Endurance training leads to an increase in the content of individual mitochondrial hemeproteins in skeletal muscle. To deduce the control mechanisms involved, cytochrome oxidase (COX) activity was compared with 1) the content of COX subunits III and IV and 2) 5-aminolevulinate synthase (ALAS) activity and mRNA content. In the plantaris muscle of female rats run daily for up to 28 days, ALAS activity was elevated 100% (P < 0.01) after 3 days and remained 150 and 125% higher (P < 0.001) after 7 and 28 days of running, respectively, than control. COX activity was also increased, but not until day 7 (40%; P < 0.05), and reached a maximal value 80% higher than control (P < 0.001) in the 28-day group. Compared with control, the content of COX subunit III and IV and ALAS mRNAs was not significantly changed by the training. The increased activities of COX and ALAS appear to be regulated by translational or posttranslational steps in the protein expression pathway. The induction of ALAS before COX suggests that the increased activity of COX may require increased synthesis of heme.  相似文献

We have found a method to activate electrically smaller nerve fibers without activating larger fibers in the same nerve trunk. The method takes advantage of the fact that action potentials are blocked with less membrane hyperpolarization in larger fibers than in smaller fibers. In our nerve stimulation system, quasitrapezoidal-shaped current pulses were delivered through a tripolar cuff electrode to effect differential block by membrane hyperpolarization. The quasitrapezoidal-shaped pulses with a square leading edge, a 350 microsecond(s) plateau, and an exponential trailing phase ensured the block of propagating action potentials and prevented the occurrence of anodal break excitation. The tripolar cuff electrode design restricted current flow inside the cuff and thus eliminated the undesired nerve stimulation due to a "virtual cathode." Experiments were performed on 13 cats. The cuff electrode was placed on the medial gastrocnemius nerve. Both compound and single fiber action potentials were recorded from L7 ventral root filaments. The results demonstrated that larger alpha motor axons could be blocked at lower current levels than smaller alpha motor axons, and that all alpha fibers could be blocked at lower current levels than gamma fibers. A statistical analysis indicated that the blocking threshold was correlated with the axonal conduction velocity or fiber diameter. This method could be used in physiological experiments and neural prostheses to achieve a small-to-large recruitment order in motor or sensory systems.  相似文献

The introduction of sit-stand worktables was examined in this study. Purchasers at four companies were interviewed regarding preparations prior to purchase, follow-up after installation, and if expectations had been fulfilled. Personnel at the four companies answered a questionnaire examining use of the worktables' sit-stand function, and the influence of some variables on this. Utilisation of the sit-stand function was less than what could be expected to relieve static musculoskeletal loading during VDU work. Pain experienced during the past year, and education on the use of the worktable gave rise to modest increases in utilisation. Investment in sit-stand worktables was satisfactory to purchasers, in terms of providing flexible furniture to suit modern organisational dynamics. Users were, in general, positive to the worktables, but showed poor compliance in using them. User and purchaser satisfaction with regard to improving individual health ergonomics was questionable in respect of the low level of usage.  相似文献

Since the handling characteristics of bicycles can affect their safety, the present experiment evaluated the manoevrability of three bicycles which differed only by handlebar configurations: racing (Maes Bend), standard, and high rise. The manoeuvrability of each bicycle was measured as subjects performed six tasks: circle, lane change, figure-eight, straight lane tracking, cornering, and slalom. Subjects were matched by riding experience and grouped by their familiarity with either race or standard bicycle. Analysis of variance showed that no bicycle times bicycle-familiarity interaction effects were significant in any one of the analyses.

The performance observed on the bicycles with high-rise and standard handlebar configurations indicated they were not significantly different from each other. On the circle, figure-eight, and slalom tasks, performance with both the high-rise and standard handlebars was significantly better than with the race handlebars. The high-rise showed a slight performance edge on tasks requiring the greatest amount of manoeuvrability, while the standard handlebars offered more control at slower speeds and on tasks requiring stability in tracking.  相似文献

