We describe the development of a preparative method to isolate molluscan catch muscle, calponin. This method is based on the ability of calponin to interact with actin in a temperature-dependent manner. After extracting thin filaments, as previously described, the extract was ultracentrifuged at 2 °C. While other surface proteins of thin filaments co-precipitated with actin, calponin, along with some minor contaminants, remained in the supernatant. Calponin was purified through cation-exchange chromatography. The yield of pure protein was four-fold higher than that achieved through high-temperature extraction. To evaluate functionally isolated proteins, we determined the effect of calponin on Mg2+-ATPase activity of hybrid and non-hybrid actomyosin. The degree of ATPase inhibition was consistent with previously published data but strongly dependent on the environmental conditions and source of actin and myosin used. Furthermore, at low concentrations, calponin could induce the ATPase activity of hybrid actomyosin. This result was consistent with data indicating that calponin can modulate actin conformation to increase the relative content of “switched on” actin monomers in thin filaments. We assume that calponin obtained by the isolation method proposed herein is a fully functional protein that can both inhibit and induce the ATPase activity.  相似文献

The nervous system expresses neuromolecules that play a crucial role in regulating physiological processes. Neuromolecule synthesis can be regulated by oxygen-dependent enzymes. Bivalves are a convenient model for studying air exposure-induced hypoxia. Here, we studied the effects of hypoxia on the expression and dynamics of neurotransmitters, and on neurotransmitter enzyme distribution, in the central nervous system (CNS) of the scallop Azumapecten farreri. We analyzed the expression of the neurotransmitters FMRFamide and serotonin (5-HT) and the choline acetyltransferase (CHAT) and universal NO-synthase (uNOS) enzymes during air exposure-induced hypoxia. We found that, in early-stage hypoxia, total serotonin content decreased in some CNS regions but increased in others. CHAT-lir cell numbers increased in all ganglia after hypoxia; CHAT probably appears de novo in accessory ganglia. Short-term hypoxia caused increased uNOS-lir cell numbers, while long-term exposure led to a reduction in their number. Thus, hypoxia weakly influences the number of FMRFamide-lir neurons in the visceral ganglion and does not affect peptide expression in the pedal ganglion. Ultimately, we found that the localization and level of synthesis of neuromolecules, and the numbers of cells expressing these molecules, vary in the scallop CNS during hypoxia exposure. This indicates their possible involvement in hypoxia resistance mechanisms.  相似文献

We study the corrosion, electrochemical, and corrosion–mechanical properties of deposited KKh30N6VSR cobalt-based, NKh15SR3 nickel-based, and Kh16N8S5 iron-based alloys in a standard NACE hydrogen-sulfide solution. It is shown that the deposited Kh16N8S5 iron-based alloy is unsuitable for these conditions due to its low corrosion resistance and susceptibility to hydrogen-induced cracking. The cobalt- and nickel-based alloys are suitable for the surfacing of the components of shutoff fittings of the oil-and-gas extraction equipment. However, as far as the “operating characteristics–cost” ratio is concerned, the nickel-based alloy outweighs the cobalt- based alloy.  相似文献

In this work, a closely spaced dual turbine concept is studied. The distance between the two side-by-side hubs is 1.05 D, where D is the rotor diameter. This configuration has a potential benefit for offshore wind developments in which power density can be maximized. The main goal is to evaluate the overall aerodynamic performance, blade loads, and wake structure of a reference wind turbine generator operating within this dual turbine configuration and to compare the effects against those for the typical single turbine configuration. For this purpose, an actuator line model has been employed together with the large eddy simulation approach for predicting the turbulence effects. This model was implemented by using the open-source computational fluid dynamics toolbox OpenFOAM. Results show a better performance for the dual turbine concept. Under same operating conditions, the aerodynamic power of each turbine within the dual concept is higher than the power of the stand alone turbine, particularly at lower operating wind speeds (approximately 2% to 3% of extra power per turbine). Comparison between the two configurations shows similar character of the tangential and normal forces acting on the blades in terms of magnitude and fluctuation, eliminating potential concerns regarding fatigue and blade design. The largest difference in the tangential and normal root bending moments are approximately 3% and 2%, respectively, between single and dual turbine configurations. Finally, wake recovery analysis shows a downwind velocity deficit that is not enhanced streamwise in the dual turbine configuration with no considerable difference after 7 D.  相似文献
