The ability to visualize cell motility occurring deep in the context of opaque tissues will allow many currently intractable issues in developmental biology and organogenesis to be addressed. In this study, we compare two-photon excitation with laser scanning confocal and conventional digital deconvolution fluorescence microscopy, using the same optical configuration, for their ability to resolve cell shape deep in Xenopus gastrula and neurula tissues. The two-photon microscope offers better depth penetration and less autofluorescence compared to confocal and conventional deconvolution imaging. Both two-photon excitation and confocal microscopy also provide improved rejection of "out-of-focus" noise and better lateral and axial resolution than conventional digital deconvolution microscopy. Deep Xenopus cells are best resolved by applying the digital deconvolution method on the two-photon images. We have also found that the two-photon has better depth penetration without any degradation in the image quality of interior sections compared to the other two techniques. Also, we have demonstrated that the quality of the image changes at different depths for various excitation powers.  相似文献

Background: Enzymes of tricarboxylic acid (TCA) have recently been recognized as tumor suppressors. Mutations in the SDHB subunit of succinate dehydrogenase (SDH) cause pheochromocytomas and paragangliomas (PCCs/PGLs) and predispose patients to malignant disease with poor prognosis. Methods: Using the human pheochromocytoma cell line (hPheo1), we knocked down SDHB gene expression using CRISPR-cas9 technology. Results: Microarray gene expression analysis showed that >500 differentially expressed gene targets, about 54%, were upregulated in response to SDHB knock down. Notably, genes involved in glycolysis, hypoxia, cell proliferation, and cell differentiation were up regulated, whereas genes involved in oxidative phosphorylation (OXPHOS) were downregulated. In vitro studies show that hPheo1 proliferation is not affected negatively and the cells that survive by shifting their metabolism to the use of glutamine as an alternative energy source and promote OXPHOS activity. Knock down of SDHB expression results in a significant increase in GLUD1 expression in hPheo1 cells cultured as monolayer or as 3D culture. Analysis of TCGA data confirms the enhancement of GLUD1 in SDHB mutated/low expressed PCCs/PGLs. Conclusions: Our data suggest that the downregulation of SDHB in PCCs/PGLs results in increased GLUD1 expression and may represent a potential biomarker and therapeutic target in SDHB mutated tumors and SDHB loss of activity-dependent diseases.  相似文献

We illustrate how channel optimized vector quantization (COVQ) can be used for channels with both bit-errors and bit-erasures. First, a memoryless channel model is presented, and the performance of COVQ's trained for this channel is evaluated for an i.i.d. Gaussian source. Then, the new method is applied in implementing an error-robust sub-band image coder, and we present image results that illustrate the resulting performance. Our experiments show that the new approach is able to outperform a traditional scheme based on separate source and channel coding.  相似文献

Renewable Energy Technologies (RETs) are often recognized as less competitive than traditional electric energy conversion systems. Obstacles with renewable electric energy conversion systems are often referred to the intermittency of the energy sources [1] and the relatively high maintenance cost. However, due to an intensified discourse on climate change and its effects, it has from a societal point of view, become more desirable to adopt and install CO2 neutral power plants. Even if this has increased the competitiveness of RETs in a political sense, the new goals for RET installations must also be met with economical viability. We propose that the direction of technical development, as well as the chosen technology in new installations, should not primarily be determined by policies, but by the basic physical properties of the energy source and the associated potential for inexpensive energy production. This potential is the basic entity that drives the payback of the investment of a specific RET power plant. With regard to this, we argue that the total electric energy conversion system must be considered if effective power production is to be achieved, with focus on the possible number of full loading hours and the Degree of Utilization [2]. This will increase the cost efficiency and economical competitiveness of RET investments, and could enhance faster diffusion of new innovations and installations without over-optimistic subsidies. This paper elaborates on the overall problem of the economy of renewable electric energy conversion systems by studying the interface between physics, engineering and economy reported for RET power plants in different scientific publications. The core objective is to show the practical use of the Degree of Utilization and how the concept is crucial for the design and economical optimization disregarding subsidies. The results clearly indicate that the future political regulative frameworks should consider the choice of renewable energy source since this strongly affects the economical output from the RET power plants.  相似文献

Liquid food production often involves continuous processing. This leads to problems in traceability systems due to mixing zones and therefore indistinct batch identities causing difficulties with regard to withdrawals or recalls. This article outlines the possible use of the concept of dynamic simulation to improve the handling of batch identities in continuous production of liquid food, a concept we call fuzzy traceability. The concept is illustrated with a realistic example from a real dairy process line.  相似文献

