The identity of a number of phosphorus-containing metabolites present in Synechocystis sp. PCC 6308 has been confirmed by 31P NMR spectroscopy. The presence of D-ribulose 1,5-bisphosphate (RuBP); DL-glyceraldehyde 3-phosphate (GlyP); D(-)3-phosphoglyceric acid (3PGA); D-ribulose 5-phosphate (Ru5P);6-phosphogluconic acid (6PGA); phosphoenolpyruvate (PEP); inorganic phosphate (Pi); uridine diphosphoglucose (UDPG); ADP and ATP were demonstrated by the pH dependence of their 31P NMR chemical shifts in spectra of perchloric acid cell extracts. Intracellular pH of cells was determined to be 7.5-7.7.  相似文献

The authors of this study investigated task switching following cerebellar damage. The study group consisted of 7 children and adolescents (M age = 13.8 years) who underwent surgical removal of a benign posterior fossa tumor. They were tested at a sufficient interval after surgery (M lag = 6.13 years) for restoration of normal cognitive skills and intelligence. Although all showed normal learning of the task compared with control participants, when rapid behavioral changes were required (short preparation time), they exhibited behavioral rigidity manifested by enhanced switching cost. These results are in line with another study on serial reaction time with the same patients (A. Berger et al., in press). They have important implications for our understanding of the cognitive sequelae of early cerebellar damage as well as the involvement of the cerebellum in task switching. Individual fluorescent polystyrene nanospheres (<10-100-nm diameter) and individual fluorescently labeled DNA molecules were dispersed on mica and analyzed using time-resolved fluorescence spectroscopy and atomic force microscopy (AFM). Spatial correlation of the fluorescence and AFM measurements was accomplished by (1) positioning a single fluorescent particle into the near diffraction-limited confocal excitation region of the optical microscope, (2) recording the time-resolved fluorescence emission, and (3) measuring the intensity of the excitation laser light scattered from the apex of an AFM probe tip and the AFM topography as a function of the lateral position of the tip relative to the sample substrate. The latter measurements resulted in concurrent high-resolution (approximately 10-20 nm laterally) images of the laser excitation profile of the confocal microscope and the topography of the sample. Superposition of these optical and topographical images enabled unambiguous identification of the sample topography residing within the excitation region of the optical microscope, facilitating the identification and structural characterization of the nanoparticle(s) or biomolecule(s) responsible for the fluorescence signal observed in step 2. These measurements also provided the lateral position of the particles relative to the laser excitation profile and the surrounding topography with nanometer-scale precision and the relationship between the spectroscopic and structural properties of the particles. Extension of these methods to the study of other types of nanostructured materials is discussed.  相似文献

Only low antibody levels were obtained from vaccinating human volunteers with single-chain peptide from the Plasmodium falciparum circumsporozoite protein (PfCSP). This resulted in modest protection against sporozoite challenge. In addition, HLA restriction limits the probability of synthesis of a vaccine effective for a diverse population. We report immunization studies with a multiple antigen peptide (MAP) system consisting of multiple copies of a B-cell epitope from the central repeat region of the PfCSP in combination with a universal T-cell epitope, the P2P30 portion of tetanus toxin. This MAP4(NANP)6P2P30 vaccine was highly immunogenic in four different strains of mice when used with various safe and nontoxic adjuvants. When this MAP vaccine was encapsulated in liposomes with lipid A and adsorbed to aluminium hydroxide and given three times at 4-week intervals, the resultant antibody prevented 100% of sporozoites from invading and developing into liver stage infection. This high degree of immunogenicity of MAP4(NANP)6P2P30 vaccine formulated in liposomes, lipid A and aluminum hydroxide provides the foundation for consideration of human trials with this formulation.  相似文献

The effect of wire delay on circuit timing typically increases when an existing layout is migrated to a new generation of process technology, because wire resistance and cross capacitances do not scale well. Hence, careful sizing and spacing of wires is an important task in migration of a processor to next generation technology. In this paper, timing optimization of signal buses is performed by resizing and spacing individual bus wires, while the area of the whole bus structure is regarded as a fixed constraint. Four different objective functions are defined and their usefulness is discussed in the context of the layout migration process. The paper presents solutions for the respective optimization problems and analyzes their properties. In an optimally-tuned bus layout, after optimizing the most critical signal delay, all signal delays (or slacks) are equal. The optimal solution of the MinMax problem is always bounded by the solution of the corresponding sum-of-delays problem. An iterative algorithm to find the optimally-tuned bus layout is presented. Examples of solutions are shown, and design implications are derived and discussed.  相似文献

