postnatal smoke exposure (PostSE). One susceptibility gene to nicotine dependence is Cytochrome P450 (CYP) 2A6, an enzyme responsible for the conversion of nicotine to cotinine in the liver. Higher CYP2A6 activity is associated with nicotine dependence and could be regulated through DNA methylation. In this study we investigated whether PostSE further impaired PreSE-induced effects on nicotine metabolism, along with Cyp2a5, orthologue of CYP2A6, mRNA expression and DNA methylation. Using a mouse model where prenatally smoke-exposed adult offspring were exposed to cigarette smoke for 3 months, enzyme activity, mRNA levels, and promoter methylation of hepatic Cyp2a5 were evaluated. We found that in male offspring, PostSE increased PreSE-induced cotinine levels and Cyp2a5 mRNA expression. In addition, both PostSE and PreSE changed Cyp2a5 DNA methylation in male groups. PreSE however decreased cotinine levels whereas it had no effect on Cyp2a5 mRNA expression or methylation. These adverse outcomes of PreSE and PostSE were most prominent in males. When considered in the context of the human health aspects, the combined effect of prenatal and adolescent smoke exposure could lead to an accelerated risk for nicotine dependence later in life.  相似文献

The concentration of endothelin-1 (ET-1) in the brain regions, heart, and throacic aorta of 1-, 4-, 6- and 8-week-old spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats was determined using radioimmunoassay. ET-1-like immunoreactivity in the brain regions of 1-week-old WKY and SHR rats was lower compared to older (6 and 8 weeks) rats. ET-1 levels in the central nervous system gradually increased with age in both SHR and WKY rats. However, the concentration of ET-1 in 8-week-old rats was lower in the brain regions of SHR compared to WKY rats. The concentration of ET-1 in the thoracic aorta of SHR (224 +/- 43 pg/g tissue) rats was lower than that of WKY (452 +/- 11 pg/g tissue) rats at 1 week of age. However, ET-1 levels gradually increased with age in SHR rats. By 8 weeks of age, levels of ET-1 in SHR (623 +/- 33 pg/g tissue) rats were higher compared to WKY (439 +/- 62 pg/g tissue) rats. In the heart, ET-1 levels were similar in WKY and SHR rats at 4 weeks of age, but at 8 weeks of age ET-1 levels were higher in SHR rats (364 +/- 33 pg/g tissue) compared to WKY rats (260 +/- 31 pg/g tissue). It appears that at 8 weeks of age when hypertension is fully expressed in rats, ET-1 levels are lower in the central nervous system and are higher in the thoracic aorta and heart of SHR compared to WKY rats.  相似文献

Xanthobacter flavus, a gram-negative facultatively autotrophic bacterium, employs the Calvin cycle for the fixation of carbon dioxide. Cells grown under autotrophic growth conditions possess an Fe(2+)-dependent fructosebisphosphate (FBP) aldolase (class II) in addition to a class I FBP aldolase. By nucleotide sequencing and heterologous expression in Escherichia coli, genes encoding transketolase (EC 2.2.1.1.; CbbT) and class II FBP aldolase (EC 4.1.2.13; CbbA) were identified. A partial open reading frame encoding a protein similar to pentose-5-phosphate 3-epimerase was identified downstream from cbbA. A phylogenetic tree of transketolase proteins displays a conventional branching order. However, the class II FBP aldolase protein from X. flavus is only distantly related to that of E. coli. The autotrophic FBP aldolase proteins from X. flavus, Alcaligenes eutrophus, and Rhodobacter sphaeroides form a tight cluster, with the proteins from gram-positive bacteria as the closest relatives.  相似文献