OBJECTIVES: An abundance of data exists documenting the association of H. pylori eradication with the reduction in duodenal ulcer recurrence. AIM: To evaluate the validity of using H. pylori eradication as a surrogate marker for the reduction in duodenal ulcer recurrence using rigorously controlled studies. METHODS: Three controlled clinical trials were conducted in patients with uncomplicated, active duodenal ulcers. Patients were treated with various combinations of omeprazole and amoxycillin. Ulcer healing and H. pylori eradication were assessed. For patients whose duodenal ulcer healed, duodenal ulcer recurrence was determined over a 6-month period in patients with H. pylori eradication and those remaining positive for H. pylori at least 4 weeks after treatment. To support the data obtained from these clinical trials, a search of the medical literature was conducted to identify additional human clinical trials in which duodenal ulcer recurrence rates were measured and categorized by H. pylori status at least 1 month post-treatment. RESULTS: In 11 controlled trials, the overall 6-18-month duodenal ulcer recurrence rate was 54% among patients remaining positive for H. pylori at least 4 weeks after treatment compared to 6% among patients with H. pylori eradication following treatment. This finding was corroborated by the uncontrolled trials, in which the duodenal ulcer recurrence rate was 64% among patients found to be H. pylori-positive and 6% for patients found to be H. pylori-negative at least 4 weeks after treatment. A time course of duodenal ulcer recurrence rates using pooled data from both controlled and uncontrolled studies demonstrated that duodenal ulcer recurrence rates for H. pylori-negative patients persisted for up to 4 years following treatment. Duodenal ulcer recurrence rates for H. pylori-positive patients increased for the first year, then levelled off. A comparison of the duodenal ulcer recurrence rates for different treatment regimens revealed that eradication regimens based on omeprazole plus antibiotics and bismuth plus antibiotics exhibited similar duodenal ulcer recurrence rates for H. pylori-positive and -negative patients. CONCLUSION: Regardless of treatment regimens, H. pylori eradication produced a consistent and significant reduction in duodenal ulcer recurrence. Therefore H. pylori eradication, 4 weeks post-therapy, can be used as a surrogate marker for reduced duodenal ulcer recurrence in investigational clinical trials.  相似文献

Multiple transmit and receive antennas can be used in wireless systems to achieve high data rate communication. Efficient space-time codes have been developed that utilize a large portion of the available capacity. These codes are designed under the assumption that the transmitter has no knowledge about the channel. In this work, on the other hand, we consider the case when the transmitter has partial, but not perfect, knowledge about the channel and how to improve a predetermined code so that this fact is taken into account. A performance criterion is derived for a frequency-nonselective fading channel and then utilized to optimize a linear transformation of the predetermined code. The resulting optimization problem turns out to be convex and can thus be efficiently solved using standard methods. In addition, a particularly efficient solution method is developed for the special case of independently fading channel coefficients. The proposed transmission scheme combines the benefits of conventional beamforming with those given by orthogonal space-time block coding. Simulation results for a narrow-band system with multiple transmit antennas and one or more receive antennas demonstrate significant gains over conventional methods in a scenario with nonperfect channel knowledge  相似文献

So far, the assumption of no channel knowledge at the transmitter has generally been inherent in the design of space-time codes. This paper, on the other hand, assumes that quantized channel information obtained from a feedback link is available at the transmitter and investigates how such channel information can be incorporated into the design of unstructured space-time block codes. Efficient codes are found by means of a gradient search over a continuous alphabet. Simulation results for an uncorrelated Rayleigh fading scenario using two and four transmit antennas and one receive antenna show the benefits of the code designs.  相似文献

An approach to optimal soft decoding for vector quantization (VQ) over a code-division multiple-access (CDMA) channel is presented. The decoder of the system is soft in the sense that the unquantized outputs of the matched filters are utilized directly for decoding (no decisions are taken), and optimal according to the minimum mean-squared error (MMSE) criterion. The derived decoder utilizes a priori source information and knowledge of the channel characteristics to combat channel noise and multiuser interference in an optimal fashion. Hadamard transform representations for the user VQs are employed in the derivation and for the implementation of the decoder. The advantages of this approach are emphasized. Suboptimal versions of the optimal decoder are also considered. Simulations show the soft decoders to outperform decoding based on maximum-likelihood (ML) multiuser detection. Furthermore, the suboptimal versions are demonstrated to perform close to the optimal, at a significantly lower complexity in the number of users. The introduced decoders are, moreover, shown to exhibit near-far resistance. Simulations also demonstrate that combined source-channel encoding, with joint source-channel and multiuser decoding, can significantly outperform a tandem source-channel coding scheme employing multiuser detection plus table lookup source decoding  相似文献
