Sarcopenia is defined as the age-related loss of skeletal muscle mass, quality, and strength. The pathophysiological mechanisms underlying sarcopenia are still not completely understood. The aim of this work was to evaluate, for the first time, the expression of NLRP3 inflammasome in bovine skeletal muscle in order to investigate the hypothesis that inflammasome activation may trigger and sustain a pro-inflammatory environment leading to sarcopenia. Samples of skeletal muscle were collected from 60 cattle belonging to three age-based groups. Morphologic, immunohistochemical and molecular analysis were performed to assess the presence of age-related pathologic changes and chronic inflammation, the expression of NLRP3 inflammasome and to determine the levels of interleukin-1β, interleukin-18 and tumor necrosis factor alpha in muscle tissue. Our results revealed the presence of morphologic sarcopenia hallmark, chronic lymphocytic inflammation and a type II fibers-selective NLRP3 expression associated to a significant decreased number of immunolabeled-fibers in aged animals. Moreover, we found a statistically significant age-related increase of pro-inflammatory cytokines such as interleukin-1β and interleukin-18 suggesting the activation of NLRP3 inflammasome. Taken together, our data suggest that NLRP3 inflammasome components may be normally expressed in skeletal muscle, but its priming and activation during aging may contribute to enhance a pro-inflammatory environment altering normal muscular anabolism and metabolism.  相似文献

We discuss the performance, of a normal metal hot electron bolometer (NHEB) that we have measured at 0.3 K. We found that the noise equivalent power was limited by the amplifier noise. To improve the NHEB power response and to make it more robust and reliable we propose to substitute the normal metal with heavily doped silicon. The heavily doped silicon behaves like a metal with lower carrier concentration and has a smaller electron–phonon thermal coupling. We have fabricated superconductor-doped silicon-superconductor contacts (S-Sm-S) and we have used them as thermometers and coolers.  相似文献

The aim of this study was to evaluate the effects of paste properties on residence time during drying in a spouted bed dryer with inert bodies. The effect of paste solids content, surface tension, and viscosity on the residence time distribution and the mean residence times were studied using factorial experimental designs. The inert bodies used were glass and polyethylene beads. The mean residence times varied from 13.6 to 16.3 and 12.2 to 17.7 min for drying on glass and polyethylene beads, respectively. The analysis of variance showed that mean residence times significantly depended on solids content and surface tension for glass beads and also on viscosity for polyethylene beads. The residence time distributions for all conditions studied fitted well to the perfect mixing cell when applying the continuous stirred vessels in series model analysis. The powder density, flowability, and particle size depended on paste properties and inert type.  相似文献

The piezoelectric films recently developed permit the design of very efficient thin sensors which are suited to interesting applications in contact problems, not only in industrial robotics but also in experimental research. After a brief introduction exemplifying the electrical principles, the primary aspects of the technique are presented and discussed with reference to possible structural engineering applications. A rather simple example regarding the interaction between a steel bar and a block of concrete is also presented. The bar is excited by dynamic cyclic loads having variable frequency. The reliability of forces detected via a PVDF sensor is evaluated by comparing the results with those obtained in parallel by means of a dynamometer strain-gauge. The comparison is promising, even if some technological problems must be resolved before this experimental technique may be applied extensively.
Resume L'étude expérimentale des forces qui se manifestent à une interface équivaut souvent à un problème technique insoluble car chaque appareillage influe de manière significative sur le comportement mécanique. En outre, si l'interface se situe dans un continuum, l'utilisation de transducteurs ou d'extensomètres devient aussi plus complexe. Dans tous les cas, les mesures peuvent se faire éventuellement à proximité de l'interface, mais non à l'interface. Des améliorations techniques récentes dans le domaine de la robotique permettent d'utiliser des films de polymères piézoélectriques (fluoride de polyvinylidène, PVDF). En raison de l'épaisseur de ce capteur qui ne dépasse pas quelques micromètres, le comportement mécanique n'est pas perturbé. Le principle électrique est typique du transducteur piézoélectrique: les charges crées par le film piézoélectrique sollicité sont concentrées sur les surfaces métallisées qui le recouvrent. Cette technique permet d'étudier de nombreux problèmes structurels. S'il s'agit de béton armé, par exemple, il est intéressant d'examiner la répartition de la pression sur un appui de poutre ou bien à la surface d'un boulon d'ancrage, les contraintes interfaciales aussi bien d'une barre d'armature chargée axialement et transversalement, que de goujons dans des structures composites. Un exemple simple montre la fiabilité de la nouvelle technique: une haute barre d'adhérence enrobée dans un bloc de béton est mise sous charge dynamique. On compare la force détectée par le capteur PVDF placé à l'interface barre d'acier-barre d'enrobage avec celle qui est mesurée au moyen du dynamomètre traditionnel placé parallèlement au support. La comparaison, établie à différentes fréquences, est satisfaisante. Cependant, pour utiliser cette nouvelle technique sur une grande échelle, il convient de résoudre quelques problèmes techniques: l'améioration de la conductivité électrique des connexions entre les surfaces métallisées de la couche sensible et les fils d'entrée et de sortie la prévention d'une éventuelle perforation du film par des particules de granulats siliceux qui sont un des constituants de base des mortiers et bétons; l'évaluation de la sensibilité d'un capteur multicouches à l'écrasement inévitable du matériau à proximité d'une barre d'acier, avec une microfissuration locale et un glissement limité. La recherche se poursuit tant au niveau technologique qu'électronique.

