The demand for biodegradable sustained release carriers with minimally invasive and less frequent administration properties for therapeutic proteins and peptides has increased over the years. The purpose of achieving sustained minimally invasive and site-specific delivery of macromolecules led to the investigation of a photo-responsive delivery system. This research explored a biodegradable prolamin, zein, modified with an azo dye (DHAB) to synthesize photo-responsive azoprolamin (AZP) nanospheres loaded with Immunoglobulin G (IgG). AZP nanospheres were incorporated in a hyaluronic acid (HA) hydrogel to develop a novel injectable photo-responsive nanosystem (HA-NSP) as a potential approach for the treatment of chorio-retinal diseases such as age-related macular degeneration (AMD) and diabetic retinopathy. AZP nanospheres were prepared via coacervation technique, dispersed in HA hydrogel and characterised via infrared spectroscopy (FTIR), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). Size and morphology were studied via scanning electron microscopy (SEM) and dynamic light scattering (DLS), UV spectroscopy for photo-responsiveness. Rheological properties and injectability were investigated, as well as cytotoxicity effect on HRPE cell lines. Particle size obtained was <200 nm and photo-responsiveness to UV = 365 nm by decreasing particle diameter to 94 nm was confirmed by DLS. Encapsulation efficiency of the optimised nanospheres was 85% and IgG was released over 32 days up to 60%. Injectability of HA-NSP was confirmed with maximum force 10 N required and shear-thinning behaviour observed in rheology studies. In vitro cell cytotoxicity effect of both NSPs and HA-NSP showed non-cytotoxicity with relative cell viability of ≥80%. A biocompatible, biodegradable injectable photo-responsive nanosystem for sustained release of macromolecular IgG was successfully developed.  相似文献

Estimating the biting fraction of mosquitoes is of critical importance for risk assessment of malaria transmission. Here, we present a novel odor-based tool that has been rigorously assessed in semi-field assays and traditional African villages for estimating the number of mosquitoes that enter houses in search of a blood meal. A standard synthetic blend (SB) consisting of ammonia, (S)-lactic acid, tetradecanoic acid, and carbon dioxide was complemented with isovaleric acid, 4,5 dimethylthiazole, 2-methyl-1-butanol, and 3-methyl-1-butanol in various combinations and concentrations, and tested for attractiveness to the malaria mosquito Anopheles gambiae. Compounds were released through low density polyethylene (LDPE) material or from nylon strips (nylon). Studies were done in a semi-field facility and two traditional villages in western Kenya. The alcohol 3-methyl-1-butanol significantly increased the attraction of SB. The other compounds proved less effective or inhibitory. Tested in a village, 3-methyl-1-butanol, released from LDPE, increased the attraction of SB. Further studies showed a significantly enhanced attraction of adding 3-methyl-1-butanol to SB compared to previously-published attractive blends both under semi-field and village conditions. Other mosquito species with relevance for public health were collected with this blend in significantly higher numbers as well. These results demonstrate the advent of a novel, reliable odor-based sampling tool for the collection of malaria and other mosquitoes. The advantage of this odor-based tool over existing mosquito sampling tools is its reproducibility, objectiveness, and relatively low cost compared to current standards of CDC light traps or the human landing catch.  相似文献

This project investigates the classic agenda‐setting hypothesis in the context of the Greek cultural market. It is hypothesized that Greek museums with higher visibility in newspaper content are related to higher visitation than museums with lower media visibility. Because of the nature of the Greek cultural market—Greece receives more than 10 million tourists during the summer months—several variables are controlled for, such as the seasonality of visitation, the type of governance of the organization, one‐time events, such as the Olympic Games, which took place in the summer of 2004, and promotion initiatives undertaken by museums. When controlling for such culturally specific variables, there is evidence supporting the agenda‐setting hypothesis within the Greek cultural market.  相似文献

One and a half years after the worst oil spill in U.S. history, we present the first quantitative assessment on the ecological impact of the spill on the salt marsh habitats along the southeastern Louisiana (LA) coast. This research combined satellite and ground data to quantify the impact of the oil and dispersant on the salt marshes in terms of their photosynthetic capacity and physiological status over a large spatial scale. Two of the most important marsh biophysical characteristics, including distribution of canopy chlorophyll content and above ground green biomass, were monitored across the southeastern LA coast during the salt marsh growing season (May–October) of 2009 (pre-spill) and 2010 (post-spill) in order to compare and isolate the spill impacted areas. The initial assessment showed that there was a significant post-spill increase in areas with reduced biomass and canopy chlorophyll (> 400 km2) during the 2010 growing season compared to 50–65 km2 during the 2009 growing season. Phenological analysis of the post oil-spill data revealed a significant decrease in the magnitude of biomass and canopy chlorophyll during the peak of the 2010 growing season. June was consistently found to be the worst month in terms of salt marsh health across LA over the 2010 phenological cycle followed by the initial signs of recovery along the fringing marsh areas proximal to the shoreline that were first impacted by oil. Interior marsh patches exhibited persistent signs of stress towards the end of the growing season. Extensive reduction in photosynthetic activity was observed during the peak of the growing season, particularly in Plaquemines Parish and St. Bernard Parish. The products generated through this study successfully delineate the critical hotspots of marsh stress so that prioritization of areas needing immediate restoration can be performed.  相似文献

