Calcineurin, a Ca2+/calmodulin-dependent protein phosphatase, plays a critical role in controlling skeletal muscle fiber type. However, little information is available concerning the expression of calcineurin in goat. Therefore, protein phosphatase 3 catalytic subunit alpha isoform (PPP3CA) gene, also called calcineurin Aα, was cloned and its expression characterized in Tianfu goat muscle. Real time quantitative polymerase chain reaction (RT-qPCR) analyses revealed that Tianfu goat PPP3CA was detected in cardiac muscle, biceps femoris muscle, abdominal muscle, longissimus dors muscle, and soleus muscle. High expression levels were found in biceps femoris muscle, longissimus muscle and abdominal muscle (p < 0.01), and low expression levels were seen in cardiac muscle and soleus muscle (p > 0.05). In addition, the spatial-temporal mRNA expression levels showed different variation trends in different muscles with the age of the goats. Western blotting further revealed that PPP3CA protein was expressed in the above-mentioned tissues, with the highest level in biceps femoris muscle, and the lowest level in soleus muscle. In this study, we isolated the full-length coding sequence of Tianfu goat PPP3CA gene, analyzed its structure, and investigated its expression in different muscle tissues from different age stages. Dynamic transboundary water resources allocation based on inflow prediction results is an important task for water resources management in river basins. This paper takes the watershed management agency as the leader and the associated area as the follower, and proposes a two-level asymmetric Nash-Harsanyi Leader-Follower game model considering inflow forecasting errors. In the proposed model, the Monte Carlo method is used to analyze the uncertainty of various stakeholder allocation results and the response regularity to the total water resource uncertainty. The quantitative relationship between the allocation results of stakeholders and the mean and standard deviation of total water resource uncertainty is subsequently established. The Huaihe River basin in China is selected as a case study. The results show the following: (1) the water allocated to the watershed management agency and three provinces has a normal distribution when the inflow forecasting error obeys the normal distribution; (2) the sum of the mean of the water allocated to stakeholders equals the mean of the forecast water resource and the sum of the standard deviations of the water allocated to stakeholders equals the standard deviation of the forecast water resource; (3) the mean and standard deviation of the allocation results have a good linear relationship with the mean and standard deviation of forecast water resource; (4) the distribution parameters of the stakeholder allocation results can be directly derived from the distribution parameters of the forecast information, thus aiding the stakeholders in making decisions and improving the practical value of the method.
Porous WS2 scaffolds with aligned lamellar pore were fabricated by freeze casting, and the pore morphology and size can be well controlled by adjusting the processing parameters during freeze casting process. The results indicated that the porosity and the compressive strength of porous WS2 were greatly affected by the concentration of gelatin. As the gelatin addition increased from 1 to 5 wt%, the porosity of porous WS2 scaffolds with initial solid content of 3 vol% decreased from 95.12 to 90.23%, and their compressive strength increased from 0.22 to 1.16 MPa. Moreover, the lamellar spacing and wall thickness could be tailored from 90 to 320 and 5 to 30 μm respectively by changing the cooling temperature. And the compressive strength of scaffolds has a slight increase with the decreases of cooling temperature. The porous WS2 scaffold with fine aligned lamellar structure and proper compressive strength are expected to be used for scaffold materials. 相似文献
