It is shown that the (normalized) area function based on the Born approximation offers a simple connection between the ultrasonic scattering response and the monochromatic ray sum in X-ray CT (computerized tomography). Because of this simple association, it is possible to apply the ultrasonic signals in the computationally direct and efficient parallel-beam X-ray CT algorithm to reproduce the vertical thickness function of an ultrasonic scatterer. The development of this imaging methodology is demonstrated for flaws of simple geometry; theoretical as well as experimental results for two model scatterers using this imaging technique are reported. Specifically, the area functions for a two-to-one spheroid and a circular cylinder are calculated and applied to a filtered backprojection algorithm of X-ray CT to obtain the vertical thickness function images. These images are then compared with the true vertical thickness functions of the targets based on their geometry. With theoretical data, this method was found to work very well. Even when experimental data containing creeping waves were used, the method produced satisfactory results for objects with continuously smooth surface.  相似文献

A study of the use of millimeter-wavelength spectral transitions to investigate the atmosphere of Mars is presented. In the model experiments investigated it is assumed that a spectrometer in the frequency range from 100 to 260 GHz looks into a modest-sized telescope of from 30 to 50 cm aperture from a near-Mars orbit. The molecules H(2)O, CO, O(2), O(3), and H(2)O(2) all have intense spectral lines in the Mars atmosphere in this frequency range and in addition are all very important in understanding the water cycle, the photochemistry, and the circularization in that atmosphere. It is shown that the altitude and the zonal distribution of H(2)O can be mapped even in atmospheric columns as dry as 0.25 precipital μm. Ozone can be mapped over the entire planet, independent of solar-lighting conditions, dust loading, or clouds in the atmosphere, because millimeter waves are insensitive to any particles that can be suspended in the Mars atmosphere. Because the signal-receiving techniques use superheterodyne devices and narrow spectral lines, zonal and meridional winds can be measured at altitudes above 10 km with a precision approaching approximately 3 m/s by the use of Doppler shifts. Temperature-pressure profiles can be measured to altitudes of 100 km by the use of CO lines in the limb-sounding mode.  相似文献

A new method of ohmic potential drop correction for use with potential step and sweep voltammetric methods is described. The method, iterative correction of the applied potential (ICAP), essentially replaces an electronic positive feedback correction with a digital positive feedback correction. This paper will present one form of the ICAP procedure, in which voltammetric current data acquired by a high-speed digital oscilloscope are used to iteratively generate a compensating potential waveform that is then synthesized by an arbitrary waveform generator. The ICAP method is advantageous for high-speed measurements, since many of the difficulties caused by electronic positive feedback compensation are eliminated.  相似文献

As computer power increases, so too does the range of interesting biomolecular phenomena and properties that can be simulated. It is now possible to simulate complicated protein conformational changes at ambient or physiological temperatures. In this regard, we are attempting to map the conformational transitions of the normal, cellular prion protein (PrP(C)) to its infectious scrapie isoform (PrP(Sc)), which causes neurodegenerative diseases in many mammals. These two forms have identical sequences and are conformational isomers, with heightened formation of beta-sheet structure in the scrapie form. Conversion can be triggered by lowering the pH, but thus far it has been impossible to characterize the conformational change at high resolution using experimental methods. Therefore, to investigate the effect of acidic pH on PrP conformation, we have performed molecular-dynamics simulations of hamster, human and bovine forms of the prion protein in water at neutral and low pH. In all cases the core of the protein is well maintained at neutral pH. At low pH, however, the protein is more dynamic, and the sheet-like structure increases both by lengthening of the native beta-sheet and by addition of a portion of the N-terminus to widen the sheet by another 2-3 strands.  相似文献

Stationary-phase Listeria monocytogenes cells that were either pH dependent acid adapted or not acid adapted were heat challenged at 60 degrees C in a two-level full factorial design for three variables. The three variables and the levels consisted of tryptic soy broth (TSB) and sterile cell-free culture supernatant (sterile TSB), the presence and absence of 1% added glucose, and pH 4.8 and pH 7. Non-acid-adapted cells were most heat resistant when challenged in TSB (mean decimal reduction times at 60 degrees C: D60 = 1.16 min). In the absence of added glucose, non-acid-adapted cells had similar D60-values for inactivations at pH 4.8 and pH 7; however, the presence of glucose caused non-acid-adapted cells challenged at pH 4.8 to be more heat sensitive (D60 = 0.65 min) than those inactivated at pH 7 (D60 = 1.03 min), indicating an interaction between glucose and pH. Overall, the significantly decreased heat resistance of the acid-adapted cells was due to the presence of glucose (D60 = 0.78 min without glucose, D60 = 0.59 min with glucose). Acid-adapted cells heat challenged in TSB had similar D60-values for inactivations at pH 4.8 and pH 7; however, acid-adapted cells in sterile TSB challenged at pH 4.8 (D60 = 0.52 min) had significantly lower heat resistance than did cells challenged at pH 7 (D60 = 0.76 min), indicating an interaction between the medium and pH. The L. monocytogenes survivor data were modeled to extract information on the frequency distribution of heat resistance within heat-challenged populations, and the frequency distribution characteristics of mean, mode, and variance were compared among treatment conditions. Significant differences in the frequency distribution data were compared with the D60-values. These data indicated that the presence and level of cross-protection is highly dependent on the physiological state of the cells and nutrient availability at the time of heat challenge. Such conditions should be considered to ensure that stressed pathogens in foods are destroyed or inactivated.  相似文献

Reactions of methylglyoxal with amino acids, methylamine, and ammonium sulfate can take place in aqueous aerosol and evaporating cloud droplets. These processes are simulated by drying droplets and bulk solutions of these compounds (at low millimolar and 1 M concentrations, respectively) and analyzing the residuals by scanning mobility particle sizing, nuclear magnetic resonance, aerosol mass spectrometry (AMS), and electrospray ionization MS. The results are consistent with imine (but not diimine) formation on a time scale of seconds, followed by the formation of nitrogen-containing oligomers, methylimidazole, and dimethylimidazole products on a time scale of minutes to hours. Measured elemental ratios are consistent with imidazoles and oligomers being major reaction products, while effective aerosol densities suggest extensive reactions take place within minutes. These reactions may be a source of the light-absorbing, nitrogen-containing oligomers observed in urban and biomass-burning aerosol particles.  相似文献
