The Sulfolobus solfataricus, strain MT4, ß-glycosidase(Ssßgly) is a thermophilic member of glycohydrolasefamily 1. To identify active-site residues, glutamic acids 206and 387 have been changed to isosteric glutamine by site-directedmutagenesis. Mutant proteins have been purified to homogeneityusing the Schistosoma japonicum glutathione S-transferase (GST)fusion system. The proteolytic cleavage of the chimeric proteinwith thrombin was only obtainable after the introduction ofa molecular spacer between the GST and the Ssß-glydomains. The Glu387 Gin mutant showed no detectable activity,as expected for the residue acting as the nucleophile of thereaction. The Glu206 Gin mutant showed 10- and 60-fold reducedactivities on aryl-galacto and aryl-glucosides, respectively,when compared with the wild type. Moreover, a significant Kmdecrease with plo-nitrophenyl-ß-D-glucoside was observed.The residual activity of the Glu206 Gln mutant lost the typicalpH dependence shown by the wild type. These data suggest thatGlu206 acts as the general acid/base catalyst in the hydrolysisreaction.  相似文献

A viable way to develop optimal control strategies for multi-particle quantum systems is to consider the framework of time-dependent density functional theory (TDDFT), where low-dimensional nonlinear Schrödinger models are developed to compute the electronic density of related high-dimensional linear Schrödinger equations. Among these models, the Kohn-Sham TDDFT system allows to accommodate control mechanisms in the same potentials that appear in the original multi-dimensional Schrödinger equations, thus allowing a physical interpretation and a laboratory implementation. The purpose of this paper is the mathematical analysis of optimal control problems governed by the time-dependent Kohn-Sham (TDKS) equations including a control potential that has the purpose to drive the evolution of the electron density to perform given tasks. For the resulting optimal control problems, existence of optimal solutions is proved and their characterization as solutions of TDKS optimality systems is investigated.  相似文献

We propose a novel asynchronous all-optical circuit for extraction and serial-to-parallel conversion of label bits from differential phase-shift keying (DPSK) packets. The circuit requires only two optical switches regardless of the number of bits to be extracted and parallelized from the packet. Experimental evidence of practical use of the circuit to four bit labels at 10 Gb/s is provided. The circuit is scalable with the number of bits, operates at low input power, and is suitable for photonic integration. The asynchronous nature of the circuit allows us to efficiently extract/read one specific label field of variable length without processing the entire label, leading to a simplified architecture of the label processing circuit  相似文献

Fast tunable wavelength conversion for all-optical packet switching   总被引:1,自引:0,他引:1  
We demonstrate a simple scheme to achieve fast tunable wavelength conversion over a wide spectral range. The wavelength conversion is obtained by four-wave mixing in a semiconductor optical amplifier using a double-pump scheme, 2048-bit sequences at 2.5 Gb/s, incoming at a given wavelength, are alternatively converted toward two different wavelengths for a total conversion interval of 20 nm. With this method, the time needed to change the destination wavelength is much shorter than the bit duration. This feature makes any guard-time spacing between packets unnecessary. Bit-error-rate measurements show that the process occurs with negligible penalties (less than 0.3 dB).  相似文献

The European ACTS project optical pan-European network (OPEN) aims at assessing the feasibility of an optical pan-European overlay network, interconnecting major European cities by means of a mesh of high-capacity optical fiber links, cross-connected through transparent photonic nodes. Both the transmission links and the routing network elements rely on wavelength division multiplexing (WDM) all-optical technologies, such as wavelength translation. This paper presents results obtained in the following domains covered within the project: network topology considerations (optimization and dimensioning); network physical layer simulation; fabrications of packaged functional modules based on advanced optoelectronic devices; laboratory demonstrations of N×10 Gb/s transmission and routing; feasibility of an optical time division multiplexing/WDM (OTDM/WDM) interface; and the field implementation of a 4×4 multiwavelength crossconnect prototype, featuring all-optical space and wavelength routing. This implementation was realized in two cross-border field trials, one conducted between Norway and Denmark and the other between France and Belgium. The final results of the Norway to Denmark field trials are presented, featuring the successful cascade of three wavelength-translating optical crossconnects (OXCs), along with the transmission over 1000 km of a mix of standard/submarine cable links for four channels at 2.5 Gb/s  相似文献

A novel all-optical label processor/erasure for DPSK spectrally separated label and payload is demonstrated. Experimental results show the recognition of two different labels at 12.5 Gbit/s at distinct outputs, while the label is simultaneously erased allowing for a new label insertion. The low power and asynchronous label processor/erasure can be scaled to distinguish a larger number of labels, to operate at higher bit rate, and then can be potentially used in an optical label switching network.  相似文献

The in vivo and in vitro effects of the insecticide deltamethrin (DM) on hepatic cytochrome P450 (Cyt P450) monooxygenase were examined in adult carp. The in vivo experiments were carried out with 0.2 microgram/l DM at 20 degrees C. The changes in the hepatic microsomal Cyt P450 content and the Cyt P450-dependent monooxygenase activities were studied in DM-treated fish. Although there were no changes in the Cyt P450 content during the exposure time, after treatment for 24 h all the investigated isoenzyme activities (para-nitrophenetole-O-deethylase, p-NPOD; aminopyrene-N-demethylase, APND; ethylmorphine-N-demethylase, EMND; 7-ethoxycoumarin-O-deethylase, ECOD; and ethoxyresorufin-O-deethylase, EROD) were significantly inhibited. After 72 h, all the activities were still lower than in the control animals. In vitro incubation of liver microsomes with DM led to a concentration-dependent decrease in total microsomal Cyt P450 content. A complete loss of Cyt P450 occurred after a 5-min incubation with 60 microM DM. The maximum in the difference spectra of microsomes was shifted to higher wavelength, showing the strong interaction of DM with Cyt P450. EROD and ECOD activities were inhibited by DM. The in vitro kinetic results on ECOD revealed that the inhibition was of non-competitive type, with K1 = 9.8 +/- 2.3 microM. This study indicates important biochemical effects of DM in fish liver, and suggests that exposure to DM may cause loss of the Cyt P450-dependent metabolism in fish.  相似文献

We experimentally investigate the probability density function (pdf) of sampled bits in common intensity-modulated direct-detection systems in linear and nonlinear regime, both single-channel and wavelength-division-multiplexed. To this aim, we use a recirculating fiber loop and selectively obtain the sample pdf for the various bits in a given sequence, which allows separating the intersymbol interference (ISI) effects. Our measurements confirm a recent theoretical simplified model, suggesting that, under the above conditions, a generalized analytic form is a good approximation of the true pdf, provided that a pattern-dependent effective noise density and noiseless signal value are assumed: all the pdfs estimated from the experimental data are indeed well fitted by this analytic form. Moreover, when nonlinear effects become dominating, the analytic form is still fitting properly the data histograms, showing, however, that the effective noise density increases. In the above regime, we clearly observe that ISI also affects the increase of the effective noise density.  相似文献
