Use of Zebrafish Embryo Assay to Evaluate Toxicity and Safety of Bioreactor-Grown Exopolysaccharides and Endopolysaccharides from European Ganoderma applanatum Mycelium for Future Aquaculture Applications

Natural mycelial exopolysaccharide (EPS) and endopolysaccharide (ENS) extracted from bioreactor-cultivated European Ganoderma applanatum mushrooms are of potential high commercial value for both food and adjacent biopharmaceutical industries. In order to evaluate their potential toxicity for aquaculture application, both EPS (0.01–10 mg/mL) and ENS (0.01–10 mg/mL) extracts were tested for Zebrafish Embryo Toxicity (ZFET); early development effects on Zebrafish Embryos (ZE) were also analyzed between 24 and 120 h post-fertilization (HPF). Both EPS and ENS are considered non-toxic with LC₅₀ of 1.41 mg/mL and 0.87 mg/mL respectively. Both EPS and ENS did not delay hatching and teratogenic defect towards ZE with <1.0 mg/mL, respectively. No significant changes in the ZE heart rate were detected following treatment with the two compounds tested (EPS: 0.01–10 mg/mL: 176.44 ± 0.77 beats/min and ENS: 0.01–10 mg/mL: 148.44 ± 17.75 beats/min) compared to normal ZE (120–180 beats/min). These initial findings support future pre-clinical trials in adult fish models with view to safely using EPS and ENS as potential feed supplements for supplements for development of the aquaculture industry.     

THE FLOW PRODUCED IN PARTICLE-SUSPENSION BY AN INFINITE ROTATING DISK:SERIES SOLUTION

To the flow produced in particle-suspension by a rotating disk of infinite extent,the asymptotic behavior at infinity is analyzed.A corresponding series solution is presented.It isshown that this approach yields widely valid solutions of high accuracy for all the cases in whichthat suction or weak injection at the disk surface exists.For the cases that strong injection or par-ticle-free band exists,the series method fails.

Reliability‐based estimation of the survival of Listeria monocytogenes in chorizos

Reliability analysis is especially important when critical decisions are to be made involving potentially severe adverse consequences such as foodborne illness. Owing to uncertainty associated with the parameters controlling survival of Listeria monocytogenes in chorizo (a Mexican‐style sausage), the time needed to reduce the count by a certain number (n) of logs (t_nD) is probabilistic. In this paper the first‐order second‐moment (FOSM) method based on Taylor series expansion is used to derive the expected value and standard deviation of t_nD as function of the operating conditions (random variables) affecting survival, namely initial water activity (a_w0) of the sausage batter, storage temperature (T) and airflow velocity (F), along with their uncertainties characterised by their means and coefficients of variation. For any given n the derived t_nD probability distribution enables one to determine an estimate of t_nD for any desired level of reliability or confidence level, such as 50% (median value), 95%, 99%, etc. Among the conclusions drawn were: (i) the variability associated with T and F has a minor effect on estimating uncertainty in t_nD, whereas the reliability of t_nD estimation is greatly influenced by the uncertainty in a_w0; and (ii) the uncertainty in a_w0 has the greatest impact when a_w0 of the sausage formulation exceeds 0.90. The approach used and discussed in this paper can be applied to any survival/inactivation study to incorporate the effect of uncertainty in the various extrinsic and intrinsic parameters on the survival kinetics of the pathogen in a food system under evaluation. Thermal decomposition behavior of γ‐irradiated poly(vinyl acetate)/poly(methyl methacrylate) miscible blends

Miscible polymer blends based on various ratios of poly(vinyl acetate) (PVAc) and poly(methyl methacrylate) (PMMA) were prepared in film form by the solution casting technique using benzene as a common solvent. The thermal decomposition behavior of these blends and their individual homopolymers before and after γ‐irradiation at various doses (50–250 kGy) was investigated. The thermogravimetric analysis technique was utilized to determine the temperatures at which the maximum value of the rate of reaction (T_max) occurs and the kinetic parameters of the thermal decomposition. The rate of reaction curves of the individual homopolymers or their blends before or after γ‐ irradiation displayed similar trends in which the T_max corresponding to all polymers was found to exist in the same position but with different values. These findings and the visual observations of the blend solutions and the transparency of the films gave support to the complete miscibility of these blends. Three transitions were observed along the reaction rate versus temperature curves; the first was around 100–200°C with no defined T_max, which may arise from the evaporation of the solvent. The second T_max was in the 340–380°C range, which depended on the polymer blend and the γ‐irradiation condition. A third transition was seen in the rate of reaction curves only for pure PVAc and its blends with PMMA with ratios up to 50%, regardless of γ‐ irradiation. We concluded that γ‐irradiation improved the thermal stability of PVAc/PMMA blends, even though the PMMA polymer was degradable by γ irradiation. © 2005 Wiley Periodicals, Inc. 原矿含泥量大造成设备的堵塞。根据现场生产实际改用粉碎后的矿粒直接给入扇形孔条筛，筛下可调刮板使细粒均匀分布在皮带下面，粗粒复盖其上。物料到达皮带首轮处，使其呈抛掷状态，然后利用分离挡板除去细粒。首轮的直径和转速是按粒度分离的决定因素。分离细粒级后使中碎机生产能力增大，降低能耗，减少磨损，提高了设备运转率。
