Heparin affinity high-performance liquid chromatography as an alternative to bioassay of CS23 mutein of recombinant human basic fibroblast growth factor

We investigated whether a chemical assay by high-performance liquid chromatography (HPLC) as an alternative to the complicated and time-consuming bioassay for CS23 mutein of recombinant human basic fibroblast growth factor (rhbFGF-CS23) using the fetal bovine heart endothelial cell line ATCC CRL 1395. Physically, chemically or enzymatically denatured rhbFGF-CS23 was subjected to heparin affinity (HA)-HPLC and the bioassay. Good agreement was observed between the results obtained by these two methods. Moreover, HA-HPLC gave much more reproducible results (RSD = 1.9%, n = 6) than the bioassay (RSD = 7.4%, n = 18). HA-HPLC is therefore a simple, accurate and reproducible alternative to the bioassay for quality control and stability studies for rhbFGF-CS23 preparations. HA-HPLC is also considered to be applicable to assays for FGFs which have heparin affinity and biological activity similar to those of the CS23 mutein.

As the cationic component of natural polymer polyionic complexes, polysaccharide chitosan or polypeptide polylysine, and as the anionic component, polysaccharide carboxymethyl cellulose (=CMC) or polypeptide poly(glutamic acid) (PGA) were used. The degree of chitosan/CMC or polylysine/PGA complex film swelling was minimum, when the amounts of the cationic and anionic groups in these polymers were balanced at 1 : 1 ratio. On the other hand, the degree of chitosan/PGA film swelling was minimum at the ratio with excess of PGA which is a linear flexible chain. Comparing the enthalpies estimated by DSC analysis, it was suggested that the larger quantity of water in the form of nonfreezable bound water was absorbed in the complex films with polysaccharide which is a chain of ring structure. Further, the compressive moduli of swollen complex films were high, when the complex consisted of polysaccharide components.     

DC‐linked hybrid generation system with an energy storage device including photovoltaic generation and gas engine cogeneration for residential houses

For the past few years, hybrid generation systems including solar panel and gas cogeneration have been used for residential houses. Solar panels can generate electronic power at daytime but not at night. But the power consumption of residential houses usually peaks in the evening. The gas engine cogeneration system can generate electronic power without such a restriction, and it also can generate heat power to warm up a house or to produce hot water. In this paper we propose a solar panel and gas engine cogeneration hybrid system with an energy storage device, combined by a DC bus. If a blackout occurs, the system still can supply electronic power for special house loads. We propose a control scheme for the system related to the charging level of the energy storage device and the voltage of the utility grid, which can be applied to both grid‐connected and standalone operation. Finally, we report experiments designed to demonstrate system operation and calculations for loss estimation. © 2012 Wiley Periodicals, Inc. Fish intake and serum levels of organochlorines among Japanese women

This study evaluates background serum levels of selected organochlorine compounds among Japanese women of reproductive age and investigates whether lifestyle factors, especially dietary factors, may be associated with these levels. A cross-sectional study was performed on 80 Japanese women, aged 26-43 years, who complained of infertility and were confirmed not to have endometriosis. The serum levels of total toxic equivalency (TEQ), 18 polychlorinated dibenzo-p-dioxins (PCDDs)/polychlorinated dibenzofurans (PCDFs), 4 coplanar polychlorinated biphenyls (cPCBs), 36 ortho-substituted polychlorinated biphenyls (PCBs), and 13 chlorinated pesticides or their metabolites were measured and data were collected on the women's age, residence, occupation, body mass index (BMI), smoking and alcohol habit and 6 dietary intakes (fish, meats, rice, vegetables, fruits and dairy products). The serum median level of total TEQ was 25.1 pg TEQ/g lipid, that of PCDDs/PCDFs/cPCBs was 11.5 pmol/g lipid, that of PCBs was 0.46 nmol/g lipid, and that of total pesticides was 1.32 nmol/g lipid. The serum levels of total TEQ, PCDDs/PCDFs/cPCBs, PCBs and pesticides were positively associated with age (P for trend=0.003, 0.01, 0.005 and 0.01, respectively) and frequent fish consumption (P for trend=0.002, 0.003, 0.0003 and 0.006, respectively). Other lifestyle factors were not associated with serum organochlorine levels. The present study suggests that Japanese women who consume fish frequently in their reproductive period tend to accumulate organochlorines in their bodies.     

