A set of 11 expression vectors was constructed, each of themharbouring a cloning cassette under the control of the promoter.Some of these vectors enable expression of foreign proteinsin the cytoplasm, while others include a synthetic sequencecoding for a very efficient secretion signal sequence. Otherfeatures are an fl origin of replication (in plus or minus orientation)and a promoterup mutation that enhances the already very highlevel of expression from these vectors. With such a versatilevector family, cloning, sequencing and sitedirected mutagenesiscan be performed on the same vector, and the level of expressioncan be defined according to the specific constraints of a givenprotein.  相似文献

The electrochemical promotion of catalysis (EPOC) of propene combustion was investigated using Pt sputtered thin film on an O2− conductor, 8 mol% Y2O3-stabilized-ZrO2 (YSZ). In order to separate the influence of the thermal migration of the O2− oxide ions from the electrolyte to the catalyst surface and the impact of an electrical polarization on the catalytic activity, several light-off experiments (cool down and heat up procedures) were successively performed under different polarizations, i.e. OCV, +2 and −2 V. These experiments have clearly shown that the presence of O2− (thermally or electrochemically induced) inhibits the catalytic activity of the platinum for the propene deep oxidation. These results demonstrate the importance to define a normalized rate enhancement ratio, ρ n , from a reference value of the catalytic rate corresponding to a Pt surface state free of O2− ions.  相似文献

Symmetric solid-electrolyte galvanic cells such as Pt/Ag Se/RbAg4+yI5–ySey/Ag Se/Pt, based on a continuous variation of the redox potential of the storage phase Ag Se with its composition, show considerable advantages over other types of cells, e.g. high accuracy, long charge-storage capability, effectively a double range of potential variation, and suppression of anodic polarization at the metalelectrolyte interface. The temperature characteristics as well as the overvoltage of this cell have been quantitatively determined and analysed. This cell can discharge in the temperature range –30 to 130° C with a current density up to 10 mA cm–2.  相似文献

Cork is a natural material produced in the Mediterranean countries. Cork stoppers are used to seal wine bottles, Cork stopper quality classification is a practical pattern classification problem. The cork stoppers are grouped into eight classes according to the degree of defects on the cork surface. These defects appear in the form of random-shaped holes, cracks, and others. As a result, the classification cork stopper is not a simple object recognition problem. This is because the pattern features are not specifically defined to a particular shape or size. Thus, a complex classification form is involved, Furthermore, there is a need to build a standard quality control system in order to reduce the classification problems in the cork stopper industry. The solution requires factory automation meeting low time and reduced cost requirements. This paper describes a cork stopper quality classification system using morphological filtering and contour extraction and following (CEF) as the feature extraction method, and a fuzzy-neural network as a classifier. This approach will be used on a daily basis. A new adaptive image thresholding method using iterative and localized scheme is also proposed, A fully functioning prototype of the system has been built and successfully tested. The test results showed a 6.7% rejection ratio, It is compared with the 40% counterpart provided by traditional systems. The human experts in the cork stopper industry rated this proposed classification approach as excellent.  相似文献

Winemakers are interested in wines with low bitterness and astringency, and therefore, there is a preference for a higher presence of skin proanthocyanidins in wines. However, the information regarding the effect of reducing or eliminating seed proanthocyanidins from the must or fermenting wine on the characteristics of finished wines is scarce. For this reason, we elaborated a wine using Monastrell grapes from which the seeds had been removed and compared it with a control wine (CW) made from intact grapes. The anthocyanin and proanthocyanidin concentration and profile, and the chromatic and sensory characteristics were studied. The absence of seed proanthocyanidins did not affect anthocyanin concentration, although the proanthocyanidin concentration was 40% lower than in the CW. The copigmentation (CA) study showed that both wines had a similar extent of CA phenomena although colour intensity was higher when seed proanthocyanidins were present. The panellist evaluated the wines elaborated without seeds as being fruitier, less astringent and with an overall higher quality.  相似文献

Carbohydrates play a pivotal role in intercellular communication processes. In particular, glycan antigens are key for sustaining homeostasis, helping leukocytes to distinguish damaged tissues and invading pathogens from healthy tissues. From a structural perspective, this cross-talk is fairly complex, and multiple membrane proteins guide these recognition processes, including lectins and Toll-like receptors. Since the beginning of this century, lectins have become potential targets for therapeutics for controlling and/or avoiding the progression of pathologies derived from an incorrect immune outcome, including infectious processes, cancer, or autoimmune diseases. Therefore, a detailed knowledge of these receptors is mandatory for the development of specific treatments. In this review, we summarize the current knowledge about four key C-type lectins whose importance has been steadily growing in recent years, focusing in particular on how glycan recognition takes place at the molecular level, but also looking at recent progresses in the quest for therapeutics.  相似文献

The feasibility of incorporating microcapsules containing Phase Change Materials (PCMs), previously obtained by a suspension polymerization process, in gypsum wallboards to increase the wall energy storage capacity was studied. Firstly, the energy storage capacity of the resulting microcapsules and the microencapsulation efficiency was maximized by studying the influence of the synthesis variable core/coating mass ratio on the suspension polymerization process. Results indicate that the higher paraffin wax to styrene monomer mass ratio, the lower microencapsulation efficiency. A mass ratio of Rubitherm® RT27 to styrene monomer equal 1.5 allowed to obtain microcapsules with the highest energy storage capacity and a good microencapsulation efficiency. It was also observed that the energy storage capacity is dependent on the particle size; the maximum capacity was obtained for a particle size of 500 μm. Finally, the thermal behaviour of three gypsum wallboards one without PCMs and the others doped with 4.7% and 7.5% by weight of microcapsules containing Rubitherm® RT27 at the optimal core/coating mass ratio was studied. Results showed that the higher the amount of microcapsules containing PCMs incorporated to the gypsum wallboard, the lower or higher the external wall temperature for heating or cooling process, respectively. Besides, the incorporation of the microcapsules to the wall increased the time required to achieve the final steady state, verifying that the material insulation capacity was enhanced by increasing PCMs content in the wall.  相似文献