A simple, rapid and sensitive procedure for the simultaneous determination of total cholesterol, tocopherols and β-carotene in meat is described. The method involves a direct saponification of the meat, a single n-hexane extraction and the analysis of the extracted compounds by normal-phase HPLC, using fluorescence (tocopherols) and UV–Vis photodiode array (cholesterol and β-carotene) detections in tandem. Rates of recovery of spiked meat samples were 93% for cholesterol, 83–86% for (α-, β- and -γ) tocopherols and 89% for β-carotene. Repeatabilities were high (CV < 6%) for all determined compounds, except for δ-tocopherol. This tocopherol, which is not usually present in meat, showed a much lower recovery percentage (73%) and repeatability (12.8%). This methodology was applied for the quantification of total cholesterol, tocopherols and β-carotene in three muscles (longissimus thoracis, longissimus lumborum and semitendinosus) of the Portuguese traditional Barrosã-PDO veal, obtained from autochthonous calves fed extensively during summer (with the least abundant green pastures) and slaughtered in early autumn (October). Barrosã-PDO veal showed median contents of total cholesterol (0.50–0.56 mg/g) and, depending on the analysed muscle, moderate to high contents of α-tocopherol (3.3–3.9 μg/g) and β-carotene (0.07–0.09 μg/g), suggesting an high sensorial and hygienic quality.  相似文献

BACKGROUND: Nisin is a commercially available bacteriocin produced by Lactococcus lactis ATCC 11454 and used as a natural agent in the biopreservation of food. In the current investigation, milk whey, a byproduct from dairy industries was used as a fermentation substrate for the production of nisin. Lactococcus lactis ATCC 11454 was developed in a rotary shaker (30 °C/36 h/100 rpm) using two different media with milk whey (i) without filtration, pH 6.8, adjusted with NaOH 2 mol L?1 and without pH adjustment, both autoclaved at 121 °C for 30 min, and (ii) filtrated (1.20 µm and 0.22 µm membrane filter). These cultures were transferred five times using 5 mL aliquots of broth culture for every new volume of the respective media. RESULTS: The results showed that culture media composed of milk whey without filtration supplied L. lactis its adaptation needs better than filtrated milk whey. Nisin titers, in milk whey without filtration (pH adjusted), was 11120.13 mg L?1 in the second transfer, and up to 1628‐fold higher than the filtrated milk whey, 6.83 mg.L?1 obtained in the firstt transfer. CONCLUSIONS: Biological processing of milk byproducts (milk whey) can be considered a profitable alternative, generating high‐value bioproducts and contributing to decreasing river disposals by dairy industries. The main goal of this work is to evaluate the extraction of sunflower oil from enzyme-treated collets using ethanol and isopropanol (IPA) as solvents. The sunflower collets are pretreated with the multienzyme complex Viscozyme L prior to solvent extraction by the Soxhlet method. The influence of the moisture content of the collets, pretreatment, processing time, and solvent type on the amount of total extracted material and the oil extraction efficiency is studied. Some quality parameters such as phospholipid content of the oil and chlorogenic acid content of the residual meal are also analyzed. At low moisture content (7%) the solvents exhibit similar oil extraction ability (98–99%), but with increasing moisture the extraction efficiency of ethanol decreases to about 85%, while no significant differences are observed for IPA. The enzymatic treatment increases the extraction efficiency for all times, especially for ethanol. It is observed that IPA is more efficient in the extraction compared to ethanol, and the amount of nonlipid material is reduced by ≈70%. In addition, the oil extracted with IPA have lower phospholipid content and the residual meal presents a higher chlorogenic acid content. Practical Applications:This work would contribute toward the use of green solvents in the extraction of sunflower oil from collets. Ethanol and isopropanol, used as solvents, present attractive advantages, including low toxicity, good operational security, as well as being obtained from a renewable source. The obtained data provide up-to-date information on the use of these alcohols in the extraction of sunflower oil from collets and the influence of operating conditions, such as moisture content, enzymatic pretreatment of the collets, and the extraction time. The effects of the SAR (14 and 38) and of the methodology of introduction of nickel (wetness impregnation and ion exchange) on catalyst activation were investigated in Ni/USY model catalysts submitted to two activation treatments: reduction (A) and reduction/calcination/reduction, successively (B). the catalysts prepared by wetness impregnation, a marked increase in the catalytic activity after treatment B was observed. On the basis of the catalyst efficiency toward ethane hydrogenolysis, the lower SAR catalyst presents a threefold higher activity. On the contrary, the catalysts prepared by ion exchange, as well as a model catalyst prepared by wetness impregnation over a USY exhaustively exchanged with nickel ions, do not present measurable increasing of the catalytic activity under the same activation conditions. Monitoring the catalyst activation by EXAFS indicates the formation of nickel clusters in the impregnated catalysts after activation treatment B, which should be responsible for the high catalytic activity. However, TEM images reveal a bimodal particle size distribution, with large (ca. 20 nm) and small nickel particles. Temperature-programmed reduction performed under the same conditions of catalyst activation suggests that only the small nickel particles are activated under the experimental conditions adopted in this work.  相似文献
