Two novel peptides, named PACAP (pituitary adenylate cyclase activating polypeptide) containing 38 (PACAP38) and 27 residues (PACAP27) were recently isolated from ovine hypothalami. In order to investigate the pituitary cell type(s) that bear a receptor for PACAP, PACAP38 was biotinylated and used for cytochemical examination of binding. The cells were also identified by immunocytochemical methods using the antisera against each of the rat anterior pituitary hormones or an antiserum against S-100 protein, a marker for pituitary folliculo-stellate (FS) cells. Biotinylated PACAP38 (biot-PACAP) exhibited adenylate cyclase stimulating activity (ACSA) comparable to PACAP38 in rat pituitary cell cultures, and displaced the bound 125I-PACAP27 to the rat pituitary membrane preparation to the same extent as PACAP38. After 2-4 days of culture, dispersed rat pituitary cells were incubated with varying concentrations of biot-PACAP at room temperature or 4 degrees C. The bound biot-PACAP38 was visualized by avidin-biotin-peroxidase complex (ABC) method with nickel intensification. Biot-PACAP-positive and pituitary hormone or S-100-positive cells were counted. More than 90% of S-100-positive cells bound biot-PACAP38. A considerable number of GH and PRL cells and a lesser number of ACTH cells also bound biot-PACAP38, whereas only a few identified LH, FSH, or TSH cells bound biot-PACAP38. These results suggest that FS cells are a major target cell type for PACAP. A recent study from our laboratory demonstrated that PACAP stimulated the release of interleukin (IL)-6 in rat pituitary cell cultures. FS cells are known to produce IL-6.  相似文献

Numerical investigation of flow hydrodynamics in a square cross‐sectioned bubble column was conducted in a transient Euler‐Euler environment by applying the simulation tool Ansys CFX 14.0. The influence of the drag coefficient (CD) was investigated and the results were also compared with drag force models. Furthermore, three different lift force models and a defined lift coefficient were studied. All results were compared with the available experimental data. All simulations were carried out for a single‐hole sparger with given aspect ratio (H/D) and superficial gas velocity.  相似文献

Mesenchymal stem cells (MSCs) are self-renewable, multipotent stem cells that regulate the phenotype and function of all immune cells that participate in anti-tumor immunity. MSCs modulate the antigen-presenting properties of dendritic cells, affect chemokine and cytokine production in macrophages and CD4+ T helper cells, alter the cytotoxicity of CD8+ T lymphocytes and natural killer cells and regulate the generation and expansion of myeloid-derived suppressor cells and T regulatory cells. As plastic cells, MSCs adopt their phenotype and function according to the cytokine profile of neighboring tumor-infiltrated immune cells. Depending on the tumor microenvironment to which they are exposed, MSCs may obtain pro- and anti-tumorigenic phenotypes and may enhance or suppress tumor growth. Due to their tumor-homing properties, MSCs and their exosomes may be used as vehicles for delivering anti-tumorigenic agents in tumor cells, attenuating their viability and invasive characteristics. Since many factors affect the phenotype and function of MSCs in the tumor microenvironment, a better understanding of signaling pathways that regulate the cross-talk between MSCs, immune cells and tumor cells will pave the way for the clinical use of MSCs in cancer immunotherapy. In this review article, we summarize current knowledge on the molecular and cellular mechanisms that are responsible for the MSC-dependent modulation of the anti-tumor immune response and we discuss different insights regarding therapeutic potential of MSCs in the therapy of malignant diseases.  相似文献

Colorectal carcinoma is acknowledged as the second leading cause of total cancer-related death in the European Region. The majority of deaths related to colorectal carcinoma are connected with liver metastatic disease. Approximately, in 25% of all patients, liver metastatic disease is diagnosed at the same time as the primary diagnosis, while up to a quarter of others would develop liver metastases in the course of the illness. In this study, we developed reaction-diffusion model and analyzed the effect of drug therapy on liver metastatic disease for a specific patient. Tumor volumes in specific time points were obtained using CT scan images. The nonlinear function for cell proliferation rate as well as data about clinically applied drug therapy was included in the model. Fitting procedure was used for parameter estimation. Good agreement of numerical and experimental results shows the feasibility and efficacy of the proposed system.  相似文献

