Mathematical modeling of viral replication dynamics, based on sequential measurements of levels of virion-associated RNA in plasma during antiretroviral treatment, has led to fundamental new insights into human immunodeficiency virus type 1 pathogenesis. We took advantage of the simian immunodeficiency virus (SIV)-infected macaque model to perform detailed measurements and mathematical modeling during primary infection and during treatment of established infection with the antiretroviral drug (R)-9-(2-phosphonylmethoxypropyl)adenine (PMPA). The calculated clearance half-life for productively infected cells during resolution of the peak viremia of primary infection was on the order of 1 day, with slightly shorter clearance half-lives calculated during PMPA treatment. Viral reproduction rates upon discontinuation of PMPA treatment after 2 weeks were approximately twofold greater than those obtained just prior to initiation of treatment in the same animals, likely reflecting accumulation of susceptible target cells during treatment. The basic reproductive ratio (R0) for the spread of SIV infection in vivo, which represents the number of productively infected cells derived from each productively infected cell at the beginning of infection, was also estimated. This parameter quantifies the extent to which antiviral therapy or vaccination must limit the initial spread of virus to prevent establishment of chronic disseminated infection. The results thus provide an important guide for efforts to develop vaccines against SIV and, by extension, human immunodeficiency virus.  相似文献

BACKGROUND: Hearing loss is generally regarded as a rare side effect of erythromycin therapy. However, our own clinical experiences in erythromycin-treated patients led us to suspect that this complication may be more common among renal allograft recipients. The purpose of this study was to evaluate the incidence, predisposing factors, clinical characteristics, and outcomes of erythromycin-induced hearing loss among renal allograft recipients. METHODS: We reviewed medical records of renal transplant patients treated for pneumonia with intravenous erythromycin lactobionate. Patients were evaluated for the occurrence of clinically significant hearing loss (including onset, duration, and reversibility), other signs and symptoms of ototoxicity (vertigo and tinnitus), daily erythromycin dose and duration of treatment, concurrent ototoxic drug therapy, renal and hepatic function, and history of previous otic disease. RESULTS: Eleven (32%) of 34 courses of intravenous erythromycin therapy resulted in hearing loss. The incidence of hearing loss was 53% (eight of 15 courses) in patients treated with 4 g of erythromycin daily compared with 16% (three of 19 courses) among those receiving 2 g/d (P = .05). In addition, courses of erythromycin were longer in those suffering auditory toxicity (9.6 +/- 4.7 days) than in nontoxic patients (5.7 +/- 3.6 days) (P < .05). Hepatic and renal function did not differ between toxic and nontoxic patients. All episodes of erythromycin-induced hearing loss were reversible. CONCLUSIONS: We conclude that clinically significant hearing loss occurs in more than 30% of renal allograft recipients treated for pneumonia with intravenous erythromycin lactobionate. Patients who require prolonged courses of erythromycin and those treated with 4 g/d are at particular risk for the development of auditory toxicity. With prompt recognition and modification of therapy, erythromycin-induced hearing loss appears to be completely reversible.  相似文献

An extra-heavy crude oil underground upgrading process is described which involves the downhole addition of a hydrogen donor additive under steam injection conditions (280-315°C and residence times of at least 24-h). Laboratory experiments showed a 4° increase in the API gravity (from 9 to 12°) of the upgraded product, a two-fold reduction in the viscosity and, an approximately 8% decrease in the asphaltene content with respect to the original crude. Further increases on the temperature led to products with improved properties reaching 15°API at 315°C. It was found that the presence of the natural formation (catalysts) and methane (natural gas) is necessary to enhance the properties of the upgraded crude oil. From GC and GC-MS results a reaction pathway is proposed that involves hydrogen transfers from tetralin to the extra-heavy crude oil resulting in the formation of 1,2-dihydronaphthalene. This compound is then transformed into naphthalene, further upgrading of crude oil through hydrogen donation. The results of the experiments carried out in the presence and absence of the mineral formation and with an inert solid (SiC) strongly indicate that the former acts as a catalyst and not as a heat transfer matrix. Isotopic labeling studies (CD4 and 13CH4) give evidences that, most probably, methane is involved in the upgrading reactions.  相似文献

This article identifies the key issues involved in the debate about affirmative action. The June 2003 Supreme Court decisions allowing consideration of race to ensure that there is a "critical mass" of African American, Latino/Latina, and Native American applicants to higher education are addressed. Social psychologists have identified key myths and provided clarifications about the need for and consequences of strategies used to promote equal opportunity for persons of color and women. A brief history of affirmative action and of the problems it was designed to solve is provided. The accomplishments, benefits, and compelling interest of diversity and affirmative action are described, as well as the concerns and counterpoints. The lack of a substantial applicant pool in psychology hinders progress toward diversity. A method for fast calculation of viscosity from molecular dynamics simulation is revisited. The method consists of using a steady-state periodic perturbation. A methodology to choose the amplitude of the external perturbation, which is one of the major practical issues in the original technique of Gosling et al. [Mol. Phys. 26: 1475 (1973)] is proposed. The amplitude of the perturbation required for fast caculations and the viscosity values for wide ranges of temperature and density of the Lennard-Jones (LJ) model fluid are reported. The viscosity results are in agreement with recent LJ viscosity calculations. Additionally, the simulations demonstrate that the proposed approach is suitable to efficiently generate viscosity data of good quality.  相似文献

Temperature control of optical focal planes comes with the intrinsic challenge of creating a pathway that is both extremely flexible mechanically and highly conductive thermally. The task is further complicated because science-caliber optical focal planes are extremely delicate, yet time, cost, and their unique nature means that their mechanical resiliency is rarely tested and documented. The mechanical engineer tasked with the thermo-mechanical design must then create a highly conductive thermal link that minimizes the tensile and shear stresses transmitted to the focal plane without design parameters on an acceptable stiffness and without data on the stiffness of previously implemented thermal links.This paper describes the development and testing of the thermal link developed for the Portable Remote Imaging Spectrometer (PRISM) instrument. It will provide experimentally determined mechanical stiffness plots in the three axes of interest. Analytical and experimental thermal conductance results for the two-arm focal-plane thermal strap (TAFTS), from cryogenic to room temperatures, are also presented. The paper also briefly describes some elements of the fabrication process followed in developing a novel design solution, which provides high conductance and symmetrical mechanical loading, while providing enhanced flexibility in all three dimensions.  相似文献

Polypyrrole hollow nanoparticles were prepared by atmospheric pressure plasma polymerization. The structure of the nanoparticles was studied using Fourier transform infrared and X‐ray photoelectron spectroscopies, thermogravimetric analysis, scanning and transmission electron microscopies and atomic force microscopy. In contrast to low‐pressure plasma polymerization of pyrrole, which can produce films and solid nanoparticles, we obtained two types of hollow nanoparticles: a fraction with single spherical core and another with a core composed of small bubbles. Thermal characterization allowed us to determine that the nanoparticles are composed of highly crosslinked polymer. A mechanism that explains the formation of both types of hollow nanoparticles as well as solid nanoparticles is proposed. Chemical characterization shows that, in addition to the expected chemical structures due to pyrrole polymerization, the high energy of the plasma at atmospheric pressure produces intense dehydrogenation and oxidation processes. The fluorescence spectrum of the nanoparticles, however, shows a peak at 482 nm indicating that some degree of π‐conjugation is present in the material. © 2014 Society of Chemical Industry  相似文献
