Microscale analysis of glycosphingolipids by TLC blotting/secondary ion mass spectrometry: a novel blood group A-active glycosphingolipid and changes in glycosphingolipid expression in rat mammary tumour cells with different metastatic potentials

The glycosphingolipid compositions of rat mammary tumour cell lines with different metastatic potentials for the lung [a parental tumour cell line (MTC) and its subclones MTLn2 (a non metastatic subclone) and MTLn3 (a subclone with high metastatic potential to the lung)] were studied using a newly developed TLC blotting/secondary ion mass spectrometry system and crude glycosphingolipids obtained from 0.5-1 x 10(7) cells of each cell line. GM3 and GM2 were the major components of the MTC cell line, but they were very minor components in the MTLn2 and MTLn3 cell lines, GDla being the major ganglioside HexNAc-fucosyl-GMla was found in the MTLn2 cells by the TLC blotting/SIMS method, and the terminal sugar linkage was shown to be a blood group A-type structure by immunostaining. These findings suggest that the ganglioside is a novel type of blood group A-active ganglioside, GalNAc alpha 1-3(fuc alpha 1 -2)GMla. No blood group A-active lipid was present in MTLn3 cells, whereas Hex-GMla and neutral glycosphingolipids with more than 5 sugar residues were.     

Differential regulation by hematopoietic growth factors of apoptosis and mitosis in acute myeloblastic leukemia cells

We evaluated the effects of various hematopoietic growth factors (HGFs) on the prevention of apoptosis in blasts from 19 patients with acute myeloblastic leukemia (AML) by assessing DNA ladder formation. After incubation without HGF, apoptosis was noted in all but two patients. HGFs prevented, did not affect, or enhanced apoptosis in 39 (60%), 14 (22%), or 12 (18%) of 65 suspension cultures, respectively. HGFs that prevented apoptosis also stimulated and/or synergized blast colony formation in 35 of 39 corresponding methylcellulose cultures. HGFs that alone stimulated colony formation also prevented apoptosis in all but two of 28 corresponding suspension cultures. In contrast, HGFs that did not prevent apoptosis also failed to stimulate growth in 17 of 26 corresponding methylcellulose cultures. HGFs that enhanced apoptosis alone never stimulated colony formation. After incubation, we noted enhanced c-fos and cjun genes as well as induction of p21 protein. An appropriate dose of HGF elevated c-fos, reduced c-jun and p21, induced G1/S transition, and inhibited apoptosis. In two patients, apoptosis was not induced after incubation. Cells not treated with HGF expressed no c-fos, c-jun, or c-myc, and remained in G0/G1. Taken together, our results support the conclusion that not only c-fos, cjun, and c-myc, but also p53 and p21 are required for blast apoptosis. HGF differentially prevents apoptosis and induces mitosis, and both events seem to be integral to the self-renewal of AML clonogenic cells.     

Electron-microscopic procedure for acrylic rubber

The electron-microscopic visualization of acrylic rubber dispersed in a heterogeneous structural resin composition consisting of a rubber-modified two-phase plastic comprising essentially a butyl acrylate rubber phase and an acrylonitrile–styrene copolymer phase, respectively, has been accomplished. This procedure consists of the following: The molded resin specimen is treated with hydrazine hydrate solution to produce the acrylic acid hydrazides. Allow the treated specimen to soak in osmium tetroxide solution. The acrylic rubber may be indirectly fixed and stained. Some micrographs of ultrathin sections of two or three resin compositions, cut by an ultramicrotome, are presented.     

Giant neurofibroma of the occipital scalp associated with lambda defect-case report (author's transl)

Calvarial bone defect associated with a hugh neurofibroma in the region of the lambdoid suture was reported in a 42-year-old man with v. Recklinghausen's neurofibromatosis. An enlarging occipital tumor was first noted at age 4 similar to or approximately 5, and an operation was performed at age 12. Lambda defect was noted at that time, but the operative diagnosis was, apparently, "meningocele". Available literature was reviewed, and the significance of calvarial bone defect in the region of the lambdoid suture as a manifestation of v. Recklinghausen's disease was stressed. Lambda defect witn no sclerosing margin, particularly if associated with hypoplasia of the ipsilateral mastoid cells, has been known to represent the primary nature of the bone defect due to mesodermal dysplasia. In the present case, plain radiographs of the skull revealed abnormal sclerotic change around the bone defect. It is suggested that such an osteoblastic change may well indicate the secondary nature of the bone defect due to the presence of overlying neurogenic tumor.

Experimental Studies on the Self-Shielding Effect in Fissile Fuel Breeding Measurement in Thorium Oxide Pellets Irradiated with 14 MeV Neutrons

The 14 MeV neutrons produced in the D-T fusion reactions have the potential of breeding Uranium-233 fissile fuel from fertile material Thorium-232. In order to estimate the amount of U-233 produced, experiments are carried out by irradiating thorium dioxide pellets with neutrons produced from a 14 MeV neutron generator. The objective of the present work is to measure the reaction rates of 232 Th + 1 n → 233 Th → 233 Pa → 233 U in different pellet thicknesses to study the self-shielding effects and adopt a procedure for correction. An appropriate assembly consisting of high-density polyethylene is designed and fabricated to slow down the high-energy neutrons, in which Thorium pellets are irradiated. The amount of fissile fuel ( 233 U) produced is estimated by measuring the 312 keV gammas emitted by Protactinium-233 (half-life of 27 days). A calibrated High Purity Germanium (HPGe) detector is used to measure the gamma ray spectrum. The amount of 233 U produced by Th 232 (n, γ) is calculated using MCNP code. The self-shielding effect is evaluated by calculating the reaction rates for different foil thickness. MCNP calculation results are compared with the experimental values and appropriate correction factors are estimated for self-shielding of neutrons and absorption of gamma rays.

Professor Vir Handa's abiding interest in low‐cost housing for the lower‐income earners is demonstrated in his paper from the University of Waterloo, Ontario. He draws experience from time spent at the Building Research Institute, Roorkee, under the United Nations TOKTEN Programme and illustrates his paper with pictures from Trinidad, where he is an External Examiner at the University of the West Indies. He argues that although shelter communities are being built without any government help and without any awareness of any codes and standards, the job of the expert should be to determine new affordable code requirements.
