Variations in composition, microhardness (in the thermomechanically affected zone) and texture in the tool domain of the dissimilar friction stir weld of AA5083-O and AA6082-T6 alloys were investigated. The contents of the major alloying elements in the weld zones were determined using inductively coupled plasma?atomic emission spectroscopy. It was observed that a slight drop in the content of the alloying elements results from the friction stir welding process with the Mg content being the most affected amongst the major alloying elements in the two alloys. By relating the mass fractions of the major alloying elements in the parent metals of both alloys to those of the stir zone, the relative proportions of the two alloys in the stir zone were estimated with the results showing that at least 60% of the materials in the stir zone are from the retreating side of the weld. It was also revealed that the changes in the hardness profile in the thermomechanically affected zone of the retreating side are predominantly influenced by changes in grain size in that domain. Finally, the investigation further revealed that the texture component in the tool shoulder domain is different from the texture component in the tool pin domain.  相似文献

A phosphatase in papaya was extracted, partially purified, and characterized. With p-nitrophenyl phosphate as substrate, the enzyme had a pH optimum of 6.0, which categorized it as an acid phosphatase, a temperature optimum of 37°C, and a Km of 1.0 mM. Heat inactivation of papaya acid phosphatase was biphasic, and the kinetics of both phases were first order reactions. D values at 60°, 65°, 70°C for the heat resistant phase were 21.0, 11.7, and 4.0 min, respectively. For the heat labile and heat resistant isozymes of papaya acid phosphatase, the activation energies, Ea, for thermal inactivation were 60.0 Kcal/mole and 37.8 Kcal/mole, respectively. The apparent molecular weight of the enzyme as determined by gel filtration was 120,000 daltons.  相似文献

High‐pressure freezing is the preferred method to prepare thick biological specimens for ultrastructural studies. However, the advantages obtained by this method often prove unattainable for samples that are difficult to handle during the freezing and substitution protocols. Delicate and sparse samples are difficult to manipulate and maintain intact throughout the sequence of freezing, infiltration, embedding and final orientation for sectioning and subsequent transmission electron microscopy. An established approach to surmount these difficulties is the use of cellulose microdialysis tubing to transport the sample. With an inner diameter of 200 μm, the tubing protects small and fragile samples within the thickness constraints of high‐pressure freezing, and the tube ends can be sealed to avoid loss of sample. Importantly, the transparency of the tubing allows optical study of the specimen at different steps in the process. Here, we describe the use of a micromanipulator and microinjection apparatus to handle and position delicate specimens within the tubing. We report two biologically significant examples that benefit from this approach, 3D cultures of mammary epithelial cells and cochlear outer hair cells. We illustrate the potential for correlative light and electron microscopy as well as electron tomography.  相似文献

Infection of 10 dairy goats of mixed breed with Staphylococcus aureus resulted in changes in concentration of certain analytes in milk, which became more pronounced as the infection progressed. Somatic cell counts reached a peak increase of 147% of pre‐infection levels, free fatty acids 49%, sodium 14%, Chloride 16% and Copper 47%. Decreases in milk concentration included lactose at a maximum 8%, iron at 47%, potassium at 10% and Calcium at 12% of pre‐infection levels. Protein, nitrogen and magnesium levels appeared unaffected by infection. Following curative treatment, changes in the analytes resolved towards normal levels by the fourth week post‐treatment.  相似文献

透水再生骨料混凝土(PRAC)是近年来新兴的一种环境友好型混凝土,利用建筑废弃物破碎所得再生骨料置换天然骨料,改善传统混凝土在城市路面建设中的弊端,实现废物资源再利用和建筑材料的可持续发展.总结了水灰比、孔隙率、再生骨料粒径和取代率、成型工艺等因素对透水再生骨料混凝土力学性能、透水性以及耐久性的影响,并提出了有待进一步研究的问题.已有研究成果表明再生骨料可以用于制备透水混凝土,应用于停车场、人行道及植生护坡等工程.  相似文献
