Starting from suvorexant (trade name Belsomra), we successfully identified interesting templates leading to potent dual orexin receptor antagonists (DORAs) via a scaffold‐hopping approach. Structure–activity relationship optimization allowed us not only to improve the antagonistic potency on both orexin 1 and orexin 2 receptors (Ox1 and Ox2, respectively), but also to increase metabolic stability in human liver microsomes (HLM), decrease time‐dependent inhibition of cytochrome P450 (CYP) 3A4, and decrease P‐glycoprotein (Pgp)‐mediated efflux. Compound 80 c [{(1S,6R)‐3‐(6,7‐difluoroquinoxalin‐2‐yl)‐3,8‐diazabicyclo[4.2.0]octan‐8‐yl}(4‐methyl‐[1,1′‐biphenyl]‐2‐yl)methanone] is a potent and selective DORA that inhibits the stimulating effects of orexin peptides OXA and OXB at both Ox1 and Ox2. In calcium‐release assays, 80 c was found to exhibit an insurmountable antagonistic profile at both Ox1 and Ox2, while displaying a sleep‐promoting effect in rat and dog models, similar to that of the benchmark compound suvorexant. 相似文献

The well-known technique of optical pumping can be used to polarize a neutron beam in a transient way. this kind of actuators in various fields around machining. In this paper an active work piece holder is presented, which is
designed for reducing the dynamic displacement between the tool and the work piece relatively. The paper shows the design
process for the fixture, the finite element analysis of the mechanical parts and the actuators. Also a specially designed
power amplifier is presented which fits best to the needs of voltage and frequency range of the active work piece holder.

A reciprocal impact hypothesis posits an influence of gender-related traits (agency and communion) on role enactment and a reciprocal impact of role enactment on gender-related traits, for both men and women. Specifically, in this study it was predicted that agency influences career success and career success influences agency. In addition, the reciprocal influence of communion and family roles was examined. A prospective study with almost 2,000 university graduates, who were tested after graduation and 1.5 years later, clearly supported the reciprocal impact hypothesis for agency and career success. Communion influenced family roles, but there was no reciprocal influence. Implications for theories of career success and of sex and gender are discussed. This article describes measurements of angular-correlation coefficients in the decay of free neutrons with the superconducting spectrometer PERKEO II. A method for measuring the β-asymmetry coefficient A is presented, as well as a new method for determining the neutrino-asymmetry coefficient B, which allows a value for the proton-asymmetry coefficient C to be obtained for the first time. Designing a high-performance twist drill is difficult due to the complex relationship between drill geometry and numerous and conflicting design goals. Earlier approaches of computer-aided twist drill design are limited to only few design aspects. This article presents a new holistic method of using computing power for twist drill design and optimization. A complete drill geometry model is used to obtain drill performance characteristics and to ensure functional capability. Numerical simulation models calculate structural stiffness and strength, torque and thrust force, coolant flow resistance, chip evacuation capability and chip flute grindability. A multi-objective geometry optimization is realized by implementing metaheuristic optimization algorithms. As a result, a numerical overall optimization of twist drill performance is possible. 相似文献
