A 4589 bp DNA segment containing the Escherichia coli panBCD gene cluster was sequenced, and found to contain 6 complete open reading frames. panB, panC, and panD were identified by subcloning and insertional mutagenesis. The orientation of panD was also confirmed by orientation-specific expression of asparate-1-decarboxylase. panB and panC lie adjacent to one another, but are separated from panD by orf3, which is oriented in the opposite direction. Interruptions in the remaining open reading frames did not affect growth on glucose-minimal medium. No significant similarity to sequences in databases was found for orf1 and orf2. Orf3 contained extensive similarity to reading frames defined by E. coli yjiP, yjiQ, yhgA, and yafD. The function of these amino acid sequences is as yet undefined.  相似文献

Membrane fusion mediated by interaction of the human immunodeficiency virus type 1 (HIV-1) envelope glycoprotein with the human CD4 molecule generally requires that the CD4 be expressed on a human cell. The failure of murine or simian cells expressing human CD4 to form syncytia upon mixing with cells expressing envelope glycoprotein could not be corrected by expression of both molecules at extremely high surface levels using vaccinia virus expression vectors. Video fluorescence microscopic analysis of fluorescent dye transfer between fusing cells indicated that the block occurred at the level of membrane fusion between individual pairs of cells. To gain insight into the basis for this fusion block, we tested the ability of fluorescent probe cells expressing envelope glycoprotein to fuse with transient animal x human hybrid giant cells expressing human CD4. The hybrid giant cells were generated either by low-pH-induced fusion of vaccinia-infected cells or by CD4/HIV-1 envelope glycoprotein-mediated cell fusion. We observed that envelope glycoprotein-expressing probe cells efficiently fused with CD4-expressing animal x human hybrid giant cells, independent of whether the CD4 was originally expressed on the animal or on the human cell. Fusion did not occur with CD4-expressing giant cells derived from animal cells alone. These results indicate that the fusion block is not due to dominant inhibitory components in the animal cell. Rather, they suggest that human cells contain an additional component(s) which, when transferred to the CD4-bearing animal cell, confers the ability to undergo membrane fusion mediated by the HIV-1 envelope glycoprotein.  相似文献

We consider the problem of efficiently sampling Web search engine query results. In turn, using a small random sample instead of the full set of results leads to efficient approximate algorithms for several applications, such as:
•  Determining the set of categories in a given taxonomy spanned by the search results;
•  Finding the range of metadata values associated with the result set in order to enable “multi-faceted search”;
•  Estimating the size of the result set;
•  Data mining associations to the query terms.
We present and analyze efficient algorithms for obtaining uniform random samples applicable to any search engine that is based on posting lists and document-at-a-time evaluation. (To our knowledge, all popular Web search engines, for example, Google, Yahoo Search, MSN Search, Ask, belong to this class.) Furthermore, our algorithm can be modified to follow the modern object-oriented approach whereby posting lists are viewed as streams equipped with a next method, and the next method for Boolean and other complex queries is built from the next method for primitive terms. In our case we show how to construct a basic sample-next(p) method that samples term posting lists with probability p, and show how to construct sample-next(p) methods for Boolean operators (AND, OR, WAND) from primitive methods. Finally, we test the efficiency and quality of our approach on both synthetic and real-world data. A preliminary version of this work has appeared in [3]. Work performed while A. The thickness (d F) and concentration (x) dependence of the Curie temperature of Ni x Cu1?x (d F) ferromagnetic alloy layers (x=0.55, 0.65, d F=[4.5 nm÷12 nm]) in contact with a vanadium layer was studied. The Curie temperature of the ferromagnetic layers depends on the thickness when it is comparable with the transition layer between the F and the vanadium layers, which is attributed to the proximity coupling of the interface region with the rest of the F layer. The present study provides valuable information for fabrication of samples with controlled exchange coupling strength for studies of superconductor/ferromagnet proximity effects.  相似文献

This paper investigated the uniaxial mechanical properties of a new type of hollow sphere structures.
