Patients with acute promyelocytic leukemia (APL) usually relapse after all-trans retinoic acid (RA) treatment because this therapy fails to eradicate the malignant clone. Our data showed that KH 1060 and other 20-epi vitamin D3 analogs alone were potent inhibitors of clonal growth of NB4 cells, an APL cell line (ED50, approximately 5 x 10(-11) M). The combination of KH 1060 and 9-cis-RA synergistically and irreversibly enhanced this effect. Neither KH 1060 nor 9-cis-RA (10(-6) M, 3 d) were strong inducers of differentiation of NB4 cells. However, 98% of the cells underwent differentiation to a mature phenotype with features of both granulocytes and monocytes after exposure to a combination of both compounds. Apoptosis only increased after incubation of NB4 cells with 9-cis-RA alone (28%) or with a combination of 9-cis-RA plus KH1060 (32%). Immunohistochemistry showed that the bcl-2 protein decreased from nearly 100% of the wild-type NB4 cells to 2% after incubation with a combination of KH 1060 and 9-cis-RA, and the bax protein increased from 50% of wild-type NB4 cells to 92% after culture with both analogs (5 x 10(-7) M, 3 d). Western blot analysis paralleled these results. Studies of APL cells from one untreated individual paralleled our results with NB4 cells. Taken together, the data demonstrated that nearly all of the NB4 cells can be irreversibly induced to differentiate terminally when exposed to the combination of KH 1060 and 9-cis-RA. 相似文献

A new forage sorghum entitled Pnina was developed in Israel. Pnina, harvested at early milk (EM) and hard dough (HD) stages of maturity, was compared with commercial sorghum variety FS‐5 and a sorghum BMR/Sudan hybrid Nutriplus. Plants grew during summer and were irrigated with 190 mm water. Pnina was semi‐dwarf (1.35 m height) and absolutely resistant to lodging. The tall varieties FS‐5 and Nutriplus suffered at HD from high lodging. Pnina was more leafy than Nutriplus and FS‐5. The dry matter (DM) content of FS‐5 and Nutriplus were at EM below the level recommended for ensilage and elevated slightly at HD, whereas DM content of Pnina was higher. Crop yields of all varieties at HD were similar and ranged between 13.3 and 14.7 t ha?1 DM. Dry matter losses during ensilage and silage pH were similar in the three varieties at HD. In vitro DM digestibility of HD silages ranged between 62.5 and 67.1%. Neutral detergent fiber (NDF) content was higher in Pnina silages. Yields of digestible silage per hectare, were similar in the three varieties harvested at HD (8.4 to 8.8 t ha?1 DM); however, yield of digestible NDF per hectare was higher in Pnina. ABSTRACT Twelve rose cultivars were selected by field tasting as potential sources of edible flowers. Hot water infusions (teas) of air‐dried petals of these cultivars were assayed for antioxidant activity, total phenols, and total anthocyanins contents. Their composition was analyzed by high‐performance liquid chromatography (HPLC). Green tea was tested in parallel as a reference antioxidant‐rich beverage. Rose petal teas from different cultivars exhibited scavenging capacity toward 2,2′‐azino‐bis‐(3‐ethylbenzothiazoline)‐6‐sulfonate cation radical (ABTS+) ranging between 712.7 and 1770.7 μM Trolox equivalents (TE) per gram of dry petals, as compared with 1227.6 μM TE/g dry weight in the green tea. The range of total phenols content in rose teas was 50.7 to 119.5 mg gallic acid equivalents (GAE) per gram of dry matter, as compared with 62.1 mg GAE/g dry weight in the green tea. The rose teas were rich in free gallic acid. The highest values of antioxidant activity, total phenols, and gallic acid contents were found in the cultivars San Francisco, Katharina Zeimet, and Mercedes and in the essential‐oil‐bearing rose Rosa damascena. The correlation coefficients between antioxidant activity, on the 1 hand, and the contents of total phenols and of gallic acid in various rose cultivars, on the other hand, were 0.79 and 0.81, respectively. No clear relationship between anthocyanin level and radical‐scavenging activity was revealed. Teas from different rose cultivars significantly differed in their sensory properties. It was concluded that dried rose petals may be used for preparing antioxidant‐rich caffeine‐free beverages, either separately or in combination with other herbal materials. 相似文献