We derive upper bounds on the transport capacity of wireless networks. The bounds obtained are solely dependent on the geographic locations and power constraints of the nodes. As a result of this derivation, we are able to conclude the optimality, in the sense of scaling of transport capacity with the number of nodes, of a multihop communication strategy for a class of network topologies.  相似文献

This study aimed to examine the effects of diallyl trisulfide (DATS), the most potent polysulfide derived from garlic, on metabolic syndrome and myocardial function in rats with metabolic syndrome (MetS). For that purpose, we used 36 male Wistar albino rats divided into control rats, rats with MetS and MetS rats treated with 40 mg/kg of DATS every second day for 3 weeks. In the first part, we studied the impact of DATS on MetS control and found that DATS significantly raised H2S, decreased homocysteine and glucose levels and enhanced lipid and antioxidative, while reducing prooxidative parameters. Additionally, this polysulfide improved cardiac function. In the second part, we investigated the impact of DATS on ex vivo induced ischemia/reperfusion (I/R) heart injury and found that DATS consumption significantly improved cardiodynamic parameters and prevented oxidative and histo-architectural variation in the heart. In addition, DATS significantly increased relative gene expression of eNOS, SOD-1 and -2, Bcl-2 and decreased relative gene expression of NF-κB, IL-17A, Bax, and caspases-3 and -9. Taken together, the data show that DATS can effectively mitigate MetS and have protective effects against ex vivo induced myocardial I/R injury in MetS rat.  相似文献

Electrophysiological investigations were carried out both in patients with carpal tunnel syndrome (CTS) and in healthy individuals. The evoked potentials were examined during stimulation of sensitive branches of n. medianus and of other nerves. The most sensitive and important for diagnosis appeared to be the electrophysiological test of alteration of responsive reaction to the stimulation of the sensitive branch of n. medianus for 4 finger and the considerable difference of evoked reactions on stimulation of skin palm branch and branch of the 1-st finger of n. medianus. The diagnostic importance of these indices corresponds well to anatomical and pathophysiological characteristics of CTS.  相似文献

The optimal microphone array, in the sense of minimum mean square errors (MMSE), includes two processing blocks: the minimum variance distortionless response (MVDR) beamformer and the single-channel Wiener filter, which acts as post-filter. In this paper, we propose a new post-filter algorithm based on assumptions that both the noise power attenuation factor (NPAF) and signal power attenuation factor (SPAF) are time invariant in the reverberant room. The algorithm recursively estimates both factors from available measurements and uses them in estimation of the post-filter parameters. Additionally, to overcome the problem of the poor performance of the MVDR beamformer in reverberant conditions, we propose the usage of the two-step (TS) MVDR algorithm. This algorithm improves the robustness of the beamformer and its ability to suppress the interferences using an estimate of the desired speaker transfer function. Although TS MVDR beamformer and proposed post-filter can work separately, or combined with other algorithms, the best performance is obtained when they work together. The performance of the proposed combination of new post-filter algorithm and TS MVDR beamformer is tested in a simulated reverberant room and compared with similar algorithms, which gave rather good results.  相似文献

In this paper, a method of floating ball wear rate identification, using two machine-learning techniques Support Vector Machine (SVM) and Improved Support Vector Machine (ISVM) are proposed. Both models are used to relate the wear rate and technological parameters of the wear resistant drip moulding using different kernel functions. The models for determining the wear rate of white iron casting with low chromium content (flotation balls), was trained and tested by using the existing exploitation data from the Bor Flotation Plant, Serbia. In order to select the best model parameters the statistical indicators for both models are presented. Results show that the ERBF (SVM) and ERBF+POLY (ISVM) achieved the best classification accuracy compare to other kernels used: the absolute mean error of ERB (SVM) is 5.85%, while the error of ERBF+POLY (ISVM) is 6.67%. The tuned ISVM model with mixture of kernels is able to accurately predict the wear rate and can be used to define the optimum chromium content in liquid metal alloys for the casting of flotation balls.  相似文献
