Interleukin 4 (IL-4) is an immune cytokine that inhibits bone resorption in mice and suppresses osteoclastic cell formation in vitro through an undefined mechanism. In this report, we have established the cellular identity of the IL-4 target cell using a variety of bone marrow/stromal cell coculture methods. Initially, we found that the majority of IL-4's inhibition of osteoclastic cell formation was due to its effect on bone marrow cells, not stromal cells. Consequently, bone marrow macrophages were used as osteoclastic cell progenitors after they had been transiently exposed to IL-4 (48 h), before the addition of stromal cells, 1,25-dihydroxyvitamin D3, and dexamethasone. In this circumstance, IL-4 impaired subsequent osteoclastic cell formation, suggesting that the macrophage may be potentially targeted by many factors known to influence osteoclast formation. Consequently, we discovered that interferon-gamma (IFN gamma), prostaglandin E (PGE), and cell-permeant cAMP analogs also impacted osteoclastic cell formation when used to selectively treat bone marrow macrophages. IFN gamma suppressed osteoclastic cell formation, whereas PGE and cAMP analog treatment led to the formation of significantly enlarged osteoclastic cells. Importantly, PGE antagonized the inhibitory effects of both IL-4 and IFN gamma on the osteoclastic cell-forming potential of bone marrow macrophages. Collectively, these findings establish bone marrow macrophages as osteoclastic cell precursors with the degree of their commitment to the osteoclast pathway sensitive to the effects of soluble mediators, including IL-4, IFN gamma, and PGE. 相似文献

We investigated hepatitis C virus (HCV) infection in 35 patients with hypertrophic cardiomyopathy and 40 patients with ischemic heart disease who were consecutively admitted to our hospital. Frequency of positive anti-HCV antibody was significantly higher in patients with hypertrophic cardiomyopathy (6 of 35 patients, 17.1%) than that in patients with ischemic heart disease (1 of 40 patients, 2.5%, p = 0.036). In three of these six patients with hypertrophic cardiomyopathy, HCV RNA was detected in myocardial tissue. In two of these three patients, HCV RNA was detected from biopsy and autopsy specimens of the ventricles, but not in the serum, suggesting that HCV may replicate in myocardial tissue and may be relevant to ventricular hypertrophy. Mechanical deformation of Pd40Ni40P20 was characterized in compression over a wide strain rate range (3.3×10−5 to 2×103 s−1) at room temperature. The compression sample fractured with a shear plane inclined 42 degree with respect to the loading axis, in contrast to 56 degree for the case of tension. This suggests the yielding of the material deviates from the classical von Mises yield criterion, but follows the Mohr-Coulomb yield criterion. Fracture stress as well as strain was found to decrease with increasing applied strain rate. The compressive stress (1.74 GPa) was also found to be higher than the tensile fracture stress at a quasi-static strain rate. Close examination of the stress–strain curves revealed that localized shear might have occurred at a compressive stress of about 1.4 GPa, much lower than the “apparent” yield stress of 1.74 GPa. However, the stress of 1.4 GPa for shear band initiation is almost the same as the fracture stress measured at a dynamic strain rate of 5×102 s−1. These results suggested that the fracture of a bulk metallic glass is sensitive to the applied loading rate. 相似文献

The properties of a quasi-one-dimensional (Q1D) electron system on liquid helium are considered and the experiments on the carrier transport are presented. It has been shown that the electron mobility changes from high values close to that for bulk helium to very small values as a function of the Q1D channel width. Such a behaviour of is connected to a gradual transition from the regime of the quasi-free movement of the carriers in a Q1D system to the conditions of strong localization of the electrons. In the last case depends on temperature exponentially with the activation energy of a few degrees.
It is predicted that in Q1D channels in regime of the localization two optical plasmon branches can exist. One of them (a high frequency branch) is connected with the oscillations of electrons in potential wells and another one has lower frequency and is due to the oscillations of dimples, which are created by localized electrons on the liquid helium surface, in the potential wells.
Possible new experiments on study of magnetotransport, plasma oscillations and phase diagram of ordered-disordered states in a Q1D electron system of a finite length over liquid helium are proposed and discussed. 相似文献

The simultaneous addition of Al and Ag to Zr–Cu binary alloys increased in the stabilization of supercooled liquid, the reduced glass transition temperature and γ value, leading to greatly enhance the glass-forming ability (GFA). The Zr–Cu–Ag–Al glassy alloy samples with diameters above 15 mm were obtained in the wide composition range of 42–50 at% Zr, 32–42 at% Cu, 5–10 at% Ag, and 5–12 at% Al. The best GFA was obtained for Zr48Cu36Ag8Al8 alloy, and the glassy samples with diameters up to 25 mm were fabricated by an injection copper mold casting. The Zr48Cu36Ag8Al8 glassy alloy exhibited high tensile and compressive fracture strength of over 1800 MPa. 相似文献