Effects of winding connections on characteristics of a five‐phase stepping motor

A method for calculating the single‐step response of five‐phase hybrid stepping motors is obtained. The effects of winding connecting methods—(1) star‐bipolar SB, (2) pentagon PD, and (3) new pentagon NP—on the single‐step response are investigated. By application of the above calculation method, the continuous rotating characteristics are studied. It is found that the maximum slewing pulse rate of NP is higher than that of SB and PD connections, and that the variation of the rotor speed at high speed with the SB and PD connections is larger than that with the NP connection. The effects of the current waveform on the oscillation characteristics and relations between the current waveform and the winding connections are investigated. © 2001 Scripta Technica, Electr Eng Jpn, 136(1): 47–56, 2001

Takashi Nakai and Toshinari Tanaka of the Forestry and Forest Products Research Institute in Japan present the results of tests, using the easy handling and reasonable cost technique of fundamental vibration frequency measurement, on larger than usual cross-sections. Modulus of elasticity measurements showed this method to have high possibilities for stress grading, overcoming the size limitations of existing commercial equipment at the saw mill or lumber yard. This paper was first presented at CIB Working Commission W18a in Vancouver in September 1988.     

MACSYM: A hierarchical parallel image processing system for event-driven pattern understanding of documents

MACSYM is a hierarchical parallel processing system for pattern understanding applications. It features event-driven parallel processing for knowledge-based understanding of document images. The system is composed of a master processor, slave processors and a large shared memory, and is equipped with versatile communication facilities. The parallel processing software system M.UM has been developed on MACSYM. It supplies a parallel processing language MacC, an extended version of C, and supports the programming for document image understanding. The Japanese newspaper layout understanding system EXPRESS is being developed on MACSYM. It analyzes a newspaper image and extracts articles in a few seconds.     

Nanoimprint with thin and uniform residual layer for various pattern densities

Pattern density variation is uncomfortable for nanoimprint lithography which uses a moldable material supplied as a thin film, because the variation of pattern density causes variations of residual layer thickness reflecting on the local pattern density. To solve the problem, a new type of mold “capacity-equalized mold”, which has constant averaged depth regardless of pattern density, was fabricated and the structure of the mold was inspected. UV nanoimprint was then carried out using the mold and thickness and uniformity of the residual layer were investigated. An average thickness of 33.2 nm with a standard deviation of 3.4 nm was obtained for the mold pattern layout with a pattern density of from 0.25 to 0.75. It was found that a standard deviation of 1.2 nm was achieved for pattern density variation of from 0.33 to 0.67 by excluding artifacts.     

Single-grain Si thin-film transistors SPICE model,analog and RF circuit applications

Single-grain thin-film transistors (SG-TFTs) fabricated inside location-controlled using μ-Czochralski process exhibit SOI-FETs like performance despite processing temperatures remaining below 350 °C. Thus, the SG-TFT is a potential technology for large-area highly-integrated electronic system and system-in-package, taking advantage of the system-on-flexible substrate and low manufacturing cost capabalities. The SG-TFT is modeled based on the BSIMSOI SPICE model where the mobility parameter is modified to fit the SG-TFT behavior. Therefore, analog and RF circuits can be designed and benchmarked. A two-stage telescopic cascode operational amplifier fabricated in a prototype 1.5 μm SG-TFT technology demonstrates DC gain of 55 dB and unity-gain bandwidth of 6.3 MHz. A prototype CMOS voltage reference demonstrates a power supply rejection ratio (PSRR) of 50 dB. With unity-gain frequency, f_T, in the GHz range, the SG-TFT can also enable RF circuits for wireless applications. A 12 dB gain RF cascode amplifier with integrated on-chip inductors operating in the 433 MHz ISM band is demonstrated.     

Evaluation of hydrogen metabolism by Escherichia coli strains possessing only a single hydrogenase in the genome

The four hydrogenase isozymes; hydrogenase 1 (Hyd-1), hydrogenase 2 (Hyd-2), hydrogenase 3 (Hyd-3) and hydrogenase 4 (Hyd-4) of Escherichia coli have been reported for their crucial functions in the hydrogen metabolism; however, their distinctive roles could not be completely understood. In this study, four ideal hydrogenase operon mutants, Δhyb hyc hyf, Δhya hyc hyf, Δhya hyb hyf, and Δhya hyb hyc, in which only a single hydrogenase is intact in the genome, were constructed as well as one quadruple mutant (Δhya hyb hyc hyf) that all four hydrogenase operons were deleted. First, single operon mutants and single-gene mutants for each hydrogenase showed different hydrogen productivity and growth in the anaerobic fermentation, indicating that bacterial phenotype regarding the hydrogen metabolism via the deletion of each operon is different with that of each single gene. Then, 4 triple hydrogenase operon mutants and one quadruple mutant were investigated to evaluate the hydrogen metabolism (hydrogen production and uptake) using glucose or glycerol as a substrate of hydrogen fermentation. With both the carbon sources, only Hyd-2 and Hyd-3 were able to produce hydrogen. Furthermore, all the hydrogenases showed hydrogen uptake activity. In addition, no hydrogen production and hydrogen uptake were detected in the quadruple mutant which does not have all 4 hydrogenases. Hydrogen production from Hyd-2 and Hyd-3 was further confirmed by complementing their operons in the cloning vector pBR322.
