Recursive algorithms to identify both subsystems of a continuous-time Wiener system are presented. The system is driven and disturbed by Gaussian white random signals. The impulse response of the linear dynamic subsystem is recovered with a correlation method. It is shown that the inverse of the non-linear characteristic of the other subsystem is a regression function. Then, to recover the inverse, two estimates are presented. The algorithms converge to the unknown impulse response, and the inverse of the characteristic, respectively. Convergence rates are presented. Moreover, results of simulation examples are given.  相似文献

The synthesis, enzymatic evaluation, and molecular modeling studies of new fluorogenic tetrapeptide‐based substrates selective for caspase 8, having the general structure Ac‐IETD–AXX, are described. Various fluorescent reporter groups (AXX), i.e., 3‐ and 4‐substituted coumarins and quinolin‐2(1H)‐ones were synthesized by von Pechmann condensation. They were subsequently coupled with the caspase‐8‐selective tetrapeptide Ac‐IETD‐OH under newly developed synthetic conditions to give the desired substrates in good yields and in high enantiomeric purity. Based on KM and Vmax values, the new compounds proved to be excellent substrates for recombinant human caspase 8. In contrast, the KM values for the same compounds as substrates for human caspase 3 were approximately 10–20‐fold higher. Molecular modeling studies based on the X‐ray crystal structures of both human caspases 3 and 8 revealed that there is sufficient room within both active sites to accommodate substrates with moderately bulky substituents in the 3‐ and 4‐positions of the fluorogenic coumarins and quinolin‐2(1H)‐ones. Automated docking of the substrates into the active sites of both human caspases 3 and 8 with the program AutoDock 3 gave structures similar to the published crystallographic structures for the same tetrapeptide bound to caspase 8 in the form of an irreversible inhibitor. The calculated binding energies for the new substrates to either caspase 3 or 8 showed little difference between the substrates, consistent with the KM data. In addition, the calculated binding energies (ΔG) to caspase 8 were considerably more negative than those to caspase 3, also consistent with the KM data. A possible molecular interaction that might explain the selectivity of the IETD tetrapeptide motif for caspase 8 over caspase 3 is discussed.  相似文献

Larval fishes were sampled in the Milk River, Missouri River drainage, Montana from May to August 2002, 2003 and 2004 to describe temporal spawning distribution in relation to spring discharge. Total larval catch‐per‐unit‐effort (CPUE) in 2002 (28.9 fish/100 m3) was an estimated 29 times greater than in 2003 (0.99 fish/100 m3) and 16 times greater than in 2004 (1.78 fish/100 m3). In 2003 and 2004 more than one third of the total catch occurred before 12 June whereas in 2002, only 5% of the total catch occurred before 12 June. Marked differences in larval species composition were also observed between years, suggesting that a later peak in discharge may benefit some species and an earlier peak others. In 2002, when flows peaked later (at 77 m3 s?1), common carp Cyprinus carpio represented 37% of the total larval catch. Common carp were proportionally less abundant in 2003 (7.2%) and 2004 (1.4%) than in 2002. In 2004, when flows peaked (at 163 m3 s?1) 32 days earlier than in 2002 but only 15 days earlier than in 2003 (at 73 m3 s?1), shorthead redhorse Moxostoma macrolepidotum and suckers Catostomus sp. were the numerically dominant taxa. These results indicate that the timing, not necessarily the magnitude, of peak spring discharge may influence spawning success in the lower Milk River, as indicated by larval fish catches. Experiments on mechanical consolidation of rapidly solidified (RS) powder of 6061 + 26 mass% Si alloy were performed using the oscillating-die extrusion method. The RS powder was wrapped in thin-wall 6061-alloy cup 35 mm in diameter and vacuum-compressed by means of 100 ton press. Bars 8 mm in diameter were extruded with cross-section reduction of λ = 19 without any preheating of the charge. Tubes with a diameter/wall thickness of 14 mm/1 mm and cross-section reduction of λ = 33 were also manufactured with success. TEM/STEM observations revealed a very fine structure of as-extruded material and bimodal distribution of quasi-spherical silicon particles. Statistical analysis revealed a silicon fine fraction of 0.1–0.7 μm and a coarse fraction 2.1–2.5 μm in diameter. Examination by means of TEM did not reveal any significant changes in the morphology of the silicon particles, even when a high extrusion ratio and the material annealing after deformation were used. Hot compression tests on as-extruded rods (λ = 19) and preliminary annealed samples were performed at a constant true strain rate of 5 × 10?3 s?1 within the temperature range of 293–823 K. High strength of the material and relatively high ductility of samples deformed by compression up to ?t ? 0.4 were observed. The maximum flow stress value for as-extruded material was reduced with deformation temperature from ~390 to ~3.5 MPa for 293 and 823 K, respectively. Annealing of the samples at 773 K/30 min was found to reduce the maximum flow stress by 30–40%. Tensile strengths of similar as-cast alloys and materials manufactured by means of other powder metallurgy methods were shown for the purpose of comparison.  相似文献

