Accumulated indirect evidence suggests nerve growth-promoting activities for acetylcholinesterase (AChE). To determine unequivocally whether such activities exist, whether they are related to the capacities of this enzyme to hydrolyze acetylcholine and enhance synapse development, and whether they are associated with alternative splicing variants of AChEmRNA, we used four recombinant human AChEDNA vectors. When Xenopus laevis embryos were injected with a vector expressing the synapse-characteristic human AChE-E6, which contains the exon 6-encoded C terminus, cultured spinal neurons expressing this enzyme grew threefold faster than co-cultured control neurons. Similar enhancement occurred in neurons expressing an insertion-inactivated human AChE-E6-IN protein, containing the same C terminus, and displaying indistinguishable immunochemical and electrophoretic migration properties from AChE-E6, but incapable of hydrolyzing acetylcholine. In contrast, the nonsynaptic secretory human AChE-I4, which contains the pseudointron 4-derived C terminus, did not affect neurite growth. Moreover, no growth promotion occurred in neurons expressing the catalytically active C-terminally truncated human AChE-E4, demonstrating a dominant role for the E6-derived C terminus in neurite extension. Also, AChE-E6 was the only active enzyme variant to be associated with Xenopus membranes. However, postsynaptic length measurements demonstrated that both AChE-E6 and AChE-E4 enhanced the development of neuromuscular junctions in vivo, unlike the catalytically inert AChE-E6-IN and the nonsynaptic AChE-I4. These findings demonstrate an evolutionarily conserved synaptogenic activity for AChE that depends on its hydrolytic capacity but not on its membrane association. Moreover, this synaptogenic effect differs from the growth-promoting activity of AChE, which is unrelated to its hydrolytic capacity yet depends on its exon 6-mediated membrane association.  相似文献

Thermal error in machine tools has been observed to be closely linked to the temperature of critical elements of the machine. It was found, in the experiments conducted herein, that there was a significant increase in the axis positioning error on account of an increase in the temperature of the machine elements due to continuous operation. However, it was also observed that the specific operating parameters of the test cycles carried out also significantly affected the positioning error. Different sets of operating parameters generated significantly different error values even though the temperature of the machine elements generated by those operating conditions was similar. As such, this observation forms a very important consideration in the development of a generic thermal error compensation system. It appears to indicate that such a system needs to be capable of evaluating the compensation depending upon the temperature values of the machine elements as well as account for the effect that different operating parameters induce upon the positioning error under the same thermal condition of the machine. This paper attempts to analyse the thermal behaviour of a three-axis vertical machining centre under the influence of various operating parameters and, through the experimental results obtained, point out and explain the effect of these parameters on the axis positioning error. This behaviour forms the basis of an improved modelling methodology that is presented separately in Part II of this paper.  相似文献

Model palm olein natural oil polyols (NOPs) with varying ratios of primary to secondary hydroxyls were synthesized, characterized, and evaluated in reaction kinetics study with isocyanate in formation of polyurethanes. Reaction rate constants and activation energies associated with primary and secondary hydroxyls of NOPs were quantified. The kinetic study in toluene shows that the NOP containing primary hydroxyls have three times higher reaction rate constants in noncatalyzed reaction with 4,4′‐diphenylmethane diisocyanate (4,4′‐MDI) compared to the model NOP containing only secondary hydroxyls, which is associated with higher activation energy of secondary hydroxyls. However, the difference in reaction rate constants of primary and secondary hydroxyls in NOPs diminished in the reactions catalyzed with dibutyltin dilaurate. Bulk polymerization reaction confirms the kinetics results in toluene, showing that the model NOP containing primary hydroxyls reached gel time at a faster rate. Evaluation of elastomers from bulk polymerization shows low degree of phase separation of hard and soft segments for elastomers based on the model NOPs. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. Dynamic Errors in Type 1 Contouring Systems   总被引:2,自引:0,他引:2  
The general nature of contouring errors experienced in type 1 servo systems as a result of command contours is described. Both transient and steady-state errors are discussed. A significant result illustrates that contouring error is essentially a result of mismatched gains in the servo drives and the contour. The relative significance of errors associated with a mismatch in time constants, modeling technique employed, and magnitude of velocity gains is discussed.  相似文献

This paper provides new lower and upper matrix bounds of the solution to the discrete algebraic Riccati equation. The lower bound always works if the solution exists. The upper bounds are presented in terms of the solution of the discrete Lyapunov equation and its upper matrix bound. The upper bounds are always calculated if the solution of the Lyapunov equation exists. A numerical example shows that the new bounds are tighter than previous results in many cases.  相似文献

In this paper, a new model-based Taylor series expansion error compensation (TSEEC) strategy is proposed to improve the contouring accuracy for computer numerically controlled (CNC) machines. In TSEEC, the contour error compensation problem is formulated as a Taylor series expansion problem, in which the value of the contour error is expanded around the reference points and the compensation components are calculated as the deviations from the reference points. Simulations show that, with perfect knowledge of the axial dynamics, zero contour errors can be achieved with TSEEC for both linear and circular contours. Due to modeling errors, external disturbances, and measurement noise, some modifications and experimentation need to be made to determine suitable parameters for implementation of the TSEEC scheme on a real machine. These measurements include a low-pass filter, a choice of a compensation target, and a compensation gain. Experimental results show the effectiveness of TSEEC in reducing contour errors and demonstrate the superiority of TSEEC over inverse feedforward compensation and cross-coupled control in improving the contouring accuracy.  相似文献

Intracellular localization of specific mRNAs is known to be a mechanism for targeting proteins to specific sites within the cell. Previous studies from this laboratory have demonstrated co-localization of mRNAs and proteins for a number of genes in absorptive enterocytes of fetal rat intestine. The present study was undertaken to examine in human enterocytes the intracellular localization patterns of mRNAs for the microvillous membrane proteins lactase-phlorizin hydrolase (LPH), sucrase-isomaltase (SI), and intestinal alkaline phosphatase (IAP), and the cytoskeletal protein beta-actin. In sections of human jejunum, mRNAs were localized by in situ hybridization using digoxigenin-labeled anti-sense RNA probes. Both LPH and SI mRNAs were localized to the apical region of villous enterocytes, whereas IAP and beta-actin mRNAs were detected both apically and basally relative to the nucleus. Therefore, in contrast to LPH, SI, and beta-actin mRNAs, which co-localize with their encoded proteins, that of IAP is present in the basal region of the cell where IAP protein has not directly been demonstrated to be present. Absorptive enterocytes from humans possess the mechanisms for intracellular mRNA localization, but not all mRNAs co-localize with their encoded proteins.  相似文献
