Fibroblast-mediated cytokine gene therapy has proven to be a promising strategy for restoring hematopoiesis following repeated chemotherapy. Interleukin 3 (IL-3) and interleukin 6 (IL-6) can synergistically promote the recovery of hematopoiesis following chemotherapy. In this investigation, combined use of fibroblast-mediated IL-3 and IL-6 gene therapy was tested for hematopoietic effects on mice with or without 5-fluorouracil administration. The results demonstrated that combined therapy with IL-3 gene-modified NIH3T3 cell (NIH3T3-IL-3) and IL-6 gene-modified fibroblast NIH3T3 cell (NIH3T3-IL-6) implantation achieves obvious stimulation of hematopoiesis in normal mice and accelerates recovery of hematopoiesis. In normal mice the quantities of platelets, neutrophils, and total white blood cells in peripheral blood increased significantly after the combined implantation of NIH3T3-IL-3 and NIH3T3-IL-6 cells. The numbers of colony-forming unit (CFU) granulocyte/macrophage (CFU-GM) and CFU megakaryocyte (CFU-MK) formed by stem cells in bone marrow was significantly higher after the combined implantation of NIH3T3-IL-3 and NIH3T3-IL-6 cells than after the implantation of NIH3T3-IL-3 alone, NIH3T3-IL-6 alone, or neomycin gene-modified NIH3T3 cells. In hematopoiesis-depressed mice induced by preinjection with 5-fluorouracil at the dose of 150 mg/kg before cell implantation, the platelets, neutrophils, and white blood cells showed accelerated recovery, and the numbers of CFU-GM and CFU-MK formed by bone marrow cells were also markedly higher after the combined implantation of NIH3T3-IL-3 and NIH3T3-IL-6 cells than in control groups. Our data show that combined use of fibroblast-mediated IL-3 and IL-6 gene therapy may be of clinical relevance for the recovery of hematopoietic depression for patients after chemotherapy.  相似文献

A consensual qualitative research strategy was used to examine data from 127 interviews conducted with 8 psychologists immediately following their sessions of brief therapy with 8 clients. Analyses revealed 3 domains relevant to countertransference: origins (including categories of family issues, needs and values, therapy specific issues, and cultural issues), triggers (including categories of content of client material, therapist comparing client with others, change in therapy structure or procedures, therapist assessing progress of therapy, therapist perception of client, and emotions), and manifestations (including categories of approach, avoidance, negative feelings, and treatment planning). The frequency of categories within and across cases was classified, and relationships among categories from the 3 domains were detected, generating hypotheses for future empirical research. This paper presents the results of microstructure and property determinations made on pure and alloyed copper sputter deposited at rates of 20–25 nm s-1. For each material, the effects of substrate temperature and post-deposition annealing on the grain size and tensile properties were investigated. Fully dense coherent structures were obtained at all substrate temperatures. The absence of the porous poorly bonded structures obtained at temperatures in the range 0.1–0.2Tm by other workers is attributed to the low gas pressure, the normal incidence adatoms and the polished substrates used in the present study.All materials exhibited high tensile strenghts when deposited at 0.2Tm Values of 700 MPa were observed with pure copper, while precipitation-hardening and dispersion-hardening alloys produced values of 1000MPa. The principal difference between these materials was in the stability of their as-deposited structure and their strength during post-deposition annealing. For example, pure copper was unstable to recrystallization at room temperature (0.2Tm), while the dispersion- strengthened alloys exhibited only minor softening at 0.8Tm and very little grain growth at 0.95Tm. The mechanical properties are discussed in relation to the microstructure produced by high rate deposition.  相似文献

In this paper, we present the principle of operation, design, and testing of a pulse echo radar unit that utilizes the properties of short helical antennas. Data was captured for targets approaching the radar at three different controlled velocities, three different target sizes, and three radar range gate settings. These data reveal the expected Doppler frequency as well as a second anomalous frequency. The effects of multipath signals, separated transmit and receive antennas, and direct antenna coupling are used to explain this anomalous frequency. These effects are mathematically simulated, and the results compare favorably with the test data  相似文献

Unusual multiphase microstructures formed in thick Ni-La and Ni-Y films that were produced by high rate sputter deposition, when the substrate was biased at several hundred volts during deposition. Films biased in this range were bombarded with an intense flux of Kr+ ions (approximately 5 × 1016 cm-2 s-1) because of the dense thermionically supported plasma used here. In these films large oriented precipitates formed near certain defects or irregularities in the substrate or film surface. The matrix in which these growths formed was amorphous or microcrystalline. No heterogeneities were observed in films deposited with a negative substrate bias lower than about 50 V. Those films were amorphous and were homogeneous in composition. Examples of the heterogeneous microstructures are presented here, and reasons for their formation are discussed.  相似文献

In order to examine the relationship between local adenosine concentrations before, during, and after ischemia and the extent of ischemic myocardial damage, measurements of interstitial fluid (ISF) nucleosides were made using microdialysis probes implanted in the ischemic region of isoflurane anesthetized Micropigs undergoing 60' coronary artery occlusion (CAO) and 3 h of reperfusion (REP). Nucleoside concentrations in the dialysate collected from the microdialysis probes were used as an index of ISF levels. Dialysate nucleoside concentrations (ADO, inosine and hypoxanthine), myocardial infarct size, and myocardial blood flow (MBF) were determined in control animals (n = 6), animals preconditioned with a single 10' cycle of CAO and REP (PC, n = 6), and those treated with the adenosine deaminase inhibitor pentostatin (n = 6, 0.2 mg/Kg i.v. 30' prior to CAO). The brief PC occlusion resulted in a transient but significant increase in dialysate ADO (6.7 +/- 1.8 microM vs. 0.67 +/- 0.1 microM at baseline). Pentostatin administration had no significant effect on either dialysate nucleosides or MBF at baseline. During the 60' CAO, dialysate ADO increased in control animals. In PC animals, however, dialysate ADO during CAO was lower than control. Pretreatment with pentostatin resulted in a six-fold augmentation in dialysate ADO during the 60 min CAO when compared to the control values (110.62 +/- 30.2 microM vs. 16.31 +/- 2.1 microM at 60 min of ischemia). Pentostatin also resulted in a significant reduction in the accumulation of inosine and hypoxanthine, indicating inhibition of adenosine deaminase activity. There were no significant differences in MBF between groups at any time point. Following 3 h REP, infarct size was 35.4 +/- 5.5%, 8.1 +/- 1.5% and 8.3 +/- 1.8% of the region at risk in control, PC, and pentostatin groups, respectively. These data suggest that marked increase in ISF ADO during CAO, may be as effective in reducing INF as a modest increase in ISF ADO prior to prolonged CAO.  相似文献

Asked 234 mental hospital employees in 5 disciplines to complete a staff attitude scale. 6 attitude factor scores were compared using a Discipline * Experience multivariate analysis of variance with education as covariate. Attitudes toward patient care were significantly different among 4 disciplines and across 6 different experience levels. A separate analysis yielded no sex differences. Using previously published means from 6 hospitals, plus data from the present sample, a multivariate Hospital * Discipline analysis was completed on 2,178 Ss. The 6 traditional hospitals differed significantly from a transitional hospital. Because of possible violations of assumptions, the last analysis requires further support.