The deep-sequencing of small RNAs has revealed that different numbers and proportions of miRNA variants called isomiRs are formed from single miRNA genes and that this effect is attributable mainly to imprecise cleavage by Drosha and Dicer. Factors that influence the degree of cleavage precision of Drosha and Dicer are under investigation, and their identification may improve our understanding of the mechanisms by which cells modulate the regulatory potential of miRNAs. In this study, we focused on the sequences and structural determinants of Drosha and Dicer cleavage sites, which may explain the generation of homogeneous miRNAs (in which a single isomiR strongly predominates) as well as the generation of heterogeneous miRNAs. Using deep-sequencing data for small RNAs, we demonstrate that the generation of homogeneous miRNAs requires more sequence constraints at the cleavage sites than the formation of heterogeneous miRNAs. Additionally, our results indicate that specific Drosha cleavage sites have more sequence determinants in miRNA precursors than specific cleavage sites for Dicer and that secondary structural motifs in the miRNA precursors influence the precision of Dicer cleavage. Together, we present the sequence and structural features of Drosha and Dicer cleavage sites that influence the heterogeneity of the released miRNAs.  相似文献

Aqueous solutions of ethoxylated alcohols which form lyotropic liquid crystals at high concentrations (40–80%) were selected as model lubricating substances. Microscopic studies under polarized light and viscosity measurements were carried out in order to confirm the presence of liquid crystalline structures in the case of alcohol solutions with ethoxylation degrees of 3, 5, 7 and 10. Microscopic images and viscosity coefficient values characteristic of various mesophases were obtained. As expected, the viscosity of LLCs decreases considerably with an increase in shearing rate which is characteristic of liquid crystals being non-Newtonian liquids. Antiseizure properties were determined by means of a four-ball machine (T-02 Tester) and characterized by scuffing load (Pt), seizure load (Poz) and limiting pressure of seizure (poz). Alcohol ethoxylates forming mesophases in aqueous solutions have the strongest effect on the Pt values which are several times higher than those measured in the presence of water. Ethoxylates with higher degrees of ethoxylation exhibit higher values of scuffing load. Those changes have been interpreted as a result of higher cloud points at which those compounds lose their amphiphilic properties. In general, the presence of mesophases in the bulk phase and particularly in the surface phase may lead to the formation of a lubricant film which separates the frictionally cooperating elements of a friction pair. The antiseizure efficiency of alcohol solutions is highest up to the load value which does not exceed the scuffing load value.  相似文献

In this article structures in biological signals are treated. The simpler—directly visible in the signals, which still demand serious methods and algorithms in the feature detection, similarity investigation and classification. The major actions in this domain are of geometric, thus simpler sort, though there are still hard problems related to simple situations. The other large class of less simple signals unsuitable for direct geometric or statistic approach, are signals with interesting frequency components and behavior, those suitable for spectroscopic analysis. Semantics of spectroscopy, spectroscopic structures and research demanded operations and transformations on spectra and time spectra are presented. The both classes of structures and related analysis methods and tools share a large common set of algorithms, all of which aiming to the full automatization. Some of the signal features present in the brain signal patterns are demonstrated, with the contexts relevant in BCI, brain computer interfaces. Mathematical representations, invariants and complete characterization of structures in broad variety of biological signals are in the central focus.  相似文献

Strong-geomaterial samples-such as monolithic rock, concrete, and firm clay-are often weakened by random discontinuities called weak joints (fissures, cracks). The paper focuses on a theoretical strength evaluation of such samples using standard methods of series reliability systems; it is assumed that the shearing failure modes generated by structural joints are independent elements of the system. Special attention is paid to the number k of elements in the system which is randomized by making use of the Poisson probability distribution. Closed-form solutions for the random strength of samples, i.e. distribution functions, are derived for randomly equivalent continua, both anisotropic and isotropic. In particular, such solutions can yield the average strength of the structured sample (the homogenized or the effective one) and its higher-order moments, etc. The examples reveal a strong reduction of the strength if weak joints concentrate around some critical angles, but primarily if the number of discontinuities increases.  相似文献
