The mechanical degradation of polysaccharides was investigated using dynamic high and ultra-high-pressure homogenization (HPH). The objectives were to reduce the molar mass of polymer chains, and simultaneously, the apparent and intrinsic viscosity of polysaccharides in solution. The influence of homogenization pressure (up to 200 MPa) and cycles was compared on polysaccharides with different physical and structure properties: namely, guar gum, hydroxyethylcellulose (HEC), sodium carboxymethylcellulose (Na-CMC), sodium alginate (Na-alginate) and gum arabic. HPH was applied on semi-dilute solutions. The apparent changes in molar mass, gyration radius and intrinsic viscosity were deduced from size exclusion chromatography coupled on-line with multi-angle laser light scattering, differential viscometer detector and differential refractive index detector (SEC/MALS/DV/DRI), while the evolution of the critical overlap concentration (C) was obtained by viscosimetry. A method based on a succession of homogenization cycles and polymer pre-concentration steps was developed to determine the minimum molar mass achieved at constant pressure. Molar mass, and intrinsic viscosity were shown to fall simultaneously while logically C increased during HPH for all polysaccharides, except gum arabic, probably because of its globular and branched structure. This highlights that the differences of polysaccharide structures and conformation (linear, branched…) exhibit a stronger impact on HPH treatments than polymer charge or molar mass. Finally, via an empirical approach linking the decrease of both molar masses and viscosities, we have evidenced a specific scaling exponent that should characterize the flexibility of the treated polymer (i.e. its ability to be degraded by HPH).  相似文献

Rheological and swelling properties of hydrogels based on pullulan crosslinked with sodium trimetaphosphate (STMP) are explained according to various polymer and crosslinking agent concentrations using 31P-nuclear magnetic resonance study. This method has allowed determining the amount of all the species present in the medium when varying both pullulan and STMP concentrations. We have clearly demonstrated with a good agreement by both 31P-NMR and rheology that a critical STMP concentration occurs which is function of pullulan concentration. This typical crosslinking agent concentration delimitates the maximum of gel structure together with the minimum of swelling.  相似文献

Two parties, Alice and Bob, wish to distill a binary secret key out of a list of correlated variables that they share after running a quantum key distribution (QKD) protocol based on continuous-spectrum quantum carriers. We present a novel construction that allows the legitimate parties to get equal bit strings out of correlated variables by using a classical channel, with as little leaked information as possible. This opens the way to securely correcting nonbinary key elements. In particular, the construction is refined to the case of Gaussian variables as it applies directly to recent continuous-variable protocols for QKD.  相似文献

Agriculture is facing increasing innovation challenges to meet current societal expectations, yet very few design science studies are devoted to it. This paper highlights some of the particularities of the objects, reasoning and organization of design in agriculture that may open fruitful scientific dialogue between design scientists and agricultural scientists. We first provide an overview of the broad range of objects that are designed in agriculture and point out their specific characteristics with regard to design. We then identify some particular challenges of design activities in agriculture and review how they have been addressed up to now. Finally, we discuss how design challenges and characteristics in agriculture can contribute to current debate in the field of design science. We propose two main lines of enquiry and debate: enhancing the links between design reasoning and organization and further conceptualizing the status of use situations in design to deal with uncertainties and complexity in design processes.  相似文献

The paper discusses the enzymatic behaviour of a series of copolymers composed of alginate grafted with poly(ε‐caprolactone) (PCL) of various lengths and degrees of substitution. The study is focused on viscosity measurements and pyrene probe fluorescence with or without two enzymes: alginate lyase, which breaks the alginate backbone; and esterase, which breaks PCL pendent groups. Alginate lyase is inactive at pH = 3.8 and degrades quickly all copolymers at pH = 6.3. The degradation is not complete and is slowed down by the presence of PCL. Esterase degrades only copolymers with long pendent PCL groups. It has no effect on copolymers when PCL has a length of 530 g mol?1. We show that the HACCP system is not fully applicable at the primary production level, and that food safety is obtained through the careful implementation of good hygiene practice (GHP) at the farm. Guides to GHP intended at primary productions cover one activity, such as cattle rearing or corn production. Yet most farms have more than one activity. Hazards from one activity can be transferred to another activity, and in general this is not considered in one-activity guides. There is a need therefore for farmers to be able to detect and control such transfers. We suggest a horizontal complement to the Guides to GHP that would consider only the application of the Step 6 and Principle 1 of the HACCP system amended as follows: “List all potential hazards transferred from one activity to another activity within the farm, conduct a hazard analysis, and consider any measures to control identified hazards”. An outline in two parts is proposed. The first part would help at checking if all generic GHP are in place; the second part would help at disclosing interactions between farm specific activities. Detected hazard transfers could then be controlled with measures that are already described in existing guides. The horizontal complement would benefit from being drafted at the European Community level.  相似文献

Graphene represents the ultimate substrate for high-resolution transmission electron microscopy, but the deposition of biological samples on this highly hydrophobic material has until now been a challenge. We present a reliable method for depositing ordered arrays of individual elongated DNA molecules on single-layer graphene substrates for high-resolution electron beam imaging and electron energy loss spectroscopy analysis. This method is a necessary step toward the observation of single elongated DNA molecules with single base spatial resolution to directly read genetic and epigenetic information.  相似文献
