BACKGROUND: Heterogeneous photocatalysis is influenced by a number of parameters involving synergistic effects; hence, an experimental strategy design that considers interactions between the main variables is needed. The response surface methodology was applied for the investigation of photodegradation of 20 mg L?1 Orange II in aqueous solutions and for optimization of color removal efficiency. Preliminary studies were performed to identify the parameters to be selected for optimization. RESULTS: The input variables considered for experimental design were: solution initial pH, oxidizing agent (H2O2) initial concentration and UV‐A irradiation time. The multivariate experimental design allowed the development of a quadratic response surface model to be used for the prediction of color removal efficiency over the full range of the experimental region. Under the optimum conditions established in the region of experimentation (pH = 6.9, [H2O2]0 = 183 mg L?1 and t = 32 min), a 100% color removal efficiency was obtained in experiments. CONCLUSIONS: It was found that the variables considered have important effects on color removal efficiency. The results demonstrate that the use of experimental design strategy is indispensable for successful investigation and adequate modeling of the process because the interdependence of the reaction parameters cannot be neglected. The cytoarchitecture and tensile characteristics of ocular lenses play a crucial role in maintaining their transparency and deformability, respectively, which are properties required for the light focusing function of ocular lens. Calcium-dependent myosin-II-regulated contractile characteristics and mechanosensitive ion channel activities are presumed to influence lens shape change and clarity. Here, we investigated the effects of load-induced force and the activity of Piezo channels on mouse lens myosin II activity. Expression of the Piezo1 channel was evident in the mouse lens based on immunoblot and immufluorescence analyses and with the use of a Piezo1-tdT transgenic mouse model. Under ex vivo conditions, change in lens shape induced by the load decreased myosin light chain (MLC) phosphorylation. While the activation of Piezo1 by Yoda1 for one hour led to an increase in the levels of phosphorylated MLC, Yoda1 treatment for an extended period led to opacification in association with increased calpain activity and degradation of membrane proteins in ex vivo mouse lenses. In contrast, inhibition of Piezo1 by GsMTx4 decreased MLC phosphorylation but did not affect the lens tensile properties. This exploratory study reveals a role for the mechanical load and Piezo1 channel activity in the regulation of myosin II activity in lens, which could be relevant to lens shape change during accommodation. 相似文献

Glioblastoma leads to a fatal course within two years in more than two thirds of patients. An essential cornerstone of therapy is chemotherapy with temozolomide (TMZ). The effect of TMZ is counteracted by the cellular repair enzyme O6-methylguanine-DNA methyltransferase (MGMT). The MGMT promoter methylation, the main regulator of MGMT expression, can change from primary tumor to recurrence, and TMZ may play a significant role in this process. To identify the potential mechanisms involved, three primary stem-like cell lines (one astrocytoma with the mutation of the isocitrate dehydrogenase (IDH), CNS WHO grade 4 (HGA)), and two glioblastoma (IDH-wildtype, CNS WHO grade 4) were treated with TMZ. The MGMT promoter methylation, migration, proliferation, and TMZ-response of the tumor cells were examined at different time points. The strong effects of TMZ treatment on the MGMT methylated cells were observed. Furthermore, TMZ led to a loss of the MGMT promoter hypermethylation and induced migratory rather than proliferative behavior. Cells with the unmethylated MGMT promoter showed more aggressive behavior after treatment, while HGA cells reacted heterogenously. Our study provides further evidence to consider the potential adverse effects of TMZ chemotherapy and a rationale for investigating potential relationships between TMZ treatment and change in the MGMT promoter methylation during relapse. 相似文献

The identification of coexpressed genes from microarray data is a challenging problem in bioinformatics and computational biology. The objective of this study is to obtain knowledge about the most important genes and clusters related to production outputs of real-world time-series microarray data in the industrial microbiology area. Each sample in the microarray data experiment is complemented with the measurement of the corresponding production and growth values. A novel aspect of this research refers to considering the relation of coexpression patterns with the measured outputs to guide the biological interpretation of results. Shape-based clustering models are developed using the pattern of gene expression values over time and further incorporating knowledge about the correlation between the change in the gene expression level and the output value. Experiments are performed for time-series microarray of bacteria, and an analysis from a biological perspective is carried out. The obtained results confirm the existence of relationships between output variables and gene expressions. Moreover, the shape-based clustering methods show promising results, being able to guide metabolic engineering actions with the identification of potential targets. 相似文献

In order to enhance properties of the polymer composite films, composite coatings based on polypyrrole (PPy) and polyethylene glycol (PEG), using various percentages of PEG (0.5%, 2%, 4%), were electrodeposited on a new titanium alloy electrode as insulating material. The structure of the coatings was investigated via infrared (FTIR) analysis, and the surface features were studied using contact angle determination, atomic force microscopy (AFM) and scanning electronic microscopy (SEM). When testing the antibacterial properties of the coatings, the best effect was found for the coating with 2% PEG concentration, which has hydrophilic character and small roughness. Such results are in concordance with mechanism of biomaterial–bacteria interaction which involves as factors affecting bacterial adhesion and growth an initial physicochemical interaction stage, where roughness and wettabilitty are factors promoting bacterial adhesion and biofilm deposition. 相似文献

This article presents a series of preliminary results regarding the electrodeposition of bismuth, tellurium, and bismuth telluride films at 60 °C from ionic liquids, containing a mixture of choline chloride and oxalic acid (ChCl–OxA). Ten millimolar concentration solutions of BiCl3 and TeO2 were used as precursors in this supporting electrolyte. Cyclic voltammetry and electrochemical impedance spectroscopy techniques were used to demonstrate the deposition processes on Pt and Cu electrodes. Long-time electrolyses (30–120 min) performed at 60 °C with potential control (between ?0.22 and ?0.37 V vs. Ag reference electrode) have resulted in films deposited on copper substrate. Film surfaces were studied by scanning electron microscopy and analyzed by energy dispersive X-ray spectroscopy. The results of this study show that ChCl–OxA ionic liquid may be considered as a promising substitute of aqueous baths for Bi, Te or Bi2Te3 film plating. 相似文献

Four lead(II) coordination polymers were isolated under hydro(solvo)thermal conditions. The applied synthetic methodology takes advantage of the coordination behaviour of a new bifunctional organoarsonate ligand, 4-(1, 2, 4-triazol-4-yl)phenylarsonic acid (H2TPAA) and involves the variation of lead(II) reactants, metal/ligand mole ratios, and solvents. The constitutional composition of the four lead(II) coordination polymers can be formulated as [Pb2(TPAA)(HTPAA)(NO3)]·6H2O (1), [Pb2(TPAA)(HTPAA)2]·DMF·0.5H2O (DMF = N, N-Dimethylformamide) (2), [Pb2Cl2(TPAA)H2O] (3), and [Pb3Cl(TPAA)(HTPAA)2H2O]Cl (4). The compounds were characterized by single-crystal and powder x-ray diffraction techniques, thermogravimetric analyses, infra-red spectroscopy, and elemental analyses. Single-crystal x-ray diffraction reveals that 1 and 2 represent two-dimensional (2D) layered structures whilst 3 and 4 form three-dimensional (3D) frameworks. The structures of 1, 2, and 4 contain one-dimensional (1D) {PbII/AsO3} substructures, while 3 is composed of 2D {PbII/AsO3} arrays. Besides their interesting topologies, 1–4 all exhibit photoluminescence properties in the solid state at room temperature. 相似文献
