Nucleotide and nucleotide sugar analysis by liquid chromatography-electrospray ionization-mass spectrometry on surface-conditioned porous graphitic carbon

We examined the analysis of nucleotides and nucleotide sugars by chromatography on porous graphitic carbon with mass spectrometric detection, a method that evades contamination of the MS instrument with ion pairing reagent. At first, adenosine triphosphate (ATP) and other triphosphate nucleotides exhibited very poor chromatographic behavior on new columns and could hardly be eluted from columns previously cleaned with trifluoroacetic acid. Satisfactory performance of both new and older columns could, however, be achieved by treatment with reducing agent and, unexpectedly, hydrochloric acid. Over 40 nucleotides could be detected in cell extracts including many isobaric compounds such as ATP, deoxyguanosine diphosphate (dGTP), and phospho-adenosine-5'-phosphosulfate or 3',5'-cyclic adenosine 5'-monophosphate (AMP) and its much more abundant isomer 2',3'-cyclic AMP. A fast sample preparation procedure based on solid-phase extraction on carbon allowed detection of very short-lived analytes such as cytidine 5'-monophosphate (CMP)-2-keto-deoxy-octulosonic acid. In animal cells and plant tissues, about 35 nucleotide sugars were detected, among them rarely considered metabolites such as uridine 5'-diphosphate (UDP)-l-arabinopyranose, UDP-L-arabinofuranose, guanosine 5'-diphosphate (GDP)-L-galactofuranose, UDP-L-rhamnose, and adenosine diphosphate (ADP)-sugars. Surprisingly, UDP-arabinopyranose was also found in Chinese hamster ovary (CHO) cells. Due to the unique structural selectivity of graphitic carbon, the method described herein distinguishes more nucleotides and nucleotide sugars than previously reported approaches.

Unser Ansatz nutzt Methoden der ,,Shot-Segmentierung“ um ein Video tempor?r zu segmentieren und einen ,,k-Means“ Algorithmus um Repr?sentanten für jeden Shot zu bestimmen. Zus?tzlich führen wir ein ,,Meta-Clustering“ auf den extrahierten Keyframes aus um kompakte Videozusammenfassungen zu erhalten. Um unsere Methoden zu testen haben wir diese auf einer Datenbank von YouTube Videos angewendet. Wir erhielten Ergebnisse, welche (1) eine Verbesserung des Retrievals und (2) kompakte Video-Zusammenfassungen zeigen.     

A flexible adaptive FIR filter VLSI IC

The architecture and features of the Motorola DSP56200 are described. The DSP56200 is an algorithm-specific cascadable digital signal processing peripheral designed to perform the computationally intensive tasks associated with finite impulse response (FIR) and adaptive FIR digital filtering applications. The DSP56200 is implemented in high-performance, low-power 1.5-μm HCMOS technology and is available in a 28-pin DIP package. The on-chip computation unit includes a 97.5-ns 24-bit×24-bit coefficient RAM, and a 256-bit×16-bit data RAM. Three modes of operation allow the part to be used as a single, dual, or single adaptive FIR filter, with up to 256 taps per chip. In the adaptive mode, the part performs the FIR filtering and least-mean-square (LMS) coefficient update operations for a single tap in 195 ns, permitting use of the part as a 19-kHz sampling rate, 256-tap adaptive FIR filter. A programmable DC tap, coefficient leakage, and adaptation coefficient parameters in the adaptive FIR mode allow the DSP56200 to be used in a wide variety of adaptive FIR filtering applications. The performance of the part in an echo canceler configuration is presented. Typical applications of the part are also described     

An abdominal CCD camera for the continuous video recording of sheep ovary images during the reproductive cycle

An intra-abdominal ovary-isolating observation system based on a charge-coupled device (CCD) camera and intended for continuously recording video images of the ovary during the reproductive cycle was constructed and surgically implanted in ewes. A time-related label was generated automatically for the identification of each frame. Results demonstrate the importance and unique capabilities of bio-implantable cameras for long-term studies.