Wireless body area networks have become popular due to recent technological developments in sensor technology. A sensor can be used to collect data from different environments of interest, process and communicate the data to other nodes in a network. By its very nature, a sensor node is limited in resource usage. Due to these limitations, numerous security challenges have emerged in their applications, hence the need for more efficient and secure cryptosystems. In this paper, we give an efficient certificateless pairing-free signcryption scheme then design a secure access control scheme that can satisfy both the properties of ciphertext authentication and public verifiability using the signcryption scheme. A formal security proof of our scheme in random oracle model is provided. In addition, we compare the efficiency of our access control scheme with other existing schemes that are based on signcryption scheme. The analysis reveals that our scheme achieves better trade-off for computational and communication cost.

The deployment of odor-baited tools for sampling and controlling malaria vectors is limited by a lack of potent synthetic mosquito attractants. A synthetic mixture of chemical compounds referred to as “the Mbita blend” (MB) was shown to attract as many host-seeking malaria mosquitoes as attracted to human subjects. We hypothesized that this effect could be enhanced by adding one or more attractive compounds to the blend. We tested changes in the capability of MB (ammonia + L-lactic acid + tetradecanoic acid +3-methyl-1-butanol + carbon dioxide) to attract host-seeking malaria mosquitoes by addition of selected dilutions of butyl-2-methylbutanoate (1:10,000), 2-pentadecanone (1:100), 1-dodecanol (1:10,000), and butan-1-amine (1:10,000,000). The experiments were conducted in semi-field enclosures and in a village in western Kenya. In semi-field enclosures, the attraction of Anopheles gambiae sensu stricto females to MB-baited traps was not enhanced by adding butyl-2-methylbutanoate. There was, however, an increase in the proportion of An. gambiae caught in traps containing MB augmented with the selected dilutions of butan-1-amine, 2-pentadecanone, and 1-dodecanol. When tested in the village, addition of butan-1-amine to MB enhanced catches of female An. gambiae sensu lato, An. funestus, and Culex mosquitoes. 1-Dodecanol increased attraction of An. gambiae s.l. to the MB, while addition of 2-pentadecanone improved trap catches of An. funestus and Culex mosquitoes. This study demonstrates the possibility of enhancing synthetic odor blends for trapping the malarial mosquitoes An. gambiae s.l. and An. funestus, as well as some culicine species. The findings provide promising results for the optimization and utilization of synthetic attractants for sampling and controlling major disease vectors.  相似文献

This study aimed to develop and assess the long-term stability of drug-loaded solid lipid nanoparticles (SLNs). The SLNs were designed to extend the release profile, overcome the problems of bioavailability and solubility, investigate toxicity, and improve the antischistosomal efficacy of praziquantel. The aim was pursued using solvent injection co-homogenization techniques to fabricate SLNs in which Compritol ATO 888 and lecithin were used as lipids, and Pluronic F127 (PF127) was used as a stabilizer. The long-term stability effect of the PF127 as a stabilizer on the SLNs was evaluated. Dynamic light scattering (DLS) was used to determine the particle size, stability, and polydispersity. The morphology of the SLNs was examined through the use of transmission electron microscopy (TEM) and scanning electron microscopy (SEM). The chemical properties, as well as the mechanical, thermal, and crystal behaviours of SLNs were evaluated using FTIR, ElastoSens Bio2, XRPD, DSC, and TGA, respectively. SLNs with PF127 depicted an encapsulation efficiency of 71.63% and a drug loading capacity of 11.46%. The in vitro drug release study for SLNs with PF127 showed a cumulative release of 48.08% for the PZQ within 24 h, with a similar release profile for SLNs’ suspension after 120 days. DLS, ELS, and optical characterization and stability profiling data indicate that the addition of PF127 as the surfactants provided long-term stability for SLNs. In vitro cell viability and in vivo toxicity evaluation signify the safety of SLNs stabilized with PF127. In conclusion, the parasitological data showed that in S. mansoni-infected mice, a single (250 mg/kg) oral dosage of CLPF-SLNs greatly improved PZQ antischistosomal efficacy both two and four weeks post-infection. Thus, the fabricated CLPF-SLNs demonstrated significant efficiency inthe delivery of PZQ, and hence are a promising therapeutic strategy against schistosomiasis.  相似文献