The selectivity of a sequence of isothermal irreversible heterogeneous reactions
within a catalytic pellet is investigated under conditions where intraparticle forced convection is a non-negligible transport mechanism. The nature of the effect of the internal filter flow depends mostly on the value of the Thiele modulus, φ2, of the desired (middle) species. The asymptotic values of the selectivity, as φ2 → 0 and φ2 → ∞, are unaltered, independent of the strength of the internal flow. The intermediate region is divided into two parts. In the subregion with lower values of φ2 the selectivity is enhanced. In the other subregion, with higher values of φ2, the selectivity is significantly retarded to values which can be markedly below the lower asymptote. These trends are common to both poor and good selective catalysts.  相似文献

BACKGROUND: Timely diagnosis and continued monitoring of patients with type I Gaucher disease is critical because skeletal involvement can permanently disable patients and visceral organ involvement can lead to abdominal pain and secondary hematologic and biochemical complications. OBJECTIVE: To seek clinical consensus for minimum recommendations for effective diagnosis and monitoring of patients with type I Gaucher disease. PARTICIPANTS, EVIDENCE, AND CONSENSUS PROCESS: Contributing authors collaborated in quarterly meetings over a 2-year period to synthesize recommendations from peer-reviewed publications and their own medical experiences. These physicians care for most patients with Gaucher disease in the United States and serve as the US Regional Coordinators for the International Collaborative Gaucher Group Registry, the world's largest database for this disorder. CONCLUSIONS: The definitive method of diagnosis is enzyme assay of beta-glucocerebrosidase activity. Schedules differ for monitoring complications of type I Gaucher disease, depending on symptoms and whether enzyme replacement therapy is used. Hematologic and biochemical involvement should be assessed by complete blood cell count, including platelets, acid phosphatase, and liver enzymes, at baseline and every 12 months in untreated patients and every 3 months and at enzyme replacement therapy changes in treated patients. Visceral involvement should be assessed at diagnosis using magnetic resonance imaging or computed tomographic scans. Skeletal involvement should be assessed at diagnosis using T1- and T2-weighted magnetic resonance imaging of the entire femora and plain radiography of the femora, spine, and symptomatic sites. Follow-up skeletal and visceral assessments are recommended every 12 to 24 months in untreated patients, and every 12 months and at enzyme replacement therapy changes in treated patients.  相似文献

On-chip power distribution grids with multiple supply voltages are discussed in this paper. Two types of interdigitated and paired power distribution grids with multiple supply voltages and multiple grounds are presented. Analytic models are also developed to estimate the loop inductance in four types of proposed power delivery schemes. Two proposed schemes, fully and pseudo-interdigitated power delivery, reduce power supply voltage drops as compared to conventional interdigitated power distribution systems with dual supplies and a single ground by, on average, 15.3% and 0.3%, respectively. The performance of the proposed on-chip power distribution grids is compared to a reference power distribution grid with a single supply and a single ground. The voltage drop in fully interdigitated and fully paired power distribution grids with multiple supplies and multiple grounds is reduced, on average, by 2.7% and 2.3%, respectively, as compared to the voltage drop of an interdigitated power distribution grid with a single supply and a single ground. The proposed power distribution grids are a better alternative to a single supply voltage and a single ground power distribution system. On-chip resonances in power distribution grids with decoupling capacitors are intuitively explained in this paper, and circuit design implications are provided. It is also noted that fully interdigitated and fully paired power distribution grids with multiple supply voltages and multiple grounds are recommended to decouple power supply voltages.  相似文献

Observations show that, when a viscous fluid flows vertically through a bed of settled flocculated aggregates, particles are emerged from locations on the upper surface of the bed, as a volcano process, and hollow tubes are created in the bed though the latter remains basically stable. A theoretical model is presented to explain this odd phenomenon from the onset of particle motion to the development of the tubular cavities and the particles' deposits around them. Analytical and numerical calculations are performed to show fluid streamlines and particle trajectories.  相似文献

Crosstalk noise in on-chip interconnect plays a major role in the performance of modern integrated circuits. Multi-aggressor capacitive and inductive coupling complicates both the modeling and mitigation of the noise. A novel method to model and analyze noise in RLC multi-line structures is proposed in this paper, exhibiting an error of up to 9% as compared to SPICE. This method is physically intuitive since it decomposes the noise produced by each of the aggressors into individual capacitive and inductive noise sources. The proposed model and related layout noise mitigation guidelines are applied to crosstalk noise reduction in multi-line structures.  相似文献
