The aim of this paper is to present a methodology for the evaluation of historic timber structures, based on numerical modeling coupled with on site measurements. A method to measure the geometry of structural elements of a timber roof using a photogrammetric modeling procedure is described. In particular, we show the utility of photogrammetric based models when applied to structural analysis by directly introducing 3D geometry files in the preprocessing module of computational software based on the Finite Element Method. The 2D and 3D FEM models of the roof are considered to compare the simple and more complex modeling, and static and dynamic analyses are performed. Both models are prepared using the photogrammetric data.  相似文献

The transport properties of the uncoupling protein (UCP) from brown adipose tissue have been studied in mutants where Cys304 has been replaced by either Gly, Ala, Ser, Thr, Ile or Trp. This position is only two residues away from the C-terminus of the protein, a region that faces the cytosolic side of the mitochondrial inner membrane. Mutant proteins have been expressed in Saccharomyces cerevisiae and their activity determined in situ by comparing yeast growth rates in the presence and absence of 2-bromopalmitate. Their bioenergetic properties have been studied in isolated mitochondria by determining the effects of fatty acids and nucleotides on the proton permeability and NADH oxidation rate. It is revealed that substitution of Cys304 by non-charged residues alters the response of UCP to fatty acids. The most effective substitution is Cys for Gly since it greatly enhances the sensitivity to palmitate, decreasing threefold the concentration required for half-maximal stimulation of respiration. The opposite extreme is the substitution by Ala which increases twofold the half-maximal concentration. We conclude that the C-terminal region participates in the fatty acid regulation of UCP activity. The observed correlation between yeast growth rates in the presence of bromopalmitate and the calculated activation constants for respiration in isolated mitochondria validates growth analysis as a method to screen the in situ activity of UCP mutants.  相似文献

The suitability of hyphenated USAED with HPLC separation and ET-AAS determination as a new rapid methodology for Se control in Se-enriched food supplements is demonstrated. Total Se determination and Se speciation are accomplished in a single sample treatment using low sample amounts (ca. 10 mg), and low extracting volume (1 mL). The total Se content in seven of the 10 Se-enriched supplements studied was in agreement with the values obtained after microwave pressurized acid digestion, MW, (test t, p = 0.05). The Se species studied were Se(IV), Se(VI), SeMet, SeMeSeCys, and SeCys2, being some of the most common found in the 10 supplements studied. Although SeMet was the Se species expected to be present at the highest concentration in most Se-enriched food supplements, we detected it in only three of the 10 samples studied. In the other seven samples, two of them had Se(IV) as the main Se species. The other five supplements had Se species that did not match with any of the five standards selected by us. We have also systematically demonstrated that ultrasonication does not alter the following Se species: Se(IV), Se(VI), SeMet, SeMeSeCys, and SeCys2. The new procedure can be easily adapted to more Se species and can be routinely used for Se control in Se-enriched food supplements. Concerning the supplements studied, our results suggest that stricter control on the Se content in enriched food supplements in terms of Se species will need to become mandatory.  相似文献

The UK, like the rest of the world, is confronting the impacts of climate change. Further changes are expected and they will have a profound effect on agriculture. Future crop production will take place against increasing CO2 levels and temperatures, decreasing water availability, and increasing frequency of extreme weather events. This review contributes to research on agricultural practices for climate change, but with a more regional perspective. The present study explores climate change impacts on UK agriculture, particularly food crop production, and how to mitigate and build resilience to climate change by adopting and/or changing soil management practices, including fertilisation and tillage systems, new crop adoption and variety choice. Some mitigation can be adopted in the shorter term, such as changes in crop type and reduction in fertiliser use, but in other cases the options will need greater investment and longer adaptation period. This is the case for new crop variety development and deployment, and possible changes to soil cultivations. Uncertainty of future weather conditions, particularly extreme weather, also affect decision‐making for adoption of practices by farmers to ensure more stable and sustainable production. Even when there is real potential for climate change mitigation, it can sometimes be more difficult to accomplish with certainty on‐farm. Better future climate projections and long‐term investments will be required to create more resilient agricultural systems in the UK in the face of climate change challenges. © 2016 Society of Chemical Industry  相似文献

Adenosine A2A receptors (A2AR) control fear memory and the underlying processes of synaptic plasticity in the amygdala. In other brain regions, A2AR activation is ensured by ATP-derived extracellular adenosine formed by ecto-5′-nucleotidase or CD73. We now tested whether CD73 is also responsible to provide for the activation of A2AR in controlling fear memory and amygdala long-term potentiation (LTP). The bilateral intracerebroventricular injection of the CD73 inhibitor αβ-methylene ADP (AOPCP, 1 nmol/ventricle/day) phenocopied the effect of the A2AR blockade by decreasing the expression of fear memory, an effect disappearing in CD73-knockout (KO) mice and in forebrain neuronal A2AR-KO mice. In the presence of PPADS (20 μM) to eliminate any modification of ATP/ADP-mediated P2 receptor effects, both AOPCP (100 μM) and the A2AR antagonist, SCH58261 (50 nM), decreased LTP magnitude in synapses of projection from the external capsula into the lateral amygdala, an effect eliminated in slices from both forebrain neuronal A2AR-KO mice and CD73-KO mice. These data indicate a key role of CD73 in the process of A2AR-mediated control of fear memory and underlying synaptic plasticity processes in the amygdala, paving the way to envisage CD73 as a new therapeutic target to interfere with abnormal fear-like emotional processing.  相似文献

Privacy-preserving billing protocols are useful in settings where a meter measures user consumption of some service, such as smart metering of utility consumption, pay-as-you-drive insurance and electronic toll collection. In such settings, service providers apply fine-grained tariff policies that require meters to provide a detailed account of user consumption. The protocols allow the user to pay to the service provider without revealing the user’s consumption measurements. Our contribution is twofold. First, we propose a general model where a meter can output meter readings to multiple users, and where a user receives meter readings from multiple meters. Unlike previous schemes, our model accommodates a wider variety of smart metering applications. Second, we describe a protocol based on polynomial commitments that improves the efficiency of previous protocols for tariff policies that employ splines to compute the price due.  相似文献

The results of NiMo/γ-Al2O3 hydrodemetalation catalyst deactivation in a long-term test are presented. A residue from Maya crude has been processed in a continuous hydroprocessing unit provided with a trickle-bed reactor (TBR) at 648 K, 10 MPa H2 pressure, 1 l/h gcat LHSV and 0.2 kg/kg ratio H2/oil. Samples of catalyst from 100, 1100, 2100 and 3100 h on-stream have been characterised by Ni, V and Mo determination and the evolution of the structural and morphological characteristics of these samples has been followed by X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS) and scanning electron microscopy (SEM) using electron backscattering detector and microanalysis by dispersion of energies (EDS). XRD and XPS results have confirmed the presence of sulphide phases with a stoichiometry close to (V, Ni, Fe)3S4. Metal deposition profiles of vanadium have shown high concentrations in the edges while nickel deposition profiles have been more uniform. The shape of these profiles has indicated that for vanadium removal reactions, the metal deposition process becomes more diffusion rate limited, resulting in a less deep penetration into the catalyst pellet.  相似文献
