In the serum of a young normal male, never transfused, a cold cytotoxic IgM antibody was found reacting exclusively with A2 homozygous cells on a French panel, as shown by population and family studies. This antibody needs about two to five times more lymphocytes or platelets to be absorbed than an immune anti-A2 of the same titre. The antigen recognized by this antibody seems to be covered by the same molecule as HLA-A2, according to redistribution experiments.  相似文献

Increased ornithine decarboxylase (ODC) activity is associated with rapid cell proliferation in many cell types. The cellular effects of early weaning on intestinal development are not well established. To investigate whether ODC is involved in intestinal growth after early weaning, we precociously weaned suckling rats on postnatal d 15 and followed through d 21 (6 d after early weaning). Age-matched suckling pups served as controls. Rat pups were killed 1, 2, 3 and 6 d after early weaning and jejunal mucosa was assayed for ODC and sucrase activities, and protein and DNA contents. Jejunal cell proliferation was monitored by bromodeoxyuridine immunohistochemistry. Elevated jejunal ODC activity 1 d after early weaning was the earliest cellular event that was detected in the current study. ODC activity peaked at d 3 (about 15-fold greater than age-matched unweaned suckling controls). Sucrase activity was elevated at d 2 after weaning and peaked at d 3 (about 10-fold greater than controls). Greater bromodeoxyuridine immunostaining in early weaned rats occurred on d 3. Protein and DNA contents were greater in jejunal mucosa of early weaned rats at d 6. Serum corticosterone levels were elevated on d 1 and d 2 after early weaning compared to controls. To explore whether the intake of nonpurified diet played a role, we also compared the induction of jejunal ODC activity in early weaned pups and pups that were food-deprived for 1 d. ODC activity was not greater in the food-deprived group compared to suckling controls while the early weaned group had 6-fold greater activity 1 d after early weaning. Early weaning stimulates jejunal cell proliferation and differentiation. The temporal sequence of increased ODC activity followed by increases in other growth variables suggests that the induction of ODC activity may act as an early marker of intestinal growth during early weaning.  相似文献

Polymethyl methacrylate (PMMA) is a useful material for dosimetry phantoms in many ways including approximate tissue equivalence, stability, accessibility and ease of use. However, recent studies indicate that PMMA may have some unanticipated variation in backscatter from one phantom to another. While the reasons behind the variations have not been identified, it has been demonstrated that the backscatter from one phantom to another may vary by as much as 15%, resulting in a dosemeter response variation of as much as 5%. This unexpected contribution to uncertainty in delivered dose to a dosemeter may be quite large compared to the normally estimated uncertainty, potentially causing problems with calibration and performance testing. This paper includes data supporting the differences in backscatter among phantoms, and results from tests on the phantoms performed in an effort to identify possible causes.  相似文献

Extracellular vesicles (EVs) are reminiscent of their cell of origin and thus represent a valuable source of biomarkers. However, for EVs to be used as biomarkers in clinical practice, simple, comparable, and reproducible analytical methods must be applied. Although progress is being made in EV separation methods for human biofluids, the implementation of EV assays for clinical diagnosis and common guidelines are still lacking. We conducted a comprehensive analysis of established EV separation techniques from human serum and plasma, including ultracentrifugation and size exclusion chromatography (SEC), followed by concentration using (a) ultracentrifugation, (b) ultrafiltration, or (c) precipitation, and immunoaffinity isolation. We analyzed the size, number, protein, and miRNA content of the obtained EVs and assessed the functional delivery of EV cargo. Our results demonstrate that all methods led to an adequate yield of small EVs. While no significant difference in miRNA content was observed for the different separation methods, ultracentrifugation was best for subsequent flow cytometry analysis. Immunoaffinity isolation is not suitable for subsequent protein analyses. SEC + ultracentrifugation showed the best functional delivery of EV cargo. In summary, combining SEC with ultracentrifugation gives the highest yield of pure and functional EVs and allows reliable analysis of both protein and miRNA contents. We propose this combination as the preferred EV isolation method for biomarker studies from human serum or plasma.  相似文献

Timing phase estimation (TPE) plays the key role in feedforward (FF) symbol timing.For reasons of performance often data-aided (DA) methods are preferred. But frequently, they turn out to be critical with respect to theimplementation and the spectrum efficiency (due to the required overhead).This paper presents three DA TPE methods for quadrature pulse amplitude modulation (PAM). In spite of their very low complexity, these methods closelyapproach the theoretical limit for timing estimation with respect to the estimation variance, even at low SNR. This enables power efficient transmission.Further, they employ a CAZAC (constant amplitude, zero auto-correlation)sequence as training-sequence (TS), or a sequence with similar correlationproperties. Since such sequences are suited for almost all DA receiver tasks, a high spectrum efficiency can be obtained by the use of a single TS. A generalization of the proposed methods for DA TPE withrespect to the choice of the TS is also shown. The presented methods can be applied to noncoherent receivers. They are suited for high data-rate applications, since they can work with an oversampling factor of 2.  相似文献

RNA helicases represent a family of enzymes that unwind double-stranded(ds) RNA in a nucleoside triphosphate (NTP)-dependent fashionand which are required in all aspects of cellular RNA metabolismand processing. The hepatitis C virus (HCV) non-structural 3(NS3) protein possesses a serine protease activity in the N-terminalone-third, whereas RNA-stimulated NTPase and helicase activitiesreside in the C-terminal portion of the 631 amino acid residuebifunctional enzyme. The HCV NS3 RNA helicase is of key importancein the life cycle of HCV, which makes it a target for the developmentof therapeutics. However, neither the precise mechanism northe substrate structure has been defined for this enzyme. Fornuclear magnetic resonance (NMR)-based drug discovery methodsand for mechanistic studies we engineered, prepared and characterizedvarious truncated constructs of the 451-residue HCV NS3 RNAhelicase. Our goal was to produce smaller fragments of the enzyme,which would be amenable to solution NMR techniques while retainingtheir native NTP and/or nucleic acid binding sites. Solutionconditions were optimized to obtain high-quality heteronuclearNMR spectra of nitrogen-15 isotope-labeled constructs, whichare typical of well-folded monomeric proteins. Moreover, NMRbinding studies and functional data directly support the correctfolding of these fragments.  相似文献

A general formulation for the scattering from multiple waveguide discontinuities is presented. It is based on the modal analysis technique in which the corresponding fields are expanded in terms of vector eigenfunctions and the boundary conditions are imposed at each junction. Analytic expressions for the global scattering matrices are derived using a recurrence procedure that requires substantially less computation than the traditional cascading techniques. The matrix equations are truncated in a manner that satisfies the conditions for a good overall convergence, which is illustrated for step discontinuities in circular waveguides. Numerical results are presented for a thick iris in a circular waveguide and for iris-matched dielectric window designs  相似文献