There are very few studies that have investigated directly the effect of an oxide film on tin whisker growth, since the ‘cracked oxide theory’ was proposed by Tu in 1994. The current study has investigated the effect of both a molybdate conversion coating and a tungstate conversion coating on tin whisker growth from Sn–Cu electrodeposits on Cu, and compared it with that from an electrochemically formed oxide produced from a potassium bicarbonate-potassium carbonate electrolyte. X-ray photoelectron spectroscopy (XPS) has been used to investigate the effect of both immersion time and applied potential on the thickness and composition of the oxide film. The XPS studies show that the oxide film formed using either of the conversion coating baths is significantly thicker than that produced from the potassium bicarbonate-potassium carbonate bath. Initial observations suggest that both the tungstate-based conversion coatings and the molybdate-based conversion coatings significantly reduce whisker growth by over 80% for all conversion coating systems compared with a native air-formed oxide and provide improved mitigation compared with the electrochemically formed oxides previously investigated.  相似文献

We assessed the value of positron emission tomography (PET) with 2-[18F]fluoro-2-deoxy-D-glucose (FDG) and 16alpha-[18F]fluoro-17beta-estradiol (FES) in women with breast cancer for predicting response to systemic therapy. Results of FES-PET were correlated with estrogen receptor (ER) status. Forty-three women with locally advanced or metastatic breast cancer underwent FDG-PET and FES-PET prior to institution of systemic therapy. All patients had measurable disease and had tumors submitted for ER determination. Cancers were considered functionally hormone sensitive if the standardized uptake value of the lesion on FES-PET was >/=1.0 (FES+) and hormone resistant if the standardized uptake value was <1.0 (FES-). Information obtained by FES-PET was compared with the results of ER assays. The tumor response to chemotherapy and hormonal therapy was correlated with intensity of uptake by both FDG-PET and FES-PET. The ER status of the breast cancers was negative (ER-) in 20 patients, positive (ER+) in 21 patients, and unknown in 2 patients. All 20 of the ER- tumors were also FES-. However, of the 21 ER+ tumors, 16 were FES+ and 5 were FES-. Thirty patients were treated initially with chemotherapy, and 21 (70%) demonstrated objective responses. We were unable to correlate the response to chemotherapy with information obtained by FDG-PET or FES-PET. Thirteen patients were treated with hormone therapy, and 8 (61%) responded to that therapy. Only 1 of the 5 patients whose tumors were ER+ but FES- received hormone therapy, and this treatment resulted in disease stabilization only. Multiple sites of disease were assessed by FES-PET in 17 patients with metastatic breast cancer. Functional hormone sensitivity, defined by FES-PET, was concordant with multiple lesions in 13 (76%). Ten patients with locally advanced breast cancer developed recurrent disease. The initial site of recurrence was the breast in 5 patients. Of the 5 patients with systemic recurrence, 4 had disease detected at the site of recurrence on the pretreatment FDG-PET study but not detected on pretreatment computed tomography. In our experience, FDG-PET imaging is more sensitive than conventional imaging methods, including computed tomography, in staging women with breast cancer. When compared with the in vitro assay of ER status, FES-PET has an apparent sensitivity of 76% and specificity of 100%. Our finding of a subset of patients who have tumors that are ER+ and FES- suggests that the functional assessment of hormone sensitivity by PET imaging can identify patients with ER+ disease whose tumors are likely to be hormone refractory.  相似文献

Biphasic electrode systems are studied for the case of the oxidation of the water-insoluble liquid N,N-didodecyl-N',N'-diethylphenylenediamine (DDPD) neat and dissolved in bis(2-ethylhexyl) phosphate (HDOP) and immersed in aqueous electrolyte media. The oxidation process in the absence of HDOP is accompanied by transfer of the anion (perchlorate or phosphate) from the water into the organic phase. However, in the presence of HDOP, oxidation is accompanied by proton exchange instead. This electrochemically driven proton exchange process occurs over a wide pH range. Organic microdroplet deposits of DDPD in HDOP at basal plane pyrolytic graphite electrodes are studied by voltammetric techniques and compared in their behavior to organic microphase deposits in mesoporous TiO2 thin films. The mesoporous TiO2 thin film acts as a host for the organic liquid and provides an alternative biphasic electrode system compared to the random microdroplet/graphite system. Two types of mesoporous TiO2 thin-film electrodes, (i) a 300-400-nm film on ITO and (ii) a 300-400-nm film on ITO sputter-coated with a 20-nm porous gold layer, are investigated.  相似文献
