The GGP1/GAS1/CWH52 gene of Saccharomyces cerevisiae encodes a major exocellular 115 kDa glycoprotein (gp115) anchored to the plasma membrane through a glycosylphosphatidylinositol (GPI). The function of gp115 is still unknown but the analysis of null mutants suggests a possible role in the control of morphogenesis. PHR1 gene isolated from Candida alibicans is homologous to the GGP1 gene. In this report we have analysed the ability of PHR1 to complement a ggp1Δ mutation in S. cerevisiae. The expression of PHR1 controlled by its natural promoter or by the GGP1 promoter has been studied. In both cases we have observed a complete complementation of the mutant phenotype. Moreover, immunological analysis has revealed that PHR1 in budding yeast gives rise to a 75–80 kDa protein anchored to the membrane through a GPI, indicating that the signal for GPI attachment present in the C. albicans gene product is functional in S. cerevisiae. 相似文献

We have previously demonstrated that UCN-01, a potent protein kinase inhibitor currently in phase I clinical trials for cancer treatment, abrogates G2 arrest following DNA damage. Here we used murine FT210 cells, which contain temperature-sensitive Cdc2 mutations, to determine if UCN-01 abrogates G2 arrest through a Cdc2-dependent pathway. We report that UCN-01 cannot induce mitosis in DNA-damaged FT210 cells at the non-permissive temperature for Cdc2 function. Failure to abrogate G2 arrest was not due to UCN-01-inactivation at the elevated temperature because parental FM3A cells, which have wild-type Cdc2, were sensitive to UCN-01-induced G2 checkpoint abrogation. Having established that UCN-01 acted through Cdc2, we next assessed UCN-01's effect on the Cdc2-inhibitory kinase, Wee1Hu, and the Cdc2-activating phosphatase, Cdc25C. We found that Wee1Hu was indeed inactivated in UCN-01-treated cells, possibly just prior to Cdc2 activation and entry of DNA-damaged cells into mitosis. This inhibition appeared, however, to be a consequence of a further upstream action since in vitro studies revealed purified Wee1Hu was relatively resistant to UCN-01-inhibition. Consistent with such an upstream action, UCN-01 also promoted the hyperphosphorylation (activation) of Cdc25C in DNA-damaged cells. Our results suggest that UCN-01 abrogates G2 checkpoint function through inhibition of a kinase residing upstream of Cdc2, Wee1Hu, and Cdc25C, and that changes observed in these mitotic regulators are downstream consequences of UCN-01's actions. 相似文献

Reverberation chambers are attractive electromagnetic compatibility test facilities, both economically and technically. Careful design and analysis of these facilities are important, if the results obtained are to be treated with a high level of confidence. Numerical modeling is an important part of the process of reverberation chamber design and analysis. Hence, it is important that the modeling techniques to be used are appropriately validated. Much of the published work to date takes either a statistical or a deterministic view of validation. This paper provides validation evidence for a low-resolution transmission line matrix (TLM) model of a reverberation chamber in a manner approximating the way in which the chamber is used, i.e., validating based on the effects of a simple device under test. A variety of statistical and heuristic approaches have been used to quantify the level of agreement, intending to set the likely lower bound for the quality of comparisons between simulations and measurements. While not drawing any "universal" conclusions about the veracity of the TLM technique, the paper concludes that a relatively simple model of a reverberation chamber provides a useful analysis of the chamber with close comparisons between modeled and measured data. 相似文献

A methodology is presented for the synthesis of the passive equivalent circuit of via holes in multilayer printed circuit boards. The paper describes the network synthesis starting from the extraction of the poles and residues from the driving point transfer functions for two port networks. The via hole is partitioned into elementary structures assumed not electromagnetically coupled and the scattering parameters for each one of them are evaluated by using a numerical approach suitably validated by comparison with results computed by other independent numerical methods. The equivalent circuit of the complete via hole is given by the cascading of the circuits of the elementary structures. An image watermarking technique based on the concept of JPEG2000 algorithm is proposed. Biorthogonal wavelet 9/7 transform is used to provide a set of coefficients suitable for watermark embedding. The statistical properties of different subbands are analyzed in order to choose the number of decomposition levels and position of subbands, which will assure the best compromise between the watermark transparency and robustness. The JPEG2000 quantization is applied to avoid insignificant wavelet coefficients, while the remaining ones are used for watermarking. The optimal and blind watermark detection is based on the nonlinear score function and appropriate model of coefficients distribution. The performance of the proposed procedure is tested on examples with various images, showing robustness under different attacks, while maintaining high image quality. 相似文献
