Direct allorecognition is the earliest and most potent immune response against a kidney allograft. Currently, it is thought that passenger donor professional antigen-presenting cells (APCs) are responsible. Further, many studies support that graft ischemia-reperfusion injury increases the probability of acute rejection. We evaluated the possible role of primary human proximal renal tubular epithelial cells (RPTECs) in direct allorecognition by CD4+ T-cells and the effect of anoxia-reoxygenation. In cell culture, we detected that RPTECs express all the required molecules for CD4+ T-cell activation (HLA-DR, CD80, and ICAM-1). Anoxia-reoxygenation decreased HLA-DR and CD80 but increased ICAM-1. Following this, RPTECs were co-cultured with alloreactive CD4+ T-cells. In T-cells, zeta chain phosphorylation and c-Myc increased, indicating activation of T-cell receptor and co-stimulation signal transduction pathways, respectively. T-cell proliferation assessed with bromodeoxyuridine assay and with the marker Ki-67 increased. Previous culture of RPTECs under anoxia raised all the above parameters in T-cells. FOXP3 remained unaffected in all cases, signifying that proliferating T-cells were not differentiated towards a regulatory phenotype. Our results support that direct allorecognition may be mediated by RPTECs even in the absence of donor-derived professional APCs. Also, ischemia-reperfusion injury of the graft may enhance the above capacity of RPTECs, increasing the possibility of acute rejection.  相似文献

The main purpose of the TARC (Transmutation by Adiabatic Resonance Crossing) experiment (PS-211), was to demonstrate the possibility to destroy efficiently Long-Lived Fission Fragments (LLFF) in Accelerator Driven Systems (ADS). The experimental set-up which consisted of a lead block with dimensions 3.3 x 3.3 x 3 m3, was installed in a CERN Proton Synchrotron (PS) beam line. The proton beam at 2.5 GeV/c and 3.5 GeV/c, was incident in the centre of the lead block assembly producing neutrons via spallation reactions. In this study, neutron flux measurements are presented in the lead block assembly using thermoluminescence and nuclear track detectors. The results are in good agreement with Monte Carlo calculations as well as with the results of the other methods used in the framework of the TARC experiment.  相似文献

We discuss new theoretical and experimental results on the dynamic rate shaping (DRS) approach for transcoding compressed video bitstreams (MPEG-1, MPEG-2, MPEG-4, H.261, as well as JPEG). We analyze the behavior of DRS assuming a first order autoregressive source. We propose a set of low complexity algorithms for both constrained and unconstrained DRS and substantiate the almost-optimal experimental performance of the memoryless algorithm by assuming a first order autoregressive source. By deriving the statistical and rate-distortion characteristics of different components of the interframe rate shaping problem, we offer an explanation as to why the set of optimal breakpoint values for any frame is somewhat invariant to the accumulated motion compensated shaping error from past frames. We also present an extensive experimental study on the various DRS algorithms (causally optimal, memoryless, and rate-based) both in their constrained and generalized forms. The study proves the computational viability of the DRS approach to transcoding and identifies a range of rate shaping ratios for which it is better than requantization, both complexity-wise as well as in performance. This result is significant in that it opens up the way to construct much simpler memoryless algorithms that give minimal penalty in achieved quality, not just for this but possibly other types of algorithms. This is also the very first use of matrix perturbation theory for tracking the spectral behavior of the autocorrelation matrix of the source signal and the motion residual it yields.  相似文献

We propose an optimal buffered compression algorithm for shape coding as defined in the forthcoming MPEG-4 international standard. The MPEG-4 shape coding scheme consists of two steps: first, distortion is introduced by down and up scaling; then, context-based arithmetic encoding is applied. Since arithmetic coding is "lossless," the down up scaling step is considered as a virtual quantizer. We first formulate the buffer-constrained adaptive quantization problem for shape coding, and then propose an algorithm for the optimal solution under buffer constraints. Previously, the fact that a conversion ratio (CR) of 1/4 makes a coded image irritating to human observers for QCIF size was reported for MPEG-4 shape coding. Therefore, careful consideration for small size images such as QCIF should be given to prevent coded images from being unacceptable. To this end, a low bit rate tuned algorithm is proposed in this paper as well. Experimental results are given using an MPEG-4 shape codec.  相似文献

OBJECTIVE: In this study the effect of the hemorheological agent pentoxifylline on the pressure of esophageal varices was investigated in portal hypertensive cirrhotic patients. METHODS: Intravariceal pressure was measured endoscopically using the direct puncture technique in 20 patients. Measurements were obtained under baseline conditions and 30 min after double-blind administration of pentoxifylline (1.4 mg/kg BW, n = 10 patients) or an identical volume of NaCl 0.9% solution (n = 10 patients). RESULTS: Under baseline conditions, intravariceal pressure was similar in pentoxifylline and placebo groups (17.3+/-5.5 mm Hg vs 18.8+/-4.6 mm Hg, respectively; p = N.S.). Placebo administration had no significant effect on intravariceal pressure (18.8+/-4.6 mm Hg vs 18.3+/-4.1 mm Hg; p = N.S.). In contrast, pentoxifylline caused a highly significant reduction of intravariceal pressure, (from 17.3+/-5.5 mm Hg to 11.4+/-5.9 mm Hg; p = 0.0001), the overall mean reduction being 36.1+/-14.1% mm Hg. CONCLUSIONS: We concluded that pentoxifylline, by reducing blood flow viscosity, caused a significant decrease in variceal pressure in patients suffering from portal hypertension.  相似文献

Chronic kidney disease (CKD) is an important global public health problem due to its high prevalence and morbidity. Although the treatment of nephrology patients has changed considerably, ineffectiveness and side effects of medications represent a major issue. In an effort to elucidate the contribution of genetic variants located in several genes in the response to treatment of patients with CKD, we performed a systematic review and meta-analysis of all available pharmacogenetics studies. The association between genotype distribution and response to medication was examined using the dominant, recessive, and additive inheritance models. Subgroup analysis based on ethnicity was also performed. In total, 29 studies were included in the meta-analysis, which examined the association of 11 genes (16 polymorphisms) with the response to treatment regarding CKD. Among the 29 studies, 18 studies included patients with renal transplantation, 8 involved patients with nephrotic syndrome, and 3 studies included patients with lupus nephritis. The present meta-analysis provides strong evidence for the contribution of variants harbored in the ABCB1, IL-10, ITPA, MIF, and TNF genes that creates some genetic predisposition that reduces effectiveness or is associated with adverse events of medications used in CKD.  相似文献

Eleven natural pigments were selected for the production of environment-friendly cotton prints. Physicochemical properties pH, conductivity, viscosity of the printing pastes were measured in order to evaluate their stability and suitability for printing. Quality control standard tests and color measurements were carried out for all the prints with their fastness properties ranging from poor to very good in some cases.  相似文献

The complex-forming interaction between two direct dyes, CI Yellow 106 and CI Blue 78, and two nonionic surfactants, namely, polyoxyethylene nonyl phenyl ether (NP-12) and polyoxyethylene stearylamine (R-11), in aqueous solutions was studied. The estimation was made by measuring surface tension and critical micelle concentration (CMC) changes as a function of dye concentration. A decrease of NP-12 surface tension at low concentration and an increase of R-11 surface tension in the presence of both dyes were observed along with a significant decrease of NP-12 CMC values. A difference in the spectrophotometric absorbance of dye solutions in the presence of both surfactants, indicating a change in the environment of the dye chromophore, was also visible. These results confirm the formation of hydrophobic complexes of NP-12 and hydrophilic complexes of R-11 with both dyes.  相似文献
